Incisor abnormalities such as loss of enamel color, hypoplasia of enamel, shortening and lengthening, irregular shape of edge, and fracture were often observed in SAM-P/2/Iw (senescence accelerated mouse-prone) more than 12 months old. On the other hand, for SAM-R/1/lw (control) mice more than 20 months old, there were only a few instances of loss of enamel color. The incidence of incisor abnormality was significantly different between P/2/Iw and R/I/Iw. The onset of abnormality was also earlier in P/2/Iw. Histologically, dens in dente and odontoma-like structures were occasionally found in the incisors of P/2/Iw. These findings add further supporting evidence that SAM-P/2/Iw is truly senescence accelerated. 相似文献

Prostaglandin E2 (PGE2) stimulated cAMP production in the MOB 3-4-F2 cell line, a subclone of the osteoblast-like MOB 3-4 cell line. After being cultured in alpha-minimum essential medium supplemented with 10% heat-inactivated foetal calf serum (HIFCS), cells responded to PGE2 (greater than or equal to 50 ng/ml) with a small, but significant, increase in cAMP production. This response did not vary with duration of culture. In 2% HIFCS-containing medium, despite their lower basal cAMP level, cells responded to PGE2 (greater than or equal to 5 ng/ml) with strikingly increased cAMP production. In addition, prolonged culture in this serum-deficient medium enhanced this response. On the other hand, culture of cells in 2% HIFCS-containing medium decreased the apparent number of PGE2 receptors, which was also enhanced by prolonged culture, without effect on their apparent affinity. Their number in 10% HIFCS-containing medium, more than that in 2% HIFCS-containing medium, was almost constant, independent of the culture period. Starvation of MOB 3-4-F2 cells in serum-deficient medium, therefore, appeared to down-regulate PGE2 receptors but increase the cAMP response to PGE2. Moreover, prolonged starvation of cells appeared to facilitate these phenomena. A heat-stable factor has been extracted from the microsomal membranes of rat brain that inhibits the activities of rat brain ceramide galactosyl- and glucosyltransferases. It is nondialyzable, susceptible to proteolytic enzymes but resistant to DNase and RNase, and has no effect on lysosomal hydrolytic enzymes. The factor could also be prepared from microsomes of bovine, mouse, and rabbit brains and in much lower concentrations, from systemic organs of rats. This inhibitor might play a role in the regulation of ceramide glycosyltransferases in vivo. 相似文献

To evaluate the role of pyrophosphate-dependent phosphofructokinase (PFP. EC 2.7.1.90) in the sugar metabolism of pollen. its occurrence and properties were studied in pollen grains of several plants including camellia ( Camellia japonica L.). In all pollen samples, PFP was strongly activated by fructose-2,6-bisphosphate (F2,6BP), and the activity of F2,6BP-activated PFP was higher than that of phosphofructokinase (PFK. EC 2.7.1.11). PFP partially purified from camellia pollen required Mg2+ for activity with an optimum at 1 m M . and was almost unaflected by a variety of metabolites at 1 m M . Its molecular mass was around 220 kDa, and apparent Km values for F6P, PPi . F1, 6BP and Pi were 294, 4, 20 and 580 u M , respectively. The levels of F2.6BP. PPi and F6P in camellia pollen were sufficent to support the forward reaction by PFP, and PFP, was 20- to 40-fold more active than PFK during pollen growth. These results suggest that pollen PFP plays a role in glycolysis but not gluconeogenesis. and the possible relevance of this to pollen tube growth is discussed. 相似文献

The heat denaturation and renaturation curves of rat liver and ascites hepatoma (AH 108A) chromatins were measured. In these renaturation curves, there are small sigmoidal regions. These sigmoidal regions remained in redenaturation curves and were largely stable to DNAase I digestion. When the chromatins were treated stepwise with NaClO4 and lysine-rich histones were removed, the sigmoidal regions in the renaturation curves disappeared. These results suggested that the sigmoidal regions reflected the interaction of DNA and lysine-rich histones. 相似文献
