The introduction of West Nile virus (WNV) into North America has been associated with relatively high rates of neurological disease and death in humans, birds, horses, and some other animals. Previous studies identified strains in both genetic lineage 1 and genetic lineage 2, including North American isolates of lineage 1, that were highly virulent in a mouse neuroinvasion model, while other strains were avirulent or significantly attenuated (D. W. C. Beasley, L. Li, M. T. Suderman, and A. D. T. Barrett, Virology 296:17-23, 2002). To begin to elucidate the basis for these differences, we compared a highly virulent New York 1999 (NY99) isolate with a related Old World lineage 1 strain, An4766 (ETH76a), which is attenuated for mouse neuroinvasion. Genomic sequencing of ETH76a revealed a relatively small number of nucleotide (5.1%) and amino acid (0.6%) differences compared with NY99. These differences were located throughout the genome and included five amino acid differences in the envelope protein gene. Substitution of premembrane and envelope genes of ETH76a into a NY99 infectious clone backbone yielded a virus with altered in vitro growth characteristics and a mouse virulence phenotype comparable to ETH76a. Further site-specific mutagenesis studies revealed that the altered phenotype was primarily mediated via loss of envelope protein glycosylation and that this was associated with altered stability of the virion at mildly acidic pH. Therefore, the enhanced virulence of North American WNV strains compared with other Old World lineage 1 strains is at least partly mediated by envelope protein glycosylation.  相似文献

Polystyrene culture dishes and polystyrene microcarriers were coated with Pronectin-F and poly-l-lysine (polylysine), either alone or in combination. Pronectin-F is a recombinant peptide containing repeats of the RGD cell-attachment sequence from fibronectin. Polylysine is a polymer ofl-lysine. Pronectin-F supported attachment of Madin-Darby Canine Kidney (MDCK) cells at concentrations as low as 0.025 g/cm2 of surface area. The cells rapidly spread after attachment. Polylysine at concentrations of 0.05–0.5 g/cm2 also supported cell attachment but cells did not rapidly spread after attachment to this substrate. Higher concentrations of polylysine could not be used because of toxicity. When the two peptides were used in conjunction, MDCK cells attached and spread at lower peptide concentrations than they did when either substrate was used alone. These findings suggest that recombinant Pronectin-F alone or in conjunction with a cationic polymer could be a useful replacement for materials such as gelatin or collagen which are currently used as microcarrier surfaces.  相似文献

Question. Can strategic burning, targeting differing ecological characteristics of native and exotic species, facilitate restoration of native understorey in weed‐invaded temperate grassy eucalypt woodlands? Location. Gippsland Plains, eastern Victoria, Australia. Methods. In a replicated, 5‐year experimental trial, the effects of repeated spring or autumn burning were evaluated for native and exotic plants in a representative, degraded Eucalyptus tereticornis grassy woodland. Treatments aimed to reduce seed banks and modify establishment conditions of exotic annual grasses, and to exhaust vegetative reserves of exotic perennial grasses. Treatments were applied to three grassland patch types, dominated by the native grass Austrodanthonia caespitosa, ubiquitous exotic annuals, or the common exotic perennial grass Paspalum dilatatum. Results. The dominant native grass Austrodanthonia caespitosa and native forbs were resilient to repeated fires, and target exotic annuals and perennials were suppressed differentially by autumn and spring fires. Exotic annuals were also suppressed by drought, reducing the overall treatment effects but indicating important opportunities for restoration. The initially sparse exotic geophyte Romulea rosea increased in cover with fire and the impact of this species on native forbs requires further investigation. There was minimal increase in diversity of subsidiary natives with fire, probably owing to lack of propagules. Conclusions. While fire is often considered to increase ecosystem invasibility, our study showed that strategic use of fire, informed by the relative responses of available native and exotic taxa, is potentially an effective step towards restoration of weed‐invaded temperate eucalypt woodlands.  相似文献

We have identified possible mechanisms for the degradation of oat phytochrome A (PHYA) mRNA. The majority of PHYA mRNA molecules appeared to be degraded prior to removal of the poly(A) tail, a pathway that differs from that reported for the degradation of other eukaryotic mRNAs. Polyadenylated PHYA mRNA contained a pattern of putative degradation products that is consistent with a 5'-->3' exoribonuclease, although the participation of a stochastic endoribonuclease cannot be excluded. The poly(A) tail of PHYA mRNA was heterogeneous in size and ranged from approximately 14 to 220 nucleotides. Early PHYA mRNA degradation events did not appear to involve site-specific endoribonucleases. Approximately 25% of the apparently full-length PHYA mRNA was poly(A) deficient. Oat H4 histone, beta-tubulin, and actin mRNA populations had lower amounts of apparently full-length mRNAs that were poly(A) deficient. Degradation of the poly(A)-deficient PHYA mRNA, a second pathway, appeared to be initiated by a 3'-->5' exoribonucleolytic removal of the poly(A) tail followed by both 5'-->3' and 3'-->5' exoribonuclease activities. Polysome-associated RNA contained putative PHYA mRNA degradation products and was a mixture of polyadenylated and deadenylated PHYA messages, suggesting that the two distinct degradation pathways are polysome associated.  相似文献

