The activity of three enzymes, aspartokinase, homoserine dehydrogenase, and homoserine kinase, has been studied in the industrial strainSaccharomyces cerevisiae IFI256 and in the mutants derived from it that are able to overproduce methionine and/or threonine. Most of the mutants showed alteration of the kinetic properties of the enzymes aspartokinase, which was less inhibited by threonine and increased its affinity for aspartate, and homoserine dehydrogenase and homoserine kinase, which both lost affinity for homoserine. Furthermore, they showed in vitro specific activities for aspartokinase and homoserine kinase that were higher than those of the wild type, resulting in accumulation of aspartate, homoserine, threonine, and/or methionine/S-adenosyl-methionine (Ado-Met). Together with an increase in the specific activity of both aspartokinase and homoserine kinase, there was a considerable and parallel increase in methionine and threonine concentration in the mutants. Those which produced the maximal concentration of these amino acids underwent minimal aspartokinase inhibition by threonine. This supports previous data that identify aspartokinase as the main agent in the regulation of the biosynthetic pathway of these amino acids. The homoserine kinase in the mutants showed inhibition by methionine together with a lack or a reduction of the inhibition by threonine that the wild type undergoes, which finding suggests an important role for this enzyme in methionine and threonine regulation. Finally, homoserine dehydrogenase displayed very similar specific activity in the mutants and the wild type in spite of the changes observed in amino acid concentrations; this points to a minor role for this enzyme in amino acid regulation.  相似文献

Abstract

End of life waste Lead (Pb) acid batteries are one of the largest sources of secondary lead production globally. Recycling lead by melting down used batteries is a commercial trade all over the world; but, regrettably, reprocessing lead from end of life batteries is reported for anthropogenic lead exposures causing harsh human health consequence and environmental pollution. The current research intends to isolate and identify Lead (Pb) solubilizing bacteria from automobile waste deposits from Agartala city in Tripura state of India. Scanning Electron Microscope equipped with energy-dispersive X-ray characterization of the grounded lead sample was carried out, and the micrographs demonstrated scattered structures across the matrix. The X-ray diffraction (XRD) spectrum indicates the presence of Lead Oxide (PbO), Lead dioxide (PbO2), and Lead sulfate (PbSO4) in the collected samples. A single bacterium viewing observable growth on Pb supplemented plates was isolated and its Pb recovering capability was estimated through ICP AES analysis. Molecular characterization of the bacterium was investigated using 16S rRNA sequencing along with isolated culture was taxonomically grouped as Cupriavidus sp. The genomic DNA sequences were submitted in NCBI GenBank with the accession number MG171197. In the present case of inspection, the ability of the bacterial strain to recover Pb from end life battery waste was carried out in laboratory scale on a shake flask for 20?days. The experiment conducted under optimum bioleaching parameters with initial pH 6, 5% w/v of microbial culture, 2% pulp density and 2?g/100?mL dextrose concentration at 30?°C temperature with a speed of 200?RPM resulted in 67% Pb recovery from the battery sample. This investigation emphasizes the significance of Pb recycling ability of native bacterial isolate for efficient Pb bio-recovery from end of life waste batteries.  相似文献

Dps (DNA-binding protein from starved cells) are dodecameric assemblies belonging to the ferritin family that can bind DNA, carry out ferroxidation, and store iron in their shells. The ferritin-like trimeric pore harbors the channel for the entry and exit of iron. By representing the structure of Dps as a network we have identified a charge-driven interface formed by a histidine aspartate cluster at the pore interface unique to Mycobacterium smegmatis Dps protein, MsDps2. Site-directed mutagenesis was employed to generate mutants to disrupt the charged interactions. Kinetics of iron uptake/release of the wild type and mutants were compared. Crystal structures were solved at a resolution of 1.8–2.2 Å for the various mutants to compare structural alterations vis à vis the wild type protein. The substitutions at the pore interface resulted in alterations in the side chain conformations leading to an overall weakening of the interface network, especially in cases of substitutions that alter the charge at the pore interface. Contrary to earlier findings where conserved aspartate residues were found crucial for iron release, we propose here that in the case of MsDps2, it is the interplay of negative-positive potentials at the pore that enables proper functioning of the protein. In similar studies in ferritins, negative and positive patches near the iron exit pore were found to be important in iron uptake/release kinetics. The unique ionic cluster in MsDps2 makes it a suitable candidate to act as nano-delivery vehicle, as these gated pores can be manipulated to exhibit conformations allowing for slow or fast rates of iron release.  相似文献

