High specific activity [3H]indole-3-acetic acid (IAA) was applied directly to root nodules of intact pea plants. After 24 h, radioactivity was detected in all plant tissues. In nodule and root tissue, only 2–3% of3H remained as IAA, and analysis by thin layer chromatography suggested that indole-3-acetyl-L-aspartic acid (IAAsp) was a major metabolite. The occurrence of IAAsp in pea root and nodule tissue was confirmed unequivocally by gas chromatography-mass spectrometry (GC-MS). The following endogenous indole compounds were also unequivocally identified in pea root nodules by GC-MS: IAA, indole-3-pyruvic acid, indole-3-lactic acid, indole-3-propionic acid, indole-3-butyric acid, and indole-3-carboxylic acid. Evidence of the occurrence of indole-3-methanol was also obtained. With the exception of IAA and indole-3-propionic acid, these compounds have not previously been unequivocally identified in a higher plant tissue.  相似文献

Indole-3-acetic acid (IAA) has been unequivocally identified in culture supernatants of Rhizobium strains by gas chromatography-mass spectrometry. A method for accurately quantitating IAA in bacterial culture supernatants, employing deuterium-labeled IAA as an internal standard, has been developed. Similar IAA concentrations were found in culture supernatants of chosen Rhizobium mutants (defective in nodule formation) and their corresponding parent strains. Since some of the mutants are known to adhere to root hairs, it can be concluded that root hair curling is not simply a consequence of IAA production by rhizobia.  相似文献

Summary Cytokinins are deactivated in radish cotyledons by conversion to 7- and 9-glucosides. In a search for inhibitors of this metabolism, the following compounds were found to be effective: (a) 6-benzylamino-2-(2-hydroxyethylamino)-9-methylpurine; (b) 3-isobutyl-1-methylxanthine; (c) papaverine; (d) theophylline; (e) caffeine; and (f) theobromine. The order of effectiveness was: (a)>(b)=(c)>(d)=(e)=(f). While the methylxanthines (b) and (d) inhibited formation of both 7- and 9-glucosides of 6-benzylaminopurine (BAP) approximately equally, compounds (a) and (c) preferentially inhibited formation of BAP 9-glucoside. Inhibition of BAP glucoside formation by (a) at 1.3 mM elevated the level of free BAP and BAP nucleotide 23- and 94-fold, respectively. While abscisic acid (ABA) suppressed conversion of zeatin riboside to zeatin 7-glucoside in radish cotyledons, it did not inhibit conversion of BAP to glucosides. Hence, ABA probably does not inhibit the glucosylating enzymes directly but rather reduces the availability of free zeatin when zeatin riboside is supplied. Auxins and nutrient supply did not affect conversion of zeatin riboside to zeatin 7-glucoside. Relative to cotyledons developed in light, those developed in darkness had a reduced capacity to convert zeatin riboside to zeatin 7-glucoside. The results presented have identified types of chemical structures which could be developed to provide more effective inhbitors of cytokininN-glucosylation.For part II in the series Inhibitors of cytokinin metabolism see Zhang et al. A lipid analysis of the tissues of a cold-seep mytilid mussel collected from the Louisiana slope of the Gulf of Mexico was used in conjunction with a compound-specific isotope analysis to demonstrate the presence of methanotrophic symbionts in the mussel gill tissue and to demonstrate the host's dependence on bacterially synthesized metabolic intermediates. The gill tissue contained large amounts of group-specific methanotrophic biomarkers, bacteriohopanoids, 4-methylsterols, lipopolysaccharide-associated hydroxy fatty acids, and type I-specific 16:1 fatty acid isomers with bond positions at delta 8, delta 10, and delta 11. Only small amounts of these compounds were detected in the mantle or other tissues of the host animal. A variety of cholesterol and 4-methylsterol isomers were identified as both free and steryl esters, and the sterol double bond positions suggested that the major bacterially derived gill sterol [11.0% 4 alpha-methyl-cholesta-8(14),24-dien-3 beta-ol] was converted to host cholesterol (64.2% of the gill sterol was cholest-5-en-3 beta-ol). The stable carbon isotope values for gill and mantle preparations were, respectively, -59.0 and -60.4% for total tissue, -60.6 and -62.4% for total lipids, -60.2 and-63.9% for phospholipid fatty acids, and -71.8 and 73.8% for sterols. These stable carbon isotope values revealed that the relative fractionation pattern was similar to the patterns obtained in pure culture experiments with methanotrophic bacteria (R.E. Summons, L.L. Jahnke, and Z. Roksandic, Geochim. Cosmochim. Acta 58: 2853-2863, 1994) further supporting the conversion of the bacteria methylsterol pool.  相似文献

When zeatin was supplied to excised leaves of Populus alba, the principal metabolites formed were adenosine, O-β-d-glucopyranosyl-cis-zeatin (derived from cis-zeatin in the commercial zeatin used), O-β-d-glucopyranosylzeatin, and two new metabolites, namely, O-β-d-glucopyranosyldihydrozeatin and O-β-d-glycopyranosyl-9-β-d-ribofuranosyldihydrozeatin, the structures of which were confirmed by unambiguous synthesis. Chromatographic studies indicated that adenosine 5′-phosphate, zeatin 7-glucopyranoside, zeatin 9-glucopyranoside, dihydrozeatin and zeatin 9-riboside were minor metabolites. The principal metabolites of zeatin 9-riboside in P. nigra leaves were the new metabolites O-β-d-glucopyranosyl-9-β-d-ribofuranosylzeatin (synthesized chemically) and O-β-d-glucopyranosl-9-β-d-ribofuranosyldihydrozeatin.  相似文献

