The effect of renal vasodilation and hypotonic volume expansion on water excretion in dogs after anesthesia and surgery

The possible effects of renal vasoconstriction from anesthesia and surgery on water excretion after hypotonic volume expansion (HVE) were studied in 18 well conditioned anesthetized dogs, with and without the infusion of phenoxybenzamine and acetylcholine into the renal artery of the cannulated kidney. In 6 dogs (Group 1 - Control) whose renal artery was infused with isosmotic saline, HVE resulted in a bilateral increase in GFR and UV (p < .05). ERPF, Cosm, C_H2O, U_NaV, U_KV, RBF, RVR and MAP did not change significantly. In 6 other dogs (Group 2), whose cannulated kidney was infused with phenoxybenzamine 50 μg/min before and during HVE, GFR increased on the infused side while CH₂O and UV increased bilaterally (p < .05). ERPF, Cosm, U_NaV, U_KV, RBF, RVR and MAP were not affected significantly. The addition of ADH, 2 mu/min into the phenoxybenzamine infusate, decreased ERPF, RBF and RVR bilaterally and CH₂O on the infused side (p < .05). It had no effect upon GFR, Cosm, U_NaV, U_KV and MAP. In another 6 dogs, (Group 3), whose cannulated renal artery was infused with acetylcholine (20 μg/min) before and during HVE, C_H2O, UV and RVR increased bilaterally (p < .05). ERPF and RBF decreased bilaterally (p < .05), whereas GFR, Cosm, U_NaV and MAP were unaffected. U_KV decreased on the infused side (p < .05). The addition of ADH (2 mu/min)_into the acetylcholine infusate, decreased C_H2O bilaterally and increased Cosm and U_KV on the control side (p < .05). It had no effect on ERPF, GFR, UV, U_NaV, RBF, RVR and MAP. These observations suggest that anesthesia and surgery produce renal vasoconstriction and this together with increased ADH release, interfere with water excretion by the kidney. Previous renal vasodilation prevents these influences of anesthesia and surgery.     

Cellular basis for tentacle adherence in the Portuguese man-of-war (Physalia physalis)

The fishing tentacles of Physalia physalis (Portuguese man-of-war) adhere to prey and human victims by the penetration of a barbed tubule connected to an intracellular nematocyst. The nematocyst is surrounded by a fibrillar system of microtubules and microfilaments that terminate in hemidesmosomal processes which anchor the nematocyst to the acellular mesoglea of the tentacle.     

Immunological and biochemical analysis of a null mutant of α-glycerolphosphate dehydrogenase from Drosophila melanogaster

Summary A Drosophila null mutant(BO-1-4) of -glycerolphosphate dehydrogenase induced by ethylmethane sulfonate(EMS) was analyzed by double immunodiffusion, enzyme immuno-inactivation, immunoelectrophoresis and two-dimensional electrophoresis. Based on all the immunological evidence, this mutant appears to express no protein that can cross-react with the antiserum specific to -glycerolphosphate dehydrogenase. A protein spot corresponding to -glycerolphosphate dehydrogenase was identified on two-dimensional gels of the soluble fly homogenates. The absence of this protein spot on two-dimensional gels of this null mutant further supported the immunological data. The activities of seven other enzymes in the related metabolic pathways were determined for the mutant and the control Drosophila. The null mutant does not show significant alterations in activities of these enzymes. Immunohistochemical localization of vitamin D-dependent calcium-binding protein in duodenum,kidney, uterus and cerebellum of chickens

Summary Calcium-binding protein (CaBP) has been localized with the immunoperoxidase method using antiserum against purified chick duodenal CaBP. Different preparative procedures were employed to investigate the experimental conditions possibly responsible for the contradictory reports in the literature of the precise cellular localization of CaBP. Freeze substitution, frozen sections followed by fixation and coagulant and non-coagulant fixatives were used with appropriate control sections to demonstrate that the true localization of CaBP in the chick duodenum is in the absorptive cell cytoplasm. The goblet cell localization reported in the literature seems to be a diffusion artifact due to inadequate fixation. CaBP was also localized in several other tissues. In the hen uterus, the tubular glands beneath the surface epithelium showed intense reaction. In the kidney, CaBP was present in the cells of the straight and convoluted segments of distal tubules. The cortex of the chick cerebellum showed the CaBP in Purkinje cells. The entire dendritic trees contained the reaction product. No other neurons in the molecular or the granular layer were stained. In the deep cerebellar nuclei, all neurons were negative and these were outlined by deeply staining axons of the Purkinje cells and their synaptic endings.     