This study investigated the effectiveness of mangrove planting initiatives in Sri Lanka. All the lagoons and estuaries in Sri Lanka were included in the study. We documented all agencies and locations, involved in mangrove planting efforts, along with the major drivers of these planting initiatives, their extents, and the possible causes of the success or failure of planting. An adapted three‐step framework and a field survey consisting of vegetation and soil surveys and questionnaires were used to evaluate the objectives. We found that about 1,000–1,200 ha of mangroves, representing 23 project sites with 67 planting efforts, have been under restoration with the participation of several governmental and nongovernmental organizations. However, about 200–220 ha showed successful mangrove restoration. Nine out of 23 project sites (i.e. 36/67 planting efforts) showed no surviving plants. The level of survival of the restoration project sites ranged from 0 to 78% and only three sites, that is, Kalpitiya, Pambala, and Negombo, showed a level of survival higher than 50%. Survival rates were significantly correlated with post‐care. Planting mangrove seedlings at the incorrect topography often entails inappropriate soil conditions for mangroves. Survival rates showed significant correlations with a range of soil parameters except soil pH. Disturbance and stress caused by cattle trampling, browsing, algal accumulation, and insect attacks, factors that may themselves relate to choosing sites with inappropriate topography and hydrology, were common to most sites. The findings are a stark illustration of the frequent mismatch between the purported aims of restoration initiatives and the realities on the ground.  相似文献

For an animal to survive in a constantly changing environment, its behavior must be shaped by the complex milieu of sensory stimuli it detects, its previous experience, and its internal state. Although taste behaviors in the fly are relatively simple, with sugars eliciting acceptance behavior and bitter compounds avoidance, these behaviors are also plastic and are modified by intrinsic and extrinsic cues, such as hunger and sensory stimuli. Here, we show that dopamine modulates a simple taste behavior, proboscis extension to sucrose. Conditional silencing of dopaminergic neurons reduces proboscis extension probability, and increased activation of dopaminergic neurons increases extension to sucrose, but not to bitter compounds or water. One dopaminergic neuron with extensive branching in the primary taste relay, the subesophageal ganglion, triggers proboscis extension, and its activity is altered by satiety state. These studies demonstrate the marked specificity of dopamine signaling and provide a foundation to examine neural mechanisms of feeding modulation in the fly.  相似文献

It is known that the fractional absorption of extrinsic iron from human milk is higher in infants and adults. A low molecular weight milk fraction has been proposed to increase the bioavailability of iron from human milk. Nevertheless, the mechanisms remained elusive. Here in we demonstrate ferric reductase activity (Km 7.73 × 10−6 M) in low molecular weight human milk fraction (10kF, filtrate derived from ultra filtration of milk whey through 10 kDa cutoff membrane), which increased ferric iron solubility and iron uptake in Caco-2 cells. The 10kF fraction was as effective as ascorbic acid (1:20 iron to ascorbic acid) in increasing the ferric iron solubility and uptake in Caco-2 cells. Further, gel filtration chromatography on peptide column led to co-elution of ferric reductase and iron solubilization activities at an apparent molecular mass of <1500 Da. Interestingly, only these fractions containing ferric reductase activity also stimulated the uptake of iron in Caco-2 cells. Thus, it is concluded that human milk possesses ferric reductase activity and is associated with ferric iron solubilization and enhanced absorption.  相似文献

Osteopontin (OPN) is a pleiotropic protein implicated in various inflammatory responses including ischemia-reperfusion (I-R) injury. Two distinct forms of the protein have been identified: an extensively studied secreted form (sOPN) and a less-well-known intracellular form (iOPN). Studies have shown that increased OPN expression parallels the time course of macrophage infiltration into injured tissue, a late event in the development of cerebral infarcts. sOPN has been suggested to promote remodeling of the extracellular matrix in the brain; the function of iOPN may be to facilitate certain signal transduction processes. Here, we studied OPN expression in adult male Sprague-Dawley rats subjected to global forebrain I-R injury. We found iOPN in the cytoplasm of both cortices and the hippocampus, but unexpectedly only the right cortex exhibited a marked increase in the iOPN level after 45 min of reperfusion. Acetaminophen, a drug recently shown to decrease apoptotic incidence, caspase-9 activation, and mitochondrial dysfunction during global I-R, significantly inhibited the increase in iOPN protein in the right cortex, suggesting a role for iOPN in the response to I-R injury in the right cortex.  相似文献