Montane temperate forests of the Cordillera de Piuchué Ecosystem Study, Isla Chiloé, Chile, are unaffected by air pollution, timber exploitation and agricultural clearing, and the current floristic assemblage has been relatively stable for the past 7500 years. The apparent absence of major perturbation at this location makes it an appropriate baseline site for ecosystem analysis. We measured soil bulk density, pH, soil organic C (SOC), total N, and NH4Cl–exchangeable cations (Ca+2, Mg+2, K+, Na+, Al+3) in 0–10 and 10–40 cm depth samples from 72 soil profiles representing three vegetation zones: Fitzroya cupressoides Forest, Pilgerodendron uvifera–Tepualia stipularis Forest, and Magellanic Moorland. Fitzroya and Pilgerodendron–Tepualia Forests were indistinguishable for all measured soil characteristics (P > 0.05, Dunn's multiple comparison test on ranked data); these included very high median SOC concentrations (0–10 cm = 49.6%) and correspondingly low bulk density values (0–10 cm = 0.07). Moorland soil median values (0-10 cm) were significantly higher for bulk density (0.12) and lower for SOC (28.5%), but not for total N (Forests = 0.99%, Moorland = 0.95%), resulting in lower median C:N ratios for the moorland (Forests = 44.7; Moorland = 30.3). Across both depths and all three vegetation zones regression analysis indicated that SOC was an excellent predictor (R2; = 0.93, P < 0.001) of (exchangeable Ca+2 + Mg+2 + K+ + Na+). Comparison with other old growth montane environments indicates that the Fitzroya and Pilgerodendron–Tepualia soil profiles are characterized by C:N ratios typical of other relatively unpolluted conifer forest soils (33.0–49.3). Soil profiles of representative polluted montane conifer forests have lower C:N ratios (16.2–23.5). Organic horizons from representative polluted montane conifer forests also retain fewer exchangeable base cations per unit SOC than are retained by organic horizons from the Cordillera de Piuchué forests.  相似文献

We evaluated whether IL-4, a cytokine critical for inducing allergic responses, also contributes to the effector phase of allergy. Pretreatment of mice with IL-4 or the related cytokine, IL-13, rapidly and dramatically increased the severity of anaphylaxis induced by cross-linking Fc(epsilon)RI or FcgammaRIII. This effect was inhibited by endogenously produced IFN-gamma, was T cell-, B cell-, and common gamma-chain-independent, and required IL-4Ralpha and Stat6. IL-4Ralpha signaling also enhanced anaphylaxis in mice infected with a nematode parasite that stimulates IL-4/IL-13 production. IL-4 exacerbated anaphylaxis by acting synergistically with vasoactive mediators to increase vascular permeability. Synergy between IL-4 and vasoactive mediators during the effector phase of allergic inflammation may both contribute to allergic immunopathology and enhance protective immunity against gastrointestinal worms.  相似文献

1. In a recent study, we distinguished two classes of amphiphilic AChE3 dimers in Torpedo tissues: class I corresponds to glycolipid-anchored dimers and class II molecules are characterized by their lack of sensitivity to PI-PLC and PI-PLD, relatively small shift in sedimentation with detergent, and absence of aggregation without detergent. 2. In the present report, we analyze the amphiphlic or nonamphiphilic properties of globular AChE forms in T28 murine neural cells, rabbit muscle, and chicken muscle. The molecular forms were identified by sucrose gradient sedimentation in the presence and absence of detergent and analyzed by nondenaturing charge-shift electrophoresis. Some amphiphilic forms showed an abnormal electrophoretic migration in the absence of detergent, because of the retention of detergent micelles. 3. We show that the amphiphilic monomers (G1a) from these tissues, as well as the amphiphilic dimers (G2a) from chicken muscle, resemble the class II dimers of Torpedo AChE. We cannot exclude that these molecules possess a glycolipidic anchor but suggest that their hydrophobic domain may be of a different nature. We discuss their relationship with other cholinesterase molecular forms.  相似文献

Infection of Bacillus subtilis by PBS2 phage, whose DNA contains uracil instead of thymine, is relatively unaffected by low concentrations of nalidixic acid which severely inhibit B. subtilis DNA synthesis. High concentrations of nalidixic acid do inhibit PBS2 DNA synthesis, but more severely reduce the burst size of PBS2 infections. Twenty-four-day-old weanling male and female rats were either lesioned in the septal area, gonadectomized, lesioned and gonadectomized, or not treated. Tests for lordotic behavior were carried out at 27 and 28 days of age after priming with estradiol alone and with estradiol plus progesterone. A second set of tests for lordotic behavior was carried out at 47 and 48 days of age. In the interim period, some of the animals were given chronic estrogen treatment. In tests given at 27 days of age, septal lesions facilitated lordotic responding after estrogen priming; no differences were observed between male and female animals. At 47 and 48 days of age, however, unless male rats had been exposed to chronic estrogens following septal lesioning, no facilitation of estrogen-induced lordotic behavior occurred. In addition, it was found that female rats gonadectomized at 24 days of age and given no exposure to estrogens between the tests at 27 and 28 days and those at 47 and 48 days of age showed reduced sensitivity to estrogens, as compared to normal or estrogen-treated females, whether lesioned or not.  相似文献