Quercetin inhibits hexose transport in a human diploid fibroblast

Summary The flavonol quercetin, a phloretin analog, inhibits transport of 2-deoxyglucose and 3-O-methylglucose in a cultured human diploid fibroblast. This inhibition is related to transport itself and not to the reported effects of flavonoids on membrane-bound ATPases. From concentration-inhibition curves at several pH's we conclude that uncharged (acid) quercetin (pK=7.65) is the inhibitory form of the molecule (K _I =10m). Quercetin, unlike phloretin, is rapidly degraded in 0.1n NaOH; the degradation products are weakly inhibitory to hexose transport.     

SMITHSONIELLA GEN. NOV., A POSSIBLE EVOLUTIONARY LINK BETWEEN THE MULTICELLULAR AND SIPHONOUS HABITS IN THE ULVOPHYCEAE,CHLOROPHYTA

A low relief, green turf-forming alga of a heterotrichous habit was discovered in the coral reef microcosm, Museum of Natural History, Smithsonian Institution. Erect filaments bore lateral, specialized sporangia and together with basal filaments possessed septal plugs between adjacent cells, grossly similar to the “pit connections” of red algae. Data are presented which: 1) establish the identity of our plant with a plant recently described as Pilinia earleae Gallagher et Humm from the Florida Gulf coast; 2) support our establishment of the new genus Smithsoniella and our transfer of P. earleae to this new taxon. Additional data on pigmentation and cytology are related to the fine structure of other selected green algae to develop and test three hypotheses, viz. Smithsoniella earleae represents either: 1) a symbiotic association between a green and a red alga; 2) an alga which belongs to either the Ulotrichales, Chaetophorales or the Chroolepidales; or 3) an alga representing an evolutionary link between filamentous forms of the Ulvophyceae and members of the coenocytic siphonalean complex (e.g., Codiales or Caulerpales) of the Chlorophyta. Data refute hypotheses 1 and 2 but do lend support to the third hypothesis.     

Parallel usage of medicinal plants by africans and their caribbean descendants

Cultivated plants are cited by anthropologists as important indicators of man’s past. Medicinal species, to a large extent, have been overlooked even though in some cases these plants represent some of the social and cultural traditions of the people who use them. A number of cultivated plants have been traced from the Old World to the New World and are generally believed to have been carried there by European explorers and early settlers. However, some evidence has been accumulating to indicate that there may have been contacts other than by European colonists. One trade route that has been neglected is that of the slave trade from west Africa to the Caribbean. Three plant species,Citrus aurantifolia, Ricinus communis andAbrus precatorius, may exemplify the role and use of this route. They also indicate the migration and assimilation of west African Fulani, Hausa, and Mandingo cultures and Obeah religion into Caribbean society.     

The lysis of Gram-negative Alcaligenes eutrophus by enzymes from Cytophaga

Summary The use of Cytophaga lysing enzymes was investigated for the liberation of poly--hydroxybutyrate (PHB) granules from the Gram-negative bacterium Alcaligenes eutrophus. Complete cell lysis was approached within a 60 minute period. Contrary to previous findings for the lysis of Gram-negative bacteria, prior removal of the outer membrane was not essential for enzymic lysis. The destabilisation of the outer membrane by the removal of divalent cations resulted in no significant improvement in the disruption process.     

How do floater male eastern bluebirds benefit by filling vacancies on territories where females already have young?

When male eastern bluebirds (Sialia sialis) are killed or deserttheir mates, replacement males usually take over the territory.We captured 25 eastern bluebird males and held them in captivityto study the behavior of replacement males and their responseto young already present in the nest. Of 17 replacement males,most courted the females, and 23% of experimental females renested,a proportion not different from that of control females. Noreplacement males were infanticidal, and none fully adoptedthe young. Replacement males spent significantly less time onthe territory and defended it significantly less than controlmales. We discuss breeding options available to a floater maleeastern bluebird and conclude that (1) floater male bluebirdsthat fill territory vacancies benefit by gaining breeding opportunitiesin the current breeding season, and (2) indifference to theyoung is the least costly option for replacement male easternbluebirds.
