The present work evaluates the influence of fish production on macrobenthic colonization over large areas (approximately 700 m2), where the colonizing populations are not nearby the disturbed area. Sampling was undertaken within newly created aquaculture earthen ponds under two contrasting conditions: white seabream (Diplodus sargus) production and no production (control). Macrobenthic and geochemical samples were collected 7, 23, 54, 93 and 180 days after filling the earthen ponds with seawater pumped from a water reservoir for the first time. The water reservoir was also sampled, and is used as a reference for the colonizing populations. Macrobenthic colonization rate in the ponds was low, probably due to the isolation of the disturbed habitat, to the large size of the defaunated area, and possibly to geochemical constraints. Initial colonization was by insect larvae (mainly chironomids), the bivalves Cerastoderma spp., the polychaetes Pseudopolydora paucibranchiata and Hydrodoides elegans, and nemerteneans. The number of species was similar in control and production ponds, even though under production higher total abundance values were observed. Although well represented in the water reservoir, the amphipod Microdeutopus gryllotalpa was only observed within the new ponds after 6 months. Preliminary results suggest that macrobenthic colonization patterns were influenced by fish production, as assemblages were significantly different among ponds. Higher food availability due to fish production may explain the results obtained, but ecological reasons, such as predation, may also contribute for shaping the macrobenthic communities. 相似文献

Here, the leaf photoacclimatory plasticity and efficiency of the tropical seagrass Thalassia testudinum were examined. Mesocosms were used to compare the variability induced by three light conditions, two leaf sections and the variability observed at the collection site. The study revealed an efficient photosynthetic light use at low irradiances, but limited photoacclimatory plasticity to increase maximum photosynthetic rates (P(max)) and saturation (E(k)) and compensation (E(c)) irradiances under high light irradiance. A strong, positive and linear association between the percentage of daylight hours above saturation and the relative maximum photochemical efficiency (F(V)/F(M)) reduction observed between basal and apical leaf sections was also found. The results indicate that T. testudinum leaves have a shade-adapted physiology. However, the large amount of heterotrophic biomass that this seagrass maintains may considerably increase plant respiratory demands and their minimum quantum requirements for growth (MQR). Although the MQR still needs to be quantified, it is hypothesized that the ecological success of this climax species in the oligotrophic and highly illuminated waters of the Caribbean may rely on the ability of the canopy to regulate the optimal leaf light environment and the morphological plasticity of the whole plant to enhance total leaf area and to reduce carbon respiratory losses. 相似文献

The central provinces of Argentina are characterized by the presence of a high number of shallow lakes, located in endorheic basins, many of which have elevated salinities as well as eutrophic or hypereutrophic condition. The zooplankton of four saline shallow lakes of the province of La Pampa was studied on a monthly basis during a 2‐year period to determine its temporal and spatial variation. The surface of these shallow lakes (<2.5 m depth) varied between 56.8 and 215.9 ha, and some have from 8.4 to 20.8 g · l–1. The more saline lakes have “clear” water and the less saline lakes “turbid” water. Fishes, Jenynsia multidentata , were present in only two lakes during the last two months of the studied period. The zooplankton was composed of 17 taxa of Rotifera, 5 taxa of Cladocera and 4 taxa of Copepoda. The low diversity and the faunistic composition are characteristic of saline environments. Although the studied lakes share 38% of the species, the faunistic similarity was higher between the two least saline lakes. The lowest diversity was found in the two most saline lakes. All four shallow lakes were characterized by their very high zooplankton density, especially in the least saline lakes (<80000 ind · l–1). The abundance is significantly correlated with the water transparency but not with salinity. The zooplankton temporal variation was characterized by the alternation of macro‐ and microzooplankton, probably regulated by competition and intrazooplanktonic predation. In each lake, the spatial abundance distribution of the macro‐ and microzooplankton was homogeneous. It was related to the shallow depht of the lakes and their polymictic condition. The Scheffer model on alternative states in shallow lakes acknowledges that it cannot be applied to saline lakes because Daphnia , the main responsible for the clear water state, is not tolerant to high salinity. Our study shows that the most saline lakes, where the halophylic Daphnia menucoensis is abundant, have also the most clear waters. Another difference that we found with regards to the mentioned model is that, in turbid lakes, it could not have had a top‐down control on macrozooplankton exerted by fishes because in these lakes fishes were practically absent. The study of cross-reactivity in allergy is key to both understanding. the allergic response of many patients and providing them with a rational treatment In the present study, protein microarrays and a co-sensitization graph approach were used in conjunction with an allergen microarray immunoassay. This enabled us to include a wide number of proteins and a large number of patients, and to study sensitization profiles among members of the LTP family. Fourteen LTPs from the most frequent plant food-induced allergies in the geographical area studied were printed into a microarray specifically designed for this research. 212 patients with fruit allergy and 117 food-tolerant pollen allergic subjects were recruited from seven regions of Spain with different pollen profiles, and their sera were tested with allergen microarray. This approach has proven itself to be a good tool to study cross-reactivity between members of LTP family, and could become a useful strategy to analyze other families of allergens. 相似文献

