Regenerating human tooth ex vivo and biological repair of dental caries are hampered by non-viable odontogenic stem cells that can regenerate different tooth components. Odontoma is a developmental dental anomaly that may contain putative post-natal stem cells with the ability to differentiate and regenerate in vivo new dental structures that may include enamel, dentin, cementum and pulp tissues. We evaluated odontoma tissues from 14 patients and further isolated and characterized human odontoma-derived mesenchymal cells (HODCs) with neural stem cell and hard tissue regenerative properties from a group of complex odontoma tissues from 1 of 14 patients. Complex odontoma was more common (9 of 14) than compound type and females (9 of 14) were more affected than males in our set of patients. HODCs were highly proliferative like dental pulp stem cells (DPSCs) but demonstrated stronger neural immunophenotype than both DPSCs and mandible bone marrow stromal cells (BMSCs) by expressing higher levels of nestin, Sox 2 and βIII-tubulin. When transplanted with hydroxyapatite/tricalcium phosphate into immunocompromised mice, HODCs differentiated and regenerated calcified hard tissues in vivo that were morphologically and quantitatively comparable to those generated by DPSCs and BMSCs. When transplanted with polycaprolactone (biodegradable carrier), HODCs differentiated to form new predentin on the surface of a dentin platform. Newly formed predentin contained numerous distinct dentinal tubules and an apparent dentin–pulp arrangement. HODCs represent unique odontogenic progenitors that readily commit to formation of dental hard tissues.  相似文献

The present study examines an antiserum prepared against antigen-reactive T cells that induces murine H-Y-specific delayed-type hypersensitivity (DTH) responses. This anti-H-Y receptor antibody (ARA) was raised in C57BL/6 male mice against splenic T lymphocytes from H-Y immune syngeneic females. Subcutaneous administration of ARA to cyclophosphamide-pretreated C57BL/6 females is able to induce H-Y-specific delayed-type footpad swelling responses. The DTH inducing capacity in ARA was selectively retained on rabbit anti-mouse immunoglobulin columns and was absorbed completely by H-Y immune lymphoid cells from C57BL/6 females. The induction of H-Y DTH reactivity was due at least in part to the activation of H-Y antigen-specific T lymphocytes that could adoptively transfer DTH-like responses to naive female mice. ARA induces DTH responses in strains with the same lgh regions, including selected strains of H-Y nonresponders. Therefore, MHC-linked lr genes do not appear to be as critical when responses are triggered by ARA instead of by antigen. Possible mechanisms for the induction of immune responses by ARA are discussed.  相似文献

The effect of temperature on germination, radial growth and pathogenic activity of two strains of Beauveria bassiana and four strains of Metarhizium anisopliae on the legume flower thrips, Megalurothrips sjostedti , was studied in the laboratory. Germination, radial growth and pathogenic activity was low for all strains at 15 C. Optimum temperature for germination, radial growth and pathogenic activity ranged between 25-30 C. The fastest growing strain at 25-30 C was M. anisopliae strain ICIPE 69. Compared to other strains, ICIPE 69 also appeared to have a broad temperature range of pathogenic activity against the pest. The significance of these findings in relation to control of M. sjostedti is discussed.  相似文献

Fopius arisanus (Sonan), a solitary koinobiont endoparasitoid of fruit flies, was introduced for testing and final release against the recently discovered species Bactrocera invadens Drew, Tsuruta and White in Africa. Laboratory experiments were conducted to assess host preference, host acceptability for oviposition, and physiological suitability of B. invadens and five other indigenous tephritid fruit fly species, namely, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), C. cosyra (Walker), C. rosa Karsch, C. fasciventris (Bezzi), and C. anonae Graham. Females of F. arisanus visited all host egg patches, but showed a stronger preference to eggs of B. invadens, which was also most accepted for oviposition. Successful development of parasitoid progenies varied greatly across hosts, with B. invadens yielding the highest parasitoid progeny and C. fasciventris yielding no F. arisanus progeny. Most of the parasitoid eggs laid in C. rosa and C. fasciventris were encapsulated. Sex ratio was not influenced by host species and it was female biased in all hosts that produced parasitoid progeny. Fopius arisanus was able to establish a new association with C. capitata, C. cosyra and to a lesser extent C. anonae. The results are discussed in the light of the potential use of F. arisanus as a biological control agent of B. invadens.  相似文献

  1. False codling moth, Thaumatotibia leucotreta is among the key constraints of the agricultural industry. Little is known about the population dynamics and genetic diversity of this pest in East Africa.
  2. The spatial–temporal population dynamics and genetic diversity of false codling moth were evaluated in citrus orchards in Kenya and Tanzania between May 2017 and August 2018 covering two fruiting seasons. The diversity of false codling moth sampled in these orchards and from solanaceous hosts, as well as from specimens received from Uganda, Sudan, and South Africa were assayed using the mitochondrial cytochrome oxidase I gene.
  3. A similar spatial–temporal pattern of false codling moth was found in both Kenya and Tanzania, with the most male moths in August 2017 and 2018. In Tanzania, the number of male moths caught at high and mid altitudes did not differ but were significantly higher than those caught at low altitude.
  4. A relatively low false codling moth genetic diversity was recorded at the sites where sampling was done in the respective countries as well as between specimens sampled from different host plants.
  5. The low false codling moth genetic diversity determined in this trial can be exploited in the management of the pest in the studied countries.

Three Pseudomonas species isolated from oil polluted soils in Lagos, Nigeria were studied for their pyrene degradation potentials. These isolates exhibited broad substrate specificities for hydrocarbon substrates including polycyclic aromatic hydrocarbons, petroleum fractions and chlorobenzoates. All three isolates tolerated salt concentrations of more than 3%. They resisted ampicillin, cenfuroxime, but susceptible to ofloxacin and ciprofloxacin. Pseudomonas sp. strain LP1 exhibited growth rates and pyrene degradation rates of 0.018 h−1 and 0.111 mg l−1 h−1 respectively, while P. aeruginosa strains LP5 and LP6 had corresponding values of 0.024, 0.082 and 0.017, 0.067 respectively. The overall respective percentage removal of pyrene obtained for strains LP1, LP5 and LP6 after a 30-day incubation period were 67.79, 66.61 and 47.09. Resting cell assay revealed that strain LP1 had the highest uptake rate. Strains LP1, LP5, and LP6 also used the ortho-cleavage pathway. Enzyme study confirmed activity of catechol 1,2-dioxygenase in all with values 0.6823, 0.9199, and 0.8344 μmol min−1 mg−1 respectively for LP1, LP3 and LP6. To the best of our knowledge, ours is the first report of pyrene-degraders from the sub-Saharan African environment.  相似文献
