Primary structure of the southern bean mosaic virus coat protein: First results

Studies on the southern bean mosaic virus coat protein have established the molecular weight of this protein, its amino acid composition, the nature of its C-terminal amino acid, and the blockage of the N-terminal residue by an acetyl group. After hydrolysis of the protein by trypsin, the hydrolysate was fractionated by ion-exchange chromatography. Among the purified tryptic peptides were isolated the N- and the C-terminal peptides where sequences were determined, principally by mass spectrometry.     

Humirianthenolides,new degraded diterpenoids from Humirianthera rupestris

The tuber of Humirianthera rupestris (Icacinaceae) contains the degraded diterpenoids 3β,20-epoxy-30α- hydroxy- 14-oxo-9β-podocarpan-19,6β-olide (humirianthenolide A), 3β,20-epoxy-3α,14α-dihydroxy-9β-podocarpan-19,6β- olide (humirianthenolide B), 3β,20; 16,14-diepoxy-3α-hydroxy-17-nor-15-oxo-9β-abiet-13-en-19,6β-olide (humirianthenolide C), 3β,20-epoxy-3α,14-dihydroxy-13-oxo-9β-podocarp-8(14)-en-19,6β-olide (humirianthenolide D), 3β,20-epoxy-3α-hidroxy-14-oxo-8α,9β-podocarpan-19,6β-olide (humirianthenolide E) and 3β,20-epoxy-3α,14β- dihydroxy-8α,9β-podocarpan-19,6β-olide (humirianthenolide F). ¹H NMR and ¹³C NMR spectroscopy were efrective for the determination of the humirianthenolide structures.     

Biochemical changes in tissue composition of Periplaneta americana (Linn.) acclimated to high and low temperatures

1. 1.|Cold acclimation apparently favours an increase of water content in fat body, but not in coxal muscle, of cockroaches.

2. 2.|A remarkable enhancement in the accumulation of total protein in fat body characterizes the cold acclimation of cockroaches, particularly adult males (175% increase in protein/DNA ratio). The increase in protein content of coxal muscle during acclimination to 15°C, observed in nymphs (16%) and males (16%) but not in females, is less pronounced than that of fat body.

3. 3.|A diminution (28–32%) in the free amino acid/DNA ratio due to cold acclimation has been recorded in both coxal muscle and fat body of nymphs and females, but not in males.

4. 4.|No qualitative change occurs in the free amino acid spectrum of haemolymph and tissues of this insect during acclimation to 15 and 35°C.

5. 5.|An augmentation (15–30%) of the RNA/DNA ratio occurs in fat body and coxal muscle of nymphs and males but in fat body alone of females following cold acclimation.

6. 6.|The glycogen reserve has been shown to increase by up to 30% in fat body and coxal muscle of cold acclimated cockroaches compared to warm acclimated ones.

Calcium gluconate production by a nonconventional fermentation method

Summary A strain of Aspergillus niger was grown in still (liquid), shake and semi-solid fermentation for calcium gluconate production from glucose, starch, or molasses. The yield from glucose or starch hydrolysate was acceptably high in both shake and semi-solid fermentation indicating that the semi-solid fermentation process offers a promising practical alternative.     

A hepatic membrane-associated factor stimulates nuclear DNA synthesis in cultured fibroblastic cells

Nuclear DNA replication in cultured mouse fibroblasts is stimulated by isolated hepatic plasma membranes in a time- and concentration-dependent manner. The plasmalemmal activity is susceptible to trypsin treatment, and to treatment with protein modifying agents, N-ethylmaleimide, N-bromosuccinimide, and 2-hydroxy-5-nitro-benzylbromide.     

Synthèse et étude conformationelle par r.m.n.-13c de β-nucléosides pyrimidiques contenant une ou deux sous-unités hexopyranosyles: Application à la conformation de l'anthelmycine

Several new peracetylated pyrimidine N-1 nucleosides containing one or two β-d-hexopyranosyl residues have been synthesized and their physical characteristics are reported. The conformation of these nucleosides has been investigated by high-resolution ¹³C-n.m.r. analysis after deblocking. It was possible to distinguish between the syn and anti rotamers of 3-β-d-glucopyranosyluracil, and to establish the favored anti conformation in solution of the nucleoside disaccharide, antibiotic anthelmycin.     

A computer program for the analysis of crossover designs.

We describe a program, CROSS, which we have written to obtain potency estimates and other parameters for bioassay data from assays of crossover design. The program permits testing of all assays for statistical validity and calculates the complete analysis of variance for assays of balanced design. The form of data input and the complete documentation of assay results make this program particularly useful for anyone carrying out crossover assays on a routine basis. The analysis of variance presented is also useful for more general biological or medical situations.