Two novel dinuclear complexes involving the antihypertensive drug valsartan and copper(II) ion have been prepared in water and DMSO. The complex compositions were determined as: [Cu(vals)(H2O)3]2.6H2O and [Cu(vals)(H2O)2DMSO]2.2H2O. They were thoroughly characterized by elemental and thermal analysis, spectrophotometric titrations and UV-visible, diffuse reflectance, FTIR, Raman and EPR spectroscopies. No effect of the ligand on two tested osteoblastic cell lines in culture (one normal MC3T3E1 and one tumoral UMR106) was observed in concentrations up to 100 μM. Higher concentrations of Valsartan are required to induce cytotoxicity in both cell lines. The antiproliferative effect of the tested complex ([Cu(vals)(H2O)3]2.6H2O) in a dose-response manner, was higher in the UMR106 osteoblastic cell line than that of the MC3T3E1 normal line at concentrations ≥ 100 μM. SUMMARY The genetic basis for the evolution of development includes genes that encode proteins expressed on the surfaces of sperm and eggs. Previous studies of the sperm acrosomal protein bindin have helped to characterize the adaptive evolution of gamete compatibility and speciation in sea urchins. The absence of evidence for bindin expression in taxa other than the Echinoidea has limited such studies to sea urchins, and led to the suggestion that bindin might be a sea urchin-specific molecule. Here we characterize the gene that encodes bindin in a broadcast-spawning asterinid sea star ( Patiria miniata ). We describe the sequence and domain structure of a full-length bindin cDNA and its single intron. In comparison with sea urchins, P. miniata bindin is larger but the two molecules share several general features of their domain structure and some sequence features of two domains. Our results extend the known evolutionary history of bindin from the Mesozoic (among the crown group sea urchins) into the early Paleozoic (and the common ancestor of eleutherozoans), and present new opportunities for understanding the role of bindin molecular evolution in sexual selection, life history evolution, and speciation among sea stars. 相似文献

