Background
We determined antibodies to the pandemic influenza A (H1N1) 2009 virus in children to assess: the incidence of (H1N1) 2009 infections in the 2009/2010 season in Germany, the proportion of subclinical infections and to compare titers in vaccinated and infected children.Methodology/Principal Findings
Eight pediatric hospitals distributed over Germany prospectively provided sera from in- or outpatients aged 1 to 17 years from April 1st to July 31st 2010. Vaccination history, recall of infections and sociodemographic factors were ascertained. Antibody titers were measured with a sensitive and specific in-house hemagglutination inhibition test (HIT) and compared to age-matched sera collected during 6 months before the onset of the pandemic in Germany. We analyzed 1420 post-pandemic and 300 pre-pandemic sera. Among unvaccinated children aged 1–4 and 5–17 years the prevalence of HI titers (≥1∶10) was 27.1% (95% CI: 23.5–31.3) and 53.5% (95% CI: 50.9–56.2) compared to 1.7% and 5.5%, respectively, for pre-pandemic sera, accounting for a serologically determined incidence of influenza A (H1N1) 2009 during the season 2009/2010 of 25,4% (95% CI : 19.3–30.5) in children aged 1–4 years and 48.0% (95% CI: 42.6–52.0) in 5–17 year old children. Of children with HI titers ≥1∶10, 25.5% (95% CI: 22.5–28.8) reported no history of any infectious disease since June 2009. Among vaccinated children, 92% (95%-CI: 87.0–96.6) of the 5–17 year old but only 47.8% (95%-CI: 33.5–66.5) of the 1–4 year old children exhibited HI titers against influenza A virus (H1N1) 2009.Conclusion
Serologically determined incidence of influenza A (H1N1) 2009 infections in children indicates high infection rates with older children (5–17 years) infected twice as often as younger children. In about a quarter of the children with HI titers after the season 2009/2010 subclinical infections must be assumed. Orotidine-5-monophosphate decarboxylase (OMPdecase) catalyzes the final step in pyrimidine biosynthesis, the conversion of orotidine-5-monophosphate (OMP) to uridine-5-monophosphate. ThepyrF gene, encoding OMPdecase, was isolated from a chromosomal library ofPseudomonas aeruginosa PAO1 by screening for complementation of anEscherichia coli and aP. aeruginosa pyrF mutant. The nucleotide sequence of a 2510-bp chromosomal DNA fragment, complementing both strains, was determined (EMBL accession number X65613). On this a 696-bp open reading frame capable of encoding the 24 kDa OMPdecase was identified. Despite a generally good correspondence to other OMPdecase sequences, theP. aeruginosa gene was unique in that it did not constitute part of an operon. ThepyrF gene was amplified by polymerase chain reaction, overexpressed in the pT7-7/E. coli BL21(DE3) system and purified to near electrophoretic homogeneity by anion exchange chromatography. The phase behaviour of cuticular waxes from leaves of Hedera helix L. and Juglans regia L. was studied by Fourier transform infrared spectroscopy. For this purpose reconstituted waxes, isolated cuticular membranes,
dewaxed polymer matrix membranes and whole leaves were studied in the horizontal attenuated total reflection and transmission
modes. Melting curves of cuticular waxes were derived from temperature-dependent changes in the absorption maximum of the
symmetric stretching mode of CH2 groups (νs, at approx. 2856–2848 cm−1). With increasing temperature absorption band doublets due to CH2 scissoring (δsciss) and rocking (δrock) movements (at approx. 1473–1471 and 730–720 cm−1, respectively) indicative of an orthorhombic arrangement of alkyl chains merged into a single peak. The area ratio of the
peaks at approx. 720 and 730 cm−1 was used as a measure for aliphatic crystallinity of plant cuticular waxes at a given temperature. The investigations of
reconstituted cuticular waxes and those still embedded in isolated cuticles or in situ on the leaf produced comparable results.
The findings are discussed in terms of the properties of the cuticular transport barrier.

Receptor-like kinases (RLKs) that function as pattern-recognition receptors (PRRs) play a key role in plant immune responses. The receptor recognizing flagellin in Arabidopsis, FLS2, is encoded by a membrane resident RLK. FLS2 is involved in preinvasive immunity against bacterial infection. Recent observations revealed that upon flagellin perception FLS2 accumulates in intracellular mobile vesicles and is then degraded. Reminiscent of ligand-induced receptor endocytosis in animals, FLS2 internalization is Wortmannin-sensitive. Mutation of the potentially phosphorylated residue threonine-867 impaired FLS2 endocytosis and flagellin-triggered responses. Furthermore, mutation of a PEST-motif abolished FLS2 endocytosis and downstream flagellin-elicited responses were affected. Thus, FLS2 endocytosis likely involves phosphorylation and ubiquitination events and appears to be interconnected with flagellin signaling. Similarly, TLR4, the mammalian PRR recognizing bacterial lipopolysaccharides (LPS) is internalized in a ligand specific manner. PURPOSE: Despite therapeutic improvements, all patients with nonresectable metastatic colorectal cancer (mCRC) acquire resistance to treatment probably due to the growth of mutated clones. In contrast to tissue-based studies, liquid biopsies have enabled the opportunity to reveal emerging resistance to treatment by detecting mutated clones and noninvasively monitoring clonal dynamics during therapy. METHODS: The courses of three patients with mCRC who were initially RAS wild-type were monitored longitudinally using liquid biopsy with long-term follow-up of up to 20 sequential samples. Detection of fragmented RAS mutated circulating cell-free DNA (cf)DNA in plasma was performed by BEAMing. In addition, plasma digital droplet PCR was used to detect and quantify BRAF and PIK3CA mutated cfDNA. Changes of mutational load were correlated with imaging data. RESULTS: A combination of liquid biopsy and radiological imaging enabled visualization of the occurrence of clonal redistribution after discontinuation of anti-EGFR mAb therapy, as well as emerging RAS mutations during therapy with anti-EGFR mAb indicating resistance. Furthermore, we found that growth of RAS mutated clones is independent of direct selective pressure by anti-EGFR therapy, which is a significant and new finding of this study. CONCLUSIONS: Our findings demonstrated the whole spectrum of clonal selection and redistribution of mutated cell clones leading to acquired resistance. Given our observation that the growth of RAS mutated clones can evolve even in the absence of anti-EGFR mAb therapy, there is a clear imperative to monitor RAS mutations in serial blood draws in all RAS wild-type patients in general and independent of the therapy. 相似文献

The analogy between H-bonded nucleobase pairs and their metalated analogues is extended to the hemiprotonated pair of 7,9-dimethylguanine
(7,9-DimeG) and the Watson-Crick and reversed Watson-Crick pair between 7,9-dimethylguaninium (7,9-DimeGH+) and 1-methylcytosine (1-MeC). The crystal structure analyses of two model compounds, trans–[Pt(CH3NH2)2(7,9-DimeG-N1)2](NO3)2 (1) and trans–[Pt(NH3)2(1-MeC-N3)(7, 9-DimeG-N1)](PF6)2· 2.5 H2O (3a) are reported. Pt binding is through N1 of 7,9-DimeG and N3 of 1-MeC. In solution, 3a exists in a mixture with Watson-Crick and reversed Watson-Crick arrangements of the two bases, depending on solvent, concentration
