Mitochondrial electron transport chain complex dysfunction in a transgenic mouse model for amyotrophic lateral sclerosis 总被引:1,自引:0,他引:1
Amyotrophic lateral sclerosis is a fatal neurodegenerative disease that causes degeneration of motoneurons. Mutation of Cu,Zn superoxide dismutase (SOD1) is one cause for this disease. In mice, expression of mutant protein causes motoneuron degeneration and paralysis resembling the human disease. Morphological change, indicative of mitochondrial damage, occurs at early stages of the disease. To determine whether mitochondrial function changes during the course of disease progression, enzyme activities of mitochondrial electron transport chain in spinal cords from mice at different disease stages were measured using three different methods: spectrophotometric assay, in situ histochemical enzyme assay, and blue native gel electrophoresis combined with in-gel histochemical reaction. The enzyme activities were decreased in the spinal cord, particularly in the ventral horn, beginning at early disease stages. This decrease persisted throughout the course of disease progression. This decrease was not detected in the spinal cords of non-transgenic animals, of mice expressing the wild-type protein, and in cerebellum and dorsal horn of the spinal cords from mice expressing mutant protein. These results demonstrate a functional defect in mitochondria in the ventral horn region and support the view that mitochondrial damage plays a role in mutant SOD1-induced motoneuron degeneration pathway. 相似文献

Legumin subunits are synthesized as precursor polypeptides and are transported into protein storage vacuoles in field bean cotyledons. We expressed a legumin subunit in yeast and found that in these cells it is also transported into the vacuoles. To elucidate vacuolar targeting information, we constructed gene fusions of different legumin propolypeptide segments with either yeast invertase or chloramphenicol acetyltransferase as reporters for analysis in yeast or plant cells, respectively. In yeast, increasing the length of the amino-terminal segment increased the portion of invertase directed to the vacuole. Only the complete legumin alpha chain (281 amino acids) directed over 90% to the vacuole. A short carboxy-terminal legumin segment (76 amino acids) fused to the carboxy terminus of invertase also efficiently targeted this fusion product to yeast vacuoles. With amino-terminal legumin-chloramphenicol acetyltransferase fusions expressed in tobacco seeds, efficient vacuolar targeting was obtained only with the complete alpha chain. Objectives
To develop an RNA aptamer specific for the methyltransferase (MTase) of dengue virus (DENV) which is essential for viral genome replication and translation acting directly on N-7 and 2′-O-methylation of the type-I cap structure of the viral RNA.Results
We identified 2′-fluoro-modified RNA aptamers that can specifically bind DENV serotype 2 (DENV2) MTase using systematic evolution of ligands by exponential enrichment technology. We truncated the chosen aptamer into a 45-mer RNA sequence that can bind DENV2 MTase with K d ~ 28 nM and inhibit N-7 methylation activity of the protein. Moreover, the 45-mer truncated aptamer could not only bind with an K d ~ 15.6 nM but also inhibit methylation activity of DENV serotype 3 (DENV3) MTase. The 45-mer aptamer competitively impeded binding of both DENV2 and DENV3 genomic RNA to MTase of each serotype.Conclusion
The selected 45-mer truncated RNA aptamer specifically and avidly bound DENV MTase and competitively inhibited its methylation activity, and thus could be useful for the development of anti-DENV agents.995.

Antennal olfactory sensory neurones responsive to host and nonhost plant volatiles in gorse pod moth Cydia succedana 下载免费PDF全文
We investigate the response profiles of the antennal olfactory sensory neurones (OSNs) in male and female gorse pod moth Cydia succedana to host and nonhost volatiles, using the single sensillum recording technique. Eight different classes of olfactory sensilla are identified in female C. succedana and five different classes of olfactory sensilla in males. Nineteen different classes of OSNs are identified from the sensilla in females, and nine different classes of OSNs in the male sensilla. All classes of sensilla, except class F7 and class M1 sensilla, co‐compartmentalize two or three OSNs in each sensillum, and the OSNs present in the same sensillum are specialized for different volatiles. Most plant‐volatile OSNs exhibit phasic‐tonic type of temporal responses, whereas the pheromone OSNs in male C. succedana show rather phasic responses. The majority of OSNs identified in C. succedana display highly specialized responses to a narrow range of volatiles, whereas only a small proportion of OSNs show broad response spectra. Two most abundant classes of OSNs exhibit highly specialized responses to β‐myrcene and (E)‐β‐ocimene, two major volatiles released by gorse (Ulex europaeus), the main host of C. succedana. By contrast, several other classes of OSNs exhibit highly specialized responses to geraniol, (Z)‐3‐hexen‐1‐ol, (±)‐α‐terpineol, citral and benzyl acetate, which are produced by various nonhost plants. Taking the results of the present study together, we suggest that C. succedana use the combinational input from a set of highly specialized OSNs for host plant volatiles and another set of highly specialized OSNs for nonhost volatiles to discriminate between hosts and nonhosts. 相似文献

