Endometrial cancer (EC) is the most common gynaecological malignancy with increasing incidence in developed countries. As gold standard, hysteroscopy confirms only 30% of suspected ECs. The detection of EC cells in the vagina by fluorescence in situ hybridization (FISH) after a smear test could reduce invasive procedures in the future. Using array-based comparative genome hybridization (aCGH) on 65 endometrial carcinomas, most frequently imbalanced regions of the tumour genome were identified. Bacterial artificial chromosomes were used to generate FISH-probes homologue to these human regions. The FISH test was hybridized on swabs specimens collected from the vaginal cavity. Samples from six patients without EC were selected as a negative control and on 13 patients with known EC as a positive control. To distinguish between benign and EC cases, the cut-off value has been defined. A first validation of this EC-FISH Test was performed with swabs from 41 patients with suspected EC. The most common genomic imbalances in EC are around the CTNNB1, FBXW7 and APC genes. The cut-off is defined at 32% of analysed cells without diploid signal pattern. This differs significantly between the positive and negative controls (p < 0.001). In a first validation cohort of 41 patients with suspected EC, the EC-FISH Test distinguishes patients with and without EC with a sensitivity of 91% and a specificity of 83%. The negative predictive value is 96%. This is the first report of a non-invasive EC-FISH Test to predict EC in women with suspected EC.  相似文献

In the Lazarev Sea, Southern Ocean, I studied the population structure and reproduction of Eukrohnia bathypelagica and E. bathyantarctica during winter and summer. A lack of unimodality in their population structures indicated at least two generations building one population and a life cycle longer than 1 year. During both seasons all maturity stages were present; therefore, continuous reproduction is very probable. Extensive breeding seasons with several releases of eggs by one generation are assumed, as adults with empty marsupial sacs continue to build new ova. E. bathypelagica carried between 86 and 128 eggs in both marsupial sacs. E. bathyantarctica had only between 8 and 13 eggs. Although self-fertilization seemed at least to be possible in E. bathyantarctica, cross-fertilization appears to be more common in both species, as most individuals developed testes and ovaries consecutively.  相似文献

Lipoic acid (LA) is a dithiol-containing cofactor that is essential for the function of α-keto acid dehydrogenase complexes. LA acts as a reversible acyl group acceptor and ‘swinging arm’ during acyl-coenzyme A formation. The cofactor is post-translationally attached to the acyl-transferase subunits of the multienzyme complexes through the action of octanoyl (lipoyl): N-octanoyl (lipoyl) transferase (LipB) or lipoic acid protein ligases (LplA). Remarkably, apicomplexan parasites possess LA biosynthesis as well as scavenging pathways and the two pathways are distributed between mitochondrion and a vestigial organelle, the apicoplast. The apicoplast-specific LipB is dispensable for parasite growth due to functional redundancy of the parasite''s lipoic acid/octanoic acid ligases/transferases. In this study, we show that LplA1 plays a pivotal role during the development of the erythrocytic stages of the malaria parasite. Gene disruptions in the human malaria parasite P. falciparum consistently were unsuccessful while in the rodent malaria model parasite P. berghei the LplA1 gene locus was targeted by knock-in and knockout constructs. However, the LplA1(−) mutant could not be cloned suggesting a critical role of LplA1 for asexual parasite growth in vitro and in vivo. These experimental genetics data suggest that lipoylation during expansion in red blood cells largely occurs through salvage from the host erythrocytes and subsequent ligation of LA to the target proteins of the malaria parasite.  相似文献

Interaction of zoospores of Ulva linza with cationic, arginine-rich oligopeptide self-assembled monolayers (SAMs) is characterized by rapid settlement. Some spores settle (ie permanently attach) in a ‘normal’ manner involving the secretion of a permanent adhesive, retraction of the flagella and cell wall formation, whilst others undergo ‘pseudosettlement’ whereby motile spores are trapped (attached) on the SAM surface without undergoing the normal metamorphosis into a settled spore. Holographic microscopy was used to record videos of swimming zoospores in the vicinity of surfaces with different cationic oligopeptide concentrations to provide time-resolved insights into processes associated with attachment of spores. The data reveal that spore attachment rate increases with increasing cationic peptide content. Accordingly, the decrease in swimming activity in the volume of seawater above the surface accelerated with increasing surface charge. Three-dimensional trajectories of individual swimming spores showed a ‘hit and stick’ motion pattern, exclusively observed for the arginine-rich peptide SAMs, whereby spores were immediately trapped upon contact with the surface.  相似文献

Frogs of the family Mantellidae are endemic to Madagascar and the Comoran island of Mayotte. Like many other animals in this biogeographical region, they have passed through millions of years of isolated evolution which led to ecological, physiological and anatomical specialization. The present study compares the intranasal anatomy of a mantellid, the Malagasy Common Marsh Frog (Mantidactylus betsileanus Boulenger, 1882) with that of the Malaysian Green Flying Frog (Rhacophorus reinwardtii Schlegel, 1840), a representative of the sister group of mantellids (the family Rhacophoridae). Histological examination revealed that the structure of the nasal cavities of M. betsileanus strongly deviates from the usual nasal morphology of anurans. In the typical condition, to which also R. reinwardtii conforms, the two parts of the nasal cavity (main chamber and accessory chambers), containing two different chemosensory systems (main olfactory organ and vomeronasal organ respectively), are connected by a slit-like longitudinal opening. In M. betsileanus, this elongated opening is almost completely reduced. Therefore, main chamber and accessory nasal chambers are markedly separated anatomically, leading to an enhanced spatial segregation of the two different organs of smell. Whether these anatomical alterations correspond to a more significant role of vomeronasal perception and might be related to the presence of characteristic pheromone-producing femoral glands in mantellid frogs requires further study.  相似文献
