A rapid and reproducible enzymatic rate assay for the quantitative determination of the concentration of active sites is presented for the alleloenzymes AdhS and AdhF from Drosophila melanogaster. Using this procedure the turnover numbers as catalytic-center activities were found to be 12.2 sec–1 for AdhF and 3.4 sec–1 for AdhS with secondary alcohols. This showed a slower dissociation of the coenzyme from the binary enzyme-NADH complex with AdhS and hence a stronger binding of NADH to this alleloenzyme. With ethanol, the catalytic-center activity was 1.4 sec–1 for AdhS and 2.8 sec–1 for AdhF, and hence the single amino acid mutation distinguishing the two alleloenzymes also affected hydride transfer.  相似文献

0-group Carcinus maenas (L.) was investigated from June 1975 to September 1976 on a shallow sandy bottom at Kvarnbukten Bay, Gullmar Fjord (58° 15′N: 11°28′E), Sweden, at an average salinity of 25% and a range of monthly mean temperatures of −0.3 to 197. °C.

The new year-class settles from August to early September at a carapace breath of 2 to 3 mm and a calorific content of 32 cal. The distribution is restricted to clusters of the mussel Mytilus edulis L. Depth, type of substratum, and patches of the eel-grass Zostera marina L. are of no importance for their spatial distribution. There is no migration to deeper water in the autumn. The carapace breadth is ≈ 9.5 mm after one year of benthic life. Sexual maturity is reached after two years. Growth occurs at temperatures above 10 °C, i.e., from August to October and from May to July. During the first year of benthic life the animals moult 7 times. The 0-group seems to be micro-carnivores feeding on the sediment meiofauna.

The individual energy budget for the first year of benthic life is: consumption (Cc) 905 cal., production (P1c) 236 cal., cast carapaces (P2c) 153 cal., respiration (Rc) 404 cal., and rejectiction (Fc) 112 cal. The assimilation efficiency (Uc−1) is 88%, the gross growth efficiency (K1c) 43%, and the net growth efficiency (K2c) 49%.

At Kvarnbukten Bay there are large variations in size between the separate year-classes. The energy content of the food consumed by the 1975/76 cohort was used as follows: 4% was stored in living biomass after one year, 36% was released to other trophic levels as dead animals and cast carapaces, 13% rejected as faeces, and 47% was lost through respiration.  相似文献

Optimization of batch pyrite bioleaching with Sulfolobus acidocaldarius was performed using statistical modelling and experimental design. First a screening design was made followed by response surface modelling. The dominating factors identified were pH, pulp density and particle size. The highest batch leaching rate after optimization was 270 mg iron·l–1·h–1 for 6% (w/v) pulp density, pH = 1.5 and particle size <20 m. This represents a 3.5-fold increase from the leaching rate of 80 mg iron·l–1·h–1 obtained under our standard laboratory conditions. Correspondence to: E. B. Lindström  相似文献

In humans and other primates, violent actions performed by victims of aggression are often directed toward an individual or object that is not the source of provocation. This psychological phenomenon is often called displaced aggression. We demonstrate that displaced aggression is either rooted in evolutionarily conserved behavioral and neuroendocrine mechanisms, or represent a convergent pattern that has arisen independently in fish and mammals. Rainbow trout that briefly encountered large, aggressive fish reacted with increased aggression toward smaller individuals. There was a strong negative correlation between received aggression and behavioral change: Individuals subjected to intense aggression were subdued, while moderate assaults induced strong agitation. Patterns of forebrain serotonin turnover and plasma cortisol suggest that the presence of socially subordinate fish had an inhibitory effect on neuroendocrine stress responses. Thus, subordinate individuals may serve as stress-reducing means of aggressive outlet, and displaced aggression toward such individuals appears to be a behavioral stress coping strategy in fishes.  相似文献

Abstract: The presence of multiple [3H] GBR-12935 binding sites in the human brain has been revealed in several recent studies. One site represents the dopamine uptake site. In rat brain it was demonstrated that [3H] GBR-12935 also binds to nondopaminergic "piperazine acceptor sites." One of these sites has been identified as cytochrome P450IID1 in canine brain. [3H] GBR-12935 binding to the piperazine acceptor sites in the human brain was investigated in the present study. A pharmacological definition of the piperazine acceptor sites is presented: the [3H]- GBR-12935 binding fraction that could be discriminated by 10 μ M GBR-12909 in the presence of 0.3 μ M mazindol. This binding fraction was saturable, with binding affinity in the range of 3–8 n M. It was also demonstrated that the piperazine acceptor or cytochrome P450-sensitive drugs cis -flupentixol and proadifen (SKF 525 A) compete for the same binding sites, suggesting the cytochrome P450 nature of the binding. The findings presented support the proposal that at least part of this fraction represents cytochrome P450IID6, the human form of P450IID1. The distribution of [3H] GBR-12935 binding to the suggested P450IID6-site in 12 brain regions was examined, without significant differences in binding densities between the regions. The significance of the present findings on the cytochrome P450 system in brain is discussed.  相似文献

The importance of proteoglycans for secretion of proteolytic enzymes was studied in the murine macrophage cell line J774. Untreated or 4beta-phorbol 12-myristate 13-acetate (PMA)-stimulated macrophages were treated with hexyl-beta-d-thioxyloside to interfere with the attachment of glycosaminoglycan chains to their respective protein cores. Activation of the J774 macrophages with PMA resulted in increased secretion of trypsin-like serine proteinase activity. This activity was completely inhibited by plasminogen activator inhibitor 1 and by amiloride, identifying the activity as urokinase plasminogen activator (uPA). Treatment of both the unstimulated or PMA-stimulated macrophages with xyloside resulted in decreased uPA activity and Western blotting analysis revealed an almost complete absence of secreted uPA protein after xyloside treatment of either control- or PMA-treated cells. Zymography analyses with gels containing both gelatin and plasminogen confirmed these findings. The xyloside treatment did not reduce the mRNA levels for uPA, indicating that the effect was at the post-translational level. Treatment of the macrophages with xylosides did also reduce the levels of secreted matrix metalloproteinase 9. Taken together, these findings indicate a role for proteoglycans in the secretion of uPA and MMP-9.  相似文献

In a prospective study 29 patients with urinary tract infections caused by sulphonamide-sensitive organisms were treated with a single oral dose of the short-acting sulphonamide sulphafurazole. Twenty-seven (93%) of the 29 patients--and possibly all 29--were cured of their infections. There was no difference in the recurrence rates after single-dose treatment and treatment for 10 days or more. Six out of eight strains of Escherichia coli causing early recurrences were sensitive to sulphonamides. These results suggest that uncomplicated infections may safely and successfully be treated by a single oral dose of a short-acting sulphonamide.  相似文献