Mitochondria isolated from potato tubers fail to undergo passive osmotic swelling when suspended in isotonic Na+ acetate or phosphate, in NaCl following addition of tripropyltin, or in Na+ nitrate following addition of an uncoupler. Swelling under each of these conditions in mitochondria from other sources has been attributed to the inward movement of Na+ on an endogenous Na+/H+ exchanger. Such a monovalent cation/H+ exchanger has also been implicated in respiration-dependent cation extrusion and contraction of swollen mitochondria. Potato mitochondria swollen in chloride and nitrate salts extrude ions and contract when respiration is initiated. The contraction reaction is slower and less efficient than that in beef heart mitochondria, but like the latter, is sensitive to uncouplers and stimulated by nigericin, butacaine, and Mg2+. These comparative studies suggest that a cation+/H+ exchanger is present in potato tuber mitochondria, but that it functions exclusively as a cation-extruding mechanism. They further suggest that cation+/H+ exchange activity is not identical in mitochondria from different sources and that these exchange components may have a directionality and regulatory features which differ with the metabolic needs of the source tissue. 相似文献

Glucose oxidase (GOD) from Aspergillus niger was expressed in Saccharomyces cerevisiae under the regime of GAL-10 promoter and GAL-7 terminator of S. cerevisiae and -amylase signal sequence of Aspergillus oryzae. The enhancement of the expression level was achieved in pH-stat feed-back controlled fed-batch culture. Strain 16F3HT, a Gram-negative, aerobic, non-motile, and oval-shaped bacterium, was isolated from river water collected from the Han River in South Korea. Growth of strain 16F3HT was observed at 10–42 °C (optimum at 25–30 °C), but no growth occurred at 4 °C. The strain is able to grow at pH 4–10 (optimum at pH 7–8) and tolerates up to 4% NaCl (w/v), with optimum growth at 0.5% NaCl. The isolate was found to be resistant to UV irradiation. Based on 16S rRNA gene sequence analysis, it is closely related to ‘Deinococcus seoulensis’ 16F1E (98.8%), Deinococcus aquaticus PB314T (98.1%) and Deinococcus caeni Ho-08T (98.0%). The level of DNA–DNA homology between the novel strain and the three related strains was 57.4, 41.2, and 35.8%, respectively. Chemotaxonomic data revealed that strain 16F3HT possesses MK-8 as the predominant respiratory quinone, an unidentified phosphoglycolipid as the major polar lipid, and C15:1 ω6c and C16:1 ω7c as the major fatty acids. The DNA G + C content was determined to be 65.7 mol%. Based on polyphasic evidence, strain 16F3HT (=KCTC 33794T = JCM 31406T) should be classified as the type strain of a novel Deinococcus species, for which the name Deinococcus knuensis sp. nov. is proposed. 相似文献

An increasing concern on resistance to multiple-antibiotics has led to the discovery of novel agents and the establishment of new precaution strategy. Numerous plant sources have been widely studied to reduce virulence of pathogenic bacteria by interfering cell-to-cell based communication called quorum sensing (QS). Leaf extracts of 17 gardening trees were collected and investigated for their anti-QS effects using a sensor strain Chromobacterium violaceum CV026. Methanolic extracts of K4 (Acer palmatum), K9 (Acer pseudosieboldianum) and K13 (Cercis chinensis) leaves were selected for further experiments based on their antagonism effect on QS without inhibiting C. violaceum CV026 growth. Subsequently, the leaf extracts on QS-mediated virulence of Pseudomonas aeruginosa PAO1 involved in biofilm formation, motility, bioluminescence, pyocyanin production, QS molecules production, and Caenorhabditis elegans killing activity were evaluated. The biofilm formation ability and swarming motility of P. aeruginosa PAO1 were decreased approximately 50% in the presence of these leaf extracts at a concentration of 1 mg/mL. The expression level of lecA::lux of P. aeruginosa PAO1 and pyocyanin production were also reduced. The three leaf extracts also decreased autoinducer (AI) production in P. aeruginosa PAO1 without direct degradation, suggesting that AI synthesis might have been suppressed by these extracts. The three leaf extracts also showed anti-infection activity in C. elegans model. Taken together, these results suggest that methanolic leaf extracts of K4, K9 and K13 have the potential to attenuate the virulence of P. aeruginosa PAO1. 相似文献