The endogenous cytokinins present in dryZea mays seed were determined using both radioimmunoassay and gas chromatography—mass spectrometry. Similar values for bases and ribosides were obtained by the two methods. The cytokinins present in embryo and endosperm were estimated separately using radioimmunoassay; similar levels of cytokinins were found in these two tissues. The major cytokinins detected on a whole-seed basis were dihydrozeatin riboside, O-glucosyldihydrozeatin riboside, zeatin 9-glucoside, zeatin, and the nucleotides of zeatin, dihydrozeatin, and isopentenyladenine. Cytokinin levels in the mature dry seed were considerably lower than cytokinin levels published in the literature for immature seed. Unexpected activity in the radioimmunoassays was detected in the wash from the DEAE cellulose column chromatography step. The compound(s) responsible for this activity did not have the solvent partitioning characteristics of a cytokinin base or riboside. They eluted as a single fraction following high-performance liquid chromatography on a Zorbax C8 column; this fraction showed no activity in theAmaranthus bioassay for cytokinins, but inhibited the activity of authentic zeatin riboside present at an optimal concentration.  相似文献

An approach to coordinated, spatially resolved, in situ carbon isotope analysis of organic matter and carbonate minerals, and sulfur three‐ and four‐isotope analysis of pyrite with an unprecedented combination of spatial resolution, precision, and accuracy is described. Organic matter and pyrite from eleven rock samples of Neoarchean drill core express nearly the entire range of δ13C, δ34S, Δ33S, and Δ36S known from the geologic record, commonly in correlation with morphology, mineralogy, and elemental composition. A new analytical approach (including a set of organic calibration standards) to account for a strong correlation between H/C and instrumental bias in SIMS δ13C measurement of organic matter is identified. Small (2–3 μm) organic domains in carbonate matrices are analyzed with sub‐permil accuracy and precision. Separate 20‐ to 50‐μm domains of kerogen in a single ~0.5 cm3 sample of the ~2.7 Ga Tumbiana Formation have δ13C = ?52.3 ± 0.1‰ and ?34.4 ± 0.1‰, likely preserving distinct signatures of methanotrophy and photoautotrophy. Pyrobitumen in the ~2.6 Ga Jeerinah Formation and the ~2.5 Ga Mount McRae Shale is systematically 13C‐enriched relative to co‐occurring kerogen, and associations with uraniferous mineral grains suggest radiolytic alteration. A large range in sulfur isotopic compositions (including higher Δ33S and more extreme spatial gradients in Δ33S and Δ36S than any previously reported) are observed in correlation with morphology and associated mineralogy. Changing systematics of δ34S, Δ33S, and Δ36S, previously investigated at the millimeter to centimeter scale using bulk analysis, are shown to occur at the micrometer scale of individual pyrite grains. These results support the emerging view that the dampened signature of mass‐independent sulfur isotope fractionation (S‐MIF) associated with the Mesoarchean continued into the early Neoarchean, and that the connections between methane and sulfur metabolism affected the production and preservation of S‐MIF during the first half of the planet's history.  相似文献

Glucosylation of adenine and 6-methylaminopurine was not detected in derooted 10-day-old radish seedlings. However, 4-(purin-6-ylamino)butanoic amide and 6-(3,4-dimethoxybenzylamino)purine (N6-substituted adenines with negligible cytokinin activity), like the highly active cytokinin 6-benzylaminopurine, were converted to 7-glucopyranosides. The N2-substituted guanine, 2-benzylaminopurin-6-one, and 6-benzylamino-2-(2-hydroxy-ethylamino)purine were also metabolized to glucosides which were probably 7-glucopyranosides. Hence glucosylation of purines is not restricted to N6-substituted adenines with strong cytokinin activity. Although only ca 1.6% of 6-benzylamino-9-(4-chlorobutyl)purine taken up by the derooted seedlings could be accounted for as 7- and 9-glucosides, a considerable proportion was metabolized to these glucosides in cotyledons excised from 2-day-old radish seedlings. The high cytokinin activity of this 9-substituted compound may be a consequence of cleavage of the 4-chlorobutyl groud at N-9.  相似文献

Demosponges are a rich natural source of unusual lipids, some of which are of interest as geochemical biomarkers. Although demosponges are animals, they often host dense communities of microbial symbionts, and it is therefore unclear which lipids can be synthesized by the animal de novo, and which require input from the microbial community. To address this uncertainty, we analyzed the lipids of Amphimdeon queenslandica, the only demosponge with a published genome. We correlated the genetic and lipid repertoires of A. queenslandica to identify which biomarkers could potentially be synthesized and/or modified by the sponge. The fatty acid profile of A. queenslandica is dominated by an unusual Δ5,9 fatty acid (cis‐5,9‐hexacosadienoic acid)—similar to what has been found in other members of the Amphimdeon genus—while the sterol profile is dominated by C27‐C29 derivatives of cholesterol. Based on our analysis of the A. queenslandica genome, we predict that this sponge can synthesize sterols de novo, but it lacks critical genes necessary to synthesize basic saturated and unsaturated fatty acids. However, it does appear to have the genes necessary to modify simpler products into a more complex “algal‐like” assemblage of unsaturated fatty acids. Ultimately, our results provide additional support for the poriferan affinity of 24‐isopropylcholestanes in Neoproterozoic‐age rocks (the “sponge biomarker” hypothesis) and suggest that some algal proxies in the geochemical record could also have animal contributions.  相似文献