Antibodies specific for neutralizing epitopes in either Human papillomavirus (HPV) capsid protein L1 or L2 can mediate protection from viral challenge and thus their accurate and sensitive measurement at high throughput is likely informative for monitoring response to prophylactic vaccination. Here we compare measurement of L1 and L2-specific neutralizing antibodies in human sera using the standard Pseudovirion-Based Neutralization Assay (L1-PBNA) with the newer Furin-Cleaved Pseudovirion-Based Neutralization Assay (FC-PBNA), a modification of the L1-PBNA intended to improve sensitivity towards L2-specific neutralizing antibodies without compromising assay of L1-specific responses. For detection of L1-specific neutralizing antibodies in human sera, the FC- PBNA and L1-PBNA assays showed similar sensitivity and a high level of correlation using WHO standard sera (n = 2), and sera from patients vaccinated with Gardasil (n = 30) or an experimental human papillomavirus type 16 (HPV16) L1 VLP vaccine (n = 70). The detection of L1-specific cross-neutralizing antibodies in these sera using pseudovirions of types phylogenetically-related to those targeted by the L1 virus-like particle (VLP) vaccines was also consistent between the two assays. However, for sera from patients (n = 17) vaccinated with an L2-based immunogen (TA-CIN), the FC-PBNA was more sensitive than the L1-PBNA in detecting L2-specific neutralizing antibodies. Further, the neutralizing antibody titers measured with the FC-PBNA correlated with those determined with the L2-PBNA, another modification of the L1-PBNA that spacio-temporally separates primary and secondary receptor engagement, as well as the protective titers measured using passive transfer studies in the murine genital-challenge model. In sum, the FC-PBNA provided sensitive measurement for both L1 VLP and L2-specific neutralizing antibody in human sera. Vaccination with TA-CIN elicits weak cross-protective antibody in a subset of patients, suggesting the need for an adjuvant. 相似文献

To assess whether anorexia nervosa affects mitochondrial function and redox state in leukocytes of anorexic patients, we measured mitochondrial oxygen consumption, membrane potential, reactive oxygen species production, glutathione levels, mitochondrial mass, and complex I and III activity in polymorphonuclear cells.Results
Mitochondrial function was impaired in the leukocytes of the anorexic patients. This was evident in a decrease in mitochondrial O2 consumption (P<0.05), mitochondrial membrane potential (P<0.01) and GSH levels (P<0.05), and an increase in ROS production (P<0.05) with respect to control subjects. Furthermore, a reduction of mitochondrial mass was detected in leukocytes of the anorexic patients (P<0.05), while the activity of mitochondrial complex I (P<0.001), but not that of complex III, was found to be inhibited in the same population.Conclusions
Oxidative stress is produced in the leukocytes of anorexic patients and is closely related to mitochondrial dysfunction. Our results lead us to propose that the oxidative stress that occurs in anorexia takes place at mitochondrial complex I. Future research concerning mitochondrial dysfunction and oxidative stress should aim to determine the physiological mechanism involved in this effect and the physiological impact of anorexia. 相似文献110.

The successful exploitation of germplasm banks, harbouring plant genetic resources indispensable for plant breeding, will depend on our ability to characterize their genetic diversity. The Vegetable Germplasm Bank of Zaragoza (BGHZ) (Spain) holds an important Capsicum annuum collection, where most of the Spanish pepper variability is represented, as well as several accessions of other domesticated and non-domesticated Capsicum spp from all over the five continents. In the present work, a total of 51 C. annuum landraces (mainly from Spain) and 51 accessions from nine Capsicum species maintained at the BGHZ were evaluated using 39 microsatellite (SSR) markers spanning the whole genome. The 39 polymorphic markers allowed the detection of 381 alleles, with an average of 9.8 alleles per locus. A sizeable proportion of alleles (41.2%) were recorded as specific alleles and the majority of these were present at very low frequencies (rare alleles). Multivariate and model-based analyses partitioned the collection in seven clusters comprising the ten different Capsicum spp analysed: C. annuum, C. chinense, C. frutescens, C. pubescens, C. bacatum, C. chacoense and C. eximium. The data clearly showed the close relationships between C. chinense and C. frutescens. C. cardenasii and C. eximium were indistinguishable as a single, morphologically variable species. Moreover, C. chacoense was placed between C. baccatum and C. pubescens complexes. The C. annuum group was structured into three main clusters, mostly according to the pepper fruit shape, size and potential pungency. Results suggest that the diversification of C. annuum in Spain may occur from a rather limited gene pool, still represented by few landraces with ancestral traits. This ancient population would suffer from local selection at the distinct geographical regions of Spain, giving way to pungent and elongated fruited peppers in the South and Center, while sweet blocky and triangular types in Northern Spain. 相似文献
