A new method is reported for labeling proteins with the positron-emitting nuclide 18F. Initially, 4-[18F]-fluorobenzylamine was prepared in two steps from aqueous [18F]fluoride in high yield. The 18F acylation agent was formed by reaction of this product with disuccinimidyl suberate. Overall yields for the 4-[18F]fluorobenzylamine succinimidyl ester ([18F]SFBS), decay corrected to the end of cyclotron bombardment, were about 30% in a synthesis time of 60 min. After a 15-min reaction, 30-45% (decay corrected) of the [18F]SFBS could be coupled to intact antibodies and their F(ab')2 and Fab fragments. Coupling yields were dependent on protein concentration but not reaction time. HPLC purification of [18F]SFBS was necessary to obtain optimal coupling efficiency and immunoreactivity. The immunoreactivities of 18F-labeled F(ab')2 and Fab fragments of an antimyosin antibody were 89 +/- 5% and 75 +/- 9%, respectively. Biodistribution studies in normal mice demonstrated similar in vivo behavior of 18F-labeled antibody fragments and those labeled with 125I by using N-succinimidyl 3-[125I]iodobenzoate. These results indicate that this method may be useful for labeling monoclonal antibodies and other proteins and peptides with 18F.  相似文献

The empty sella turcica is defined as the herniation of the subarachnoid space within the sella with displacement of the pituitary towards the posteroinferior wall. By autopsy studies, the incidence in the general population is around 20%. The association of prolactinoma and empty sella has been coincidental & infrequently reported. As such for microadenoma, visual field testing and screening for hypopituitarism is not needed, but if it is associated with empty sella, both visual field testing and screening for hypopituitarism is necessary.  相似文献

Investigations were made to explore the potential of an epigeic earthworm Eisenia foetida to transform textile mill sludge spiked with poultry droppings in to value added product, i.e., vermicompost. The growth and reproduction of E. foetida was monitored in a range of different feed mixtures for 77 days in the laboratory under controlled experimental conditions. The maximum growth was recorded in 100% cow dung (CD). Replacement of poultry droppings by cow dung in feed mixtures and vice versa had little or no effect on worm growth rate and reproduction potential. Worms grew and reproduced favourably in 70% poultry droppings (PD)+30% solid textile mill sludge (STMS) and 60% PD+40% STMS feed mixtures. Greater percentage of STMS in the feed mixture significantly affected the biomass gain and cocoon production. Net weight gain by earthworms in 100% CD was 2.9-18.2 fold higher than different STMS containing feed mixtures. The mean number of cocoon production was between 23.4+/-4.65 (in 100% CD) and 3.6+/-1.04 (in 50% PD+50% STMS) cocoons earthworm(-1) for different feed mixtures tested. Vermicomposting resulted in significant reduction in C:N ratio and increase in nitrogen and phosphorus contents. Total potassium, total calcium and heavy metals (Fe, Zn, Pb and Cd) contents were lower in the final product than initial feed mixtures. Our trials demonstrated vermicomposting as an alternate technology for the recycling and environmentally safe disposal/management of textile mill sludge using an epigeic earthworm E. foetida if mixed with poultry droppings.  相似文献

To investigate the effect of inland groundwater salinity on growth performance, feed conversion efficiency, nutrient retention and intestinal enzyme activity in milkfish, two experiments were conducted. In the first experiment (Expt I), a 100‐day monoculture of Chanos chanos [mean body weight (BW): 2.2 g] at different salinities (0, 10, 15, 20 and 25‰) was carried out in ponds fertilized with cowdung (about 10 000 kg ha?1 year?1) and poultry droppings (about 3000 kg ha?1 year?1). The fish were fed a compounded supplementary diet (containing 40% protein) at 5% BW day?1. Studies have revealed that growth increased with each increase in the salinity level; the highest values in weight gain and energy assimilated were observed in ponds maintained at 25‰ salinity [weight: 322.2 g and specific growth rate (SGR): 8.3]. Highest values of condition factor (0.7) and exponential value (n) of the length–weight relationship (LWR; n = 3.25) were also observed in ponds maintained at 25‰ salinity. Dissolved oxygen (DO), biological oxygen demand (BOD), pH and nutrient release remained at the optimal level during the culture period. High values of chlorophyll a, net primary productivity (NPP), phytoplankton and zooplankton population coincided with the highest values of alkalinity and turbidity in ponds maintained at 25‰ salinity. Multivariate analysis revealed a significant positive correlation of chlorides (r = 0.91), conductivity (r = 0.89) and hardness (r = 0.96) with fish growth. Productivity indicating parameters viz. NPP (r = 0.45), nitrate (r = 0.94) and o‐PO4 (r = 0.52) also showed a significant positive correlation with fish weight gain. In the second experiment (Expt II), milkfish (mean BW: 3.7 g) fry were exposed to different levels of salinity (0.0, 10, 15, 20, 25 and 30‰), and maintained for 90 days in the laboratory. Significantly (P < 0.05) high growth (percentage increase in BW: 183.1 and SGR: 1.2), feed conversion efficiency (64.5%) and intestinal enzyme activity (protease 5.1, amylase 4.1 and cellulolytic 3.2) were observed in the group maintained at 25 ppt salinity in comparison with other groups similarly maintained at low or high salinity levels. Carcass composition, muscle and liver glycogen levels were also significantly (P < 0.05) affected by salinity changes. The significance of these findings is discussed in this paper.  相似文献

