Haptoglobin (HP) is a serum protein that has the capability of binding the extracorpuscular haemoglobin released during haemolysis. It plays an important role in protection of haemolytic disease by reducing the oxidative and peroxidative potential at free haemoglobin. The present study was aimed to determine the prevalence of HP polymorphism among different Indian populations, anthropologically belonging to diverse ethnicity. The polymorphism was screened among 642 unrelated individuals belonging to 14 population groups of India including both tribal and non-tribal caste groups from different geographical regions of India with distinct linguistic affiliations. An attempt is also made to understand the distribution of HP polymorphism among the studied populations. The result reveals the HP gene to be polymorphic in all the studied populations. Except the two tribal populations (Thotis of Andhra Pradesh and Patelias of Rajasthan) and one caste population (Rajput of Himachal Pradesh), all the studied populations are found to obey the Hardy-Weinberg equilibrium. The significance of the present study is elucidated with the prevalence of high mutant HP*2 allele frequency in India. Selection could be one of the most plausible explanations for this high HP frequency because of its uniformly high occurrence among all the studied populations. 相似文献

Background
Human leishmaniasis is caused by more than 20 Leishmania species and has a wide range of symptoms. Our recent studies have demonstrated the essential role of sphingolipid degradation in the virulence of Leishmania (Leishmania) major, a species responsible for localized cutaneous leishmaniasis in the Old World. In this study, we investigated the function of sphingolipid degradation in Leishmania (Leishmania) amazonensis, an etiological agent of localized and diffuse cutaneous leishmaniasis in South America.Methodology/Principal Findings
First, we identified the enzyme LaISCL which is responsible for sphingolipid degradation in L. amazonensis. Primarily localized in the mitochondrion, LaISCL shows increased expression as promastigotes progress from replicative log phase to non-replicative stationary phase. To study its function, null mutants of LaISCL (Laiscl−) were generated by targeted gene deletion and complemented through episomal gene add-back. In culture, loss of LaISCL leads to hypersensitivity to acidic pH and poor survival in murine macrophages. In animals, Laiscl− mutants exhibit severely attenuated virulence towards C57BL6 mice but are fully infective towards BALB/c mice. This is drastically different from wild type L. amazonensis which cause severe pathology in both BALB/c and C57BL 6 mice.Conclusions/Significance
A single enzyme LaISCL is responsible for the turnover of sphingolipids in L. amazonensis. LaISCL exhibits similar expression profile and biochemical property as its ortholog in L. major. Deletion of LaISCL reduces the virulence of L. amazonensis and the outcome of Laiscl−-infection is highly dependent on the host''s genetic background. Therefore, compared to L. major, the role of sphingolipid degradation in virulence is substantially different in L. amazonensis. Future studies may reveal whether sphingolipid degradation is required for L. amazonensis to cause diffuse cutaneous infections in humans. 相似文献44.
Biological systems employing microorganisms have been used as an alternative to conventional chemical techniques for synthesizing gold nanoparticles. In the present study, gold nanoparticles have been synthesized from the supernatant broth (SB) and live cell filtrate (LCF) of the industrially important fungus Penicillium rugulosum. Additionally, potato dextrose broth (PDB) medium which is used for the growth of the fungus has also been able to synthesize gold nanoparticles. The size of the particles has been investigated by Bio-TEM before purification as well as after purification to find the difference in morphology pattern of the nanoparticles. Different characterization techniques like X-ray diffraction (XRD), infra-red (FTIR), X-ray photoelectron (XPS) and UV–vis spectroscopy have been used for analysis of the particles. SB of the fungus has yielded nanoparticles with better morphology and hence further optimization studies were conducted for controlling the size and shape of the above by altering pH and concentration of gold salt. A pH range of 4–6 has favored the synthesis process whereas increasing concentration of gold salt (beyond 2 mM) has resulted in the formation of bigger sized and aggregated nanoparticles. The optimized nanoparticles have been used to conjugate with isolated genomic DNA of bacteria Escherichia coli and Staphylococcus aureus. Visual observation of agarose gel electrophoresis images confirmed the binding of gold nanoparticles (4 μL and 6 μL) with isolated DNA (2 μL) fragments of both the organisms. The slight red shift of the surface plasmon (SP) band and minor aggregations noticed in Bio-TEM images for the DNA conjugated gold nanoparticles indicates that the genomic DNA could stabilize the particles against aggregation owing to negatively charged phosphate backbone. 相似文献

