Summary Protoplast formation in Curvularia inaequalis was achieved using non-commercial and commercial snail gut enzymes or Trichoderma harzianum enzymes. The cells were grown for enzyme treatment on cellophane sheets or in liquid cultures for varying periods of time. The production of T. harzianum enzymes is discussed. The highest protoplast yields were 2.6x107 protoplasts/ml enzyme solution. Protoplasts were shown to have zero to four nuclei. Protoplast regeneration was succesfully carried out in semisolid agar.  相似文献

Cotransformants of yeast cells by two partially homologous plasmids, one of which is incapable of autonomous replication, has been used to construct multiply marked recombinant plasmids. Only simultaneous elimination of three yeast markers was registered when episomal plasmid, carrying Ade2 gene, and integrative plasmid, carrying yeast genes LEU2 and URA3, were cotransformed. Transformants, in which yeast genes LEU2, URA3 and HIS3 are linked, have been isolated by analogous technique. The genetic analysis has confirmed existence of plasmid cointegrates in the transformant cells, which carry three yeast genes, bacterial DNA fragment and 2 micrometers DNA fragment, coding for replicative functions. Recombination in the region of bacterial plasmid pBR322 might have resulted in formation of such plasmids. Plasmid recombination in cotransformants has been used to construct multiply marked circular chromosomes, having included yeast genes LEU2, URA3 and TRP1, centromere of the IV yeast chromosome and the sequence coding for their replication in yeast as well as in E. coli cells.  相似文献

The skeleton is the first pool of magnesium in the animal body. 3. For intestinal absorption, renal metabolism, bone accretion and resorption, magnesium shows analogies with calcium. 4. Magnesium exchange between extracellular, cellular and skeletal compartments are very precisely controlled. 5. Parathyroid hormone, 1 alpha, 25-dihydroxy-vitamin D3, calcitonin and estrogens are the principal hormone systems implicated in magnesium metabolism. 6. 1. The effect of estradiol and pituitary hormones on the titre of serum vitellogenin has been studied in Rana esculenta by rocket immunoelectrophoresis. 2. Hepatic synthesis of vitellogenin depends on physiological doses of estradiol. 3. Gonadotrophins enhance the uptake, presumably by acting directly on the oocyte plasma membrane. 4. In addition, our data support direct pituitary intervention on liver synthesis and/or release of vitellogenin. 5. Hormonal response, as evaluated by vitellogenin serum titres, tends to increase from November to July. This could be the expression of a modification, throughout the sexual cycle, of liver sensitivity to the hormones.  相似文献

Investigations were carried out into the prevalence of tinea capitis infection among school children in Ile-Ife, Nigeria. The prevalence rate of clinical infection was found to be 14.02%. The infection was found to be more frequent among pupils between ages 5 and 10 years, and more prevalent among the poor than the rich (p<0.005). Families having between 1 and 4 children had lower prevalence than families having minimum of five children (p<0.10). Approximately one-third of the clinically infected children were not receiving any treatment while about two-thirds of the remaining two-thirds were using local native remedies, some of which had resulted in deaths of some children. The most common causative agent isolated from the lesions was Microsporum audouinii.  相似文献

By means of a cytological method involving BrdU incorporation and acridine orange fluorescence staining in combination with embryo manipulation, we studied X-chromosome activity in female preimplantation mouse embryos with special reference to the correlation between X-chromosome inactivation and cell differentiation. There was no sign of asynchronous replication between the two X chromosomes from the one-cell to intermediate blastocyst stage. The allocyclic X chromosome, first detected in late blastocysts, was paternal in origin, mostly replicating early in the S phase and limited to the trophectoderm. Subsequent X-chromosome inactivation occurring in the primary endoderm was also characterized by the involvement of the paternal X and early replication. Both X chromosomes continued to replicate synchronously in the embryonic ectoderm or epiblast at this stage. It was evident that overt cell differentiation preceded the appearance of the asynchronously replicating X chromosome in the trophectoderm and primary endoderm. This finding seems to support the view that cell differentiation is an important correlate of X-chromosome inactivation.  相似文献

A method is given to predict the unitary free energies of complexation between drug-like and nucleoside-like biomolecules in a range of mixed solvent compositions. A stability maximum for the actinomycin (A)-deoxyguanosine (D) complex at 8% MeOH (v/v) in methanol/water mixtures is correctly predicted by the theory in agreement with existing experimental data. The molecular surface areas of A and D exposed to the solvent are found to diminish by 36.4 A(2) upon association. The 'microthermodynamic differential surface tension' of the solvophobic theory obtained for nucleoside-like and organic molecules in contact with MeOH/H2O can be used to predict the solvent effect free energies in other such molecular or biopolymeric associations in solution.  相似文献

Independent experiments have shown that both protein folding (G. Velicelebi and J.M. Sturtevant, Biochemistry 18 (1979) 1180) and drug-biomolecule complexation (D.M. Crothers and D.I. Ratner, Biochemistry 7 (1968) 1823) in a wide range of compositions of methanol/water mixed solvents exhibit a maximum at 8% (v/v) MeOH. This hitherto unexplained phenomenon is shown to be given a priori by the 'solvophobic theory' developed earlier by Sinanoglu which had related the solvent effects including water in biochemistry to the then introduced 'molecular surface areas' and to 'microthermodynamic cavity inner surface tensions' and in a different version to interfacial microtensions between side chains and the solvent. Both analyses carried out in the present paper in detail for MeOH/water mixtures show how the denaturation or complexation free energies are predicted for the entire range of MeOH/water compositions from only data at one point. The molecular surface area changes for the conformational processes are obtained as well as the free energies in the hypothetical but theoretically important in vacuo limits with no solvent present.  相似文献