We had earlier shown that human foetal epithelial cells (WISH), growth-inhibited by interferon gamma (IFNgamma), were reversibly detained at a point prior to DNA synthesis. In the present study, we determined the window of action of IFNgamma in the G1 phase duration and the exact point of detention of WISH cells in cell cycle progression with respect to the known points of detention by the inhibitors of DNA replication initiation (aphidicolin and carbonyl diphosphonate) and of activation of replication protein A (6-dimethylaminopurine), of which RPA activation being the earlier event compared to DNA replication initiation in cell cycle progression. WISH cells, which were released from IFNgamma-induced arrest, permeabilised and exposed independently to these inhibitors show that IFNgamma detains WISH cells prior to initiation of DNA synthesis. Further, exposure of IFNalpha-synchronized (at G0/G1) or mimosine-synchronized (at G1/S) WISH cells to IFNgamma, which was added at different time points post-release from the synchronizing agent, showed that the cells were promptly responsive to the growth inhibitory action of IFNgamma only during the first 11h in G1 phase. Taken together, these results suggest that IFNgamma inhibits growth of WISH cells by detaining them at a point prior to initiation of DNA synthesis and that the IFN acts within the first 11h in G1 phase of the cell cycle.  相似文献

This study describes a method for predicting and classifying oxygen-binding pro- teins. Firstly, support vector machine (SVM) modules were developed using amino acid composition and dipeptide composition for predicting oxygen-binding pro- teins, and achieved maximum accuracy of 85.5% and 87.8%, respectively. Sec- ondly, an SVM module was developed based on amino acid composition, classify- ing the predicted oxygen-binding proteins into six classes with accuracy of 95.8%, 97.5%, 97.5%, 96.9%, 99.4%, and 96.0% for erythrocruorin, hemerythrin, hemo- cyanin, hemoglobin, leghemoglobin, and myoglobin proteins, respectively. Finally, an SVM module was developed using dipeptide composition for classifying the oxygen-binding proteins, and achieved maximum accuracy of 96.1%, 98.7%, 98.7%, 85.6%, 99.6%, and 93.3% for the above six classes, respectively. All modules were trained and tested by five-fold cross validation. Benzanthrone, an anthraquinone dye intermediate, is commonly used for the synthesis of a number of polycyclic vat and disperse dyes. Our prior studies have shown that benzanthrone can be metabolized by rat hepatic microsomal cytochrome P450 (P450) (Biochem. Int., 18, 1989, 1237). In this study, the interaction of benzanthrone with rat hepatic microsomal P-450 and its effect on xenobiotic metabolism have been investigated. Parenteral administration of benzanthrone (40 mg/kg body weight) for 3, 7, or 21 days caused no change in the relative body weight or organ weight of rats. The levels of P450 were found to be reduced (33%-50%) in all the benzanthrone-exposed animals at all the time periods. In vitro addition of benzanthrone caused a spectral change with oxidized P450 and concentration-dependent reduction in the carbon monoxide spectrum of dithionite-reduced P450. The addition of benzanthrone to hepatic microsomes prepared from phenobarbital-treated rats resulted in spectral changes characterized by an absorbance maximum at 397 nm indicative of type I binding. In vitro addition of benzanthrone showed a concentration-dependent inhibition of hepatic aminopyrine N-demethylase (APD) and ethoxyresorufin-O-deethylase (ERD) activities with respective I50 values of 9.5 x 10(-4) and 8.0 x 10(-5) M. However, the inhibition of aryl hydrocarbon hydroxylase (AHH) even at the highest concentration of benzanthrone (10(-2) M), was of the order of only 29%. In vivo administration of benzanthrone also led to the inhibition of APD, AHH, and ERD activities at all treatment times although the magnitude of inhibition was of a lower order.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Mitochondrial evolution entailed the origin of protein import machinery that allows nuclear-encoded proteins to be targeted to the organelle, as well as the origin of cleavable N-terminal targeting sequences (NTS) that allow efficient sorting and import of matrix proteins. In hydrogenosomes and mitosomes, reduced forms of mitochondria with reduced proteomes, NTS-independent targeting of matrix proteins is known. Here, we studied the cellular localization of two glycolytic enzymes in the anaerobic pathogen Trichomonas vaginalis: PPi-dependent phosphofructokinase (TvPPi-PFK), which is the main glycolytic PFK activity of the protist, and ATP-dependent PFK (TvATP-PFK), the function of which is less clear. TvPPi-PFK was detected predominantly in the cytosol, as expected, while all four TvATP-PFK paralogues were imported into T. vaginalis hydrogenosomes, although none of them possesses an NTS. The heterologous expression of TvATP-PFK in Saccharomyces cerevisiae revealed an intrinsic capability of the protein to be recognized and imported into yeast mitochondria, whereas yeast ATP-PFK resides in the cytosol. TvATP-PFK consists of only a catalytic domain, similarly to “short” bacterial enzymes, while ScATP-PFK includes an N-terminal extension, a catalytic domain, and a C-terminal regulatory domain. Expression of the catalytic domain of ScATP-PFK and short Escherichia coli ATP-PFK in T. vaginalis resulted in their partial delivery to hydrogenosomes. These results indicate that TvATP-PFK and the homologous ATP-PFKs possess internal structural targeting information that is recognized by the hydrogenosomal import machinery. From an evolutionary perspective, the predisposition of ancient ATP-PFK to be recognized and imported into hydrogenosomes might be a relict from the early phases of organelle evolution.  相似文献