Obestatin is purported to be a peptide hormone encoded in preproghrelin. We studied the metabolic effects of continuous infusion of obestatin via subcutaneously implanted osmotic mini-pumps. Administration of up to 500nmol/kg body weight/day obestatin did not change 24h cumulative food intake or body weight in rats. Similarly, no effects were observed when obestatin was infused at 1000nmol/kg body weight/day for seven days. This dose of obestatin infused during a 24h fast did not alter weight loss, suggesting that obestatin has no effect on energy expenditure, and this dose did not alter glucose or insulin responses during an IPGTT. Obestatin was originally proposed to interact with GPR39 and subsequently the receptor for GLP-1. While both receptors are expressed in pancreatic islets, incubation with obestatin did not alter insulin release from islets in vitro. Moreover, obestatin did not bind to INS-1 beta-cells or HEK cells overexpressing GLP-1 receptors or displace GLP-1 binding to these cells. Although riverine carbon fluxes are a minor component of the global carbon cycle, thetransfer of organic carbon from land to ocean represents a flux of potential carbon storage, irre-versible over 10~3 to 10~4 a. Future carbon transfers through river basins are expected to accelerate,with respect to both sources and sinks, because of the large-scale human driven land-use and land-cover changes. Thus, the increased amounts of carbon transported to and sequestered inmarine sediments (through fertilization by river-borne inorganic nutrients) may be an important netsink for anthropogenic CO_2. Particularly, the humid tropics of South Asia are regions very sensitiveto this lateral C transport because of high precipitation and high rates of land use and cover change. In this paper we report on the role of upland tributaries in the transport processes influ-encing the lateral carbon and nitrogen fluxes of the Godavari, a large tropical river of India. By far,dissolved inorganic carbon (DIC) is the dominant form of carbon transport in the river basin. It con-stitutes as much as 75% to the total carbon load. Particulate and dissolved organic carbon (POC and DOC) fluxes account for 21% and 4%, respectively. In the upper basin, DOC fluxes exceedthat of POC due to large-scale anthropogenic activities. In contrast, tributaries in the central basinare characterized by comparable fluxes of POC and DOC. However, downriver POC export is 35%less than the import from upriver and tributaries due to the entrainment of sediments in river channels and dam sites. We argue that for highly disturbed watersheds in tropical regions, down-stream transport of sediments and carbon requires long-term sampling programmes. 相似文献

Covalent coupling of lipid to protein antigen, BSA, modifies the immunogenicity leading to selective induction of delayed type hypersensitivity in mice with no or very little concomitant antibody production. The mode of linkage of lipid to protein, however, controls its tissue distribution and retention in the body and cell uptake in vitro. Whereas D-BSA accumulates in the draining lymph node after foot pad inoculation, DA-BSA stays at the site. Further, DA-BSA is eliminated much more slowly and D-BSA more quickly from the body than the native antigen. Although both lipid-conjugates are taken up by lymphoid cells in vitro more than the native antigen, DA-BSA binds significantly more than D-BSA. On the basis of in vitro blastogenic response and enumeration of antigen sensitive cells and in vivo tests of delayed type hypersensitivity, DA-BSA appears superior to D-BSA but neither was as potent as BSA in CFA. 相似文献

Beta-lactam- and in particular carbapenem-resistant Enterobacteriaceae represent a major public health threat. Despite strong variation of resistance across geographical settings, there is limited understanding of the underlying drivers. To assess these drivers, we developed a transmission model of cephalosporin- and carbapenem-resistant Klebsiella pneumoniae. The model is parameterized using antibiotic consumption and demographic data from eleven European countries and fitted to the resistance rates for Klebsiella pneumoniae for these settings. The impact of potential drivers of resistance is then assessed in counterfactual analyses. Based on reported consumption data, the model could simultaneously fit the prevalence of extended-spectrum beta-lactamase-producing and carbapenem-resistant Klebsiella pneumoniae (ESBL and CRK) across eleven European countries over eleven years. The fit could explain the large between-country variability of resistance in terms of consumption patterns and fitted differences in hospital transmission rates. Based on this fit, a counterfactual analysis found that reducing nosocomial transmission and antibiotic consumption in the hospital had the strongest impact on ESBL and CRK prevalence. Antibiotic consumption in the community also affected ESBL prevalence but its relative impact was weaker than inpatient consumption. Finally, we used the model to estimate a moderate fitness cost of CRK and ESBL at the population level. This work highlights the disproportionate role of antibiotic consumption in the hospital and of nosocomial transmission for resistance in gram-negative bacteria at a European level. This indicates that infection control and antibiotic stewardship measures should play a major role in limiting resistance even at the national or regional level. 相似文献

A new class of antenna chromophores so called ‘tetrazolates’ have not been explored much for lanthanide luminescencent complexes. However, we have already published several articles considering pyridineoxide tetrazolates as sensitizer with lanthanide ions. Although this class of antenna attracted much less attention because of its poor photoluminescence quantum yields (tris‐pyridineoxide tetrazolate europium complex = 13% in solution) we tried and successfully achieved to improve the photoluminescence quantum yields for this particular antenna molecule by replacing coordinated water from the inner coordination sphere of europium ion by introducing phosphine oxides as additional chromophore. In the present article the two bis‐phosphine oxides attach two molecules of tris‐pyridineoxide tetrazolate europium(III) complex which leads to the improvement of the overall molar absorption coefficients as well as photo‐physical properties of the complexes. We found more than two‐fold increase (31% in solution) in photoluminescence quantum yield with one of the coordinated phosphine oxides comparing with that of tris‐pyridineoxide tetrazolate europium(III) complex. 相似文献
