Using a water maze, it has been shown that both wild-type and apoE4-expressing female mice are at greater risk of developing age-related hippocampal-dependent impairments in spatial learning and memory than age-matched male mice of the same genotype. In addition, apoE4-expressing female mice were more sensitive to 137Cs gamma-radiation-induced impairment in spatial learning and memory than age-matched male mice of the same genotype. These findings imply that androgen receptors (ARs) contribute to spatial learning and memory, posing the question as to whether transgenic expression of AR in female mice might modulate hippocampal-dependent learning and memory under baseline conditions and after local brain irradiation. Hippocampal-dependent novel location recognition was comparable in wild-type and AR-Tg female mice. This function was impaired after irradiation in AR-Tg but not wild-type mice. In contrast, sham-irradiated wild-type and AR-Tg female mice showed hippocampal-independent novel location recognition, and this was not affected by radiation. After the second day of hidden platform training, in a water maze probe trial, sham-irradiated and irradiated AR-Tg female mice showed spatial memory retention but irradiated wild-type mice did not. After the third day of hidden platform training, only irradiated wild-type female mice did not show spatial memory retention in the water maze probe trial. Both sham-irradiated and irradiated wild-type and AR-Tg female mice showed passive avoidance learning and memory. These data support an important role for AR in spatial memory retention in water maze probe trials in female mice under baseline conditions and after cranial irradiation.  相似文献

The effect of single amino acid substitutions associated with the Italian (E22K), Arctic (E22G), Dutch (E22Q) and Iowa (D23N) familial forms of Alzheimer's disease and cerebral amyloid angiopathy on the structure of the 21-30 fragment of the Alzheimer amyloid β-protein (Aβ) is investigated by replica-exchange molecular dynamics simulations. The 21-30 segment has been shown in our earlier work to adopt a bend structure in solution that may serve as the folding nucleation site for Aβ. Our simulations reveal that the 24-28 bend motif is retained in all E22 mutants, suggesting that mutations involving residue E22 may not affect the structure of the folding nucleation site of Aβ. Enhanced aggregation in Aβ with familial Alzheimer's disease substitutions may result from the depletion of the E22-K28 salt bridge, which destabilizes the bend structure. Alternately, the E22 mutations may affect longer-range interactions outside the 21-30 segment that can impact the aggregation of Aβ. Substituting at residue D23, on the other hand, leads to the formation of a turn rather than a bend motif, implying that in contrast to E22 mutants, the D23N mutant may affect monomer Aβ folding and subsequent aggregation. Our simulations suggest that the mechanisms by which E22 and D23 mutations affect the folding and aggregation of Aβ are fundamentally different.  相似文献

Background  

We advocate unifying classical and genomic classification of bacteriophages by integration of proteomic data and physicochemical parameters. Our previous application of this approach to the entirely sequenced members of the Podoviridae fully supported the current phage classification of the International Committee on Taxonomy of Viruses (ICTV). It appears that horizontal gene transfer generally does not totally obliterate evolutionary relationships between phages.  相似文献

Thirteen species of fleshy fungi were identified from two agricultural fields during the 1989 and 1990 growing seasons at the Rural Development Center (RDC) and Horticulture Farm, Coastal Plain Experiment Station, Tifton, Georgia. The three most common fungi wereCoprinus plicatilis, Cyathus olla, andPanaeolina foenisecii. At the former site, residue management practices were established for evaluating crop production and at the Horticulture Farm different crops were grown with conservation tillage employing a Low Input Sustainable Vegetable Production system. Tillage treatments at the RDC were no-till, row-till, ridge-plant, and moldboard-plowing. Residue management of triticale included burning or cutting the stubble at a height of 20 or 60 cm at harvest. At the Horticulture Farm, seven winter cover crops were compared to a fallow control. No significant differences were shown between observations of fungal species and individual treatments at the Horticulture Farm during 1989 and 1990. However, there were treatment differences for observed fungi at the RDC farm on specific dates for both years. Furthermore, no individual treatment(s) during the investigation increased observation rates of specific fungal species. Non-burned debris treatments showed consistently higher populations and diversity of fungi than burn debris plots.  相似文献

Ischemic dysfunction, including contracture, has been attributed to lack of ATP, although previous work has not been consistent with this concept. We describe here a model of no flow ischemic arrest, characterized by depressed levels of mechanical function upon reperfusion and high energy phosphate stores within normal limits. The decreased mechanical function bears an inverse relationship to myocardial lactate levels after twenty-minutes of reperfusion in the absence or presence of dichloroacetic acid (DCA). Post-ischemic non-DCA treated hearts attained peak work of only 25% of that of controls, while those treated with DCA following ischemia performed almost as well as controls. ATP and CP levels remained high in both DCA treated and non-DCA treated hearts. Lactate levels were high in hearts immediately following ischemia, but were reduced to control levels in post-ischemic hearts perfused with DCA within twenty minutes, whereas those not treated with DCA had lactate levels two to three times that of controls within the same time period. Pyruvate dehydrogenase (PDH) activity was reduced in non-DCA treated post ischemic hearts after twenty minutes reperfusion but was elevated above controls in hearts reperfused with DCA. The data indicates that DCA increases mechanical performance of the isolated post-ischemic rat heart and the proposed mechanism for this increase is the oxidative removal of lactate resulting from an increase in PDH activity.  相似文献

In husks, the activity of gibberellin-like substances extracted with aqueous methanol (M-“free” GAs) showed a maximum on the 9th day after pollination. In developing embryos, M-“free” GAs showed no biological activity, whereas biological active component(s) were obtained when the embryos were extracted with Tris buffer. The “free” GAs found in the buffer homogenates (B-“free” GAs) of developing embryos showed a maximum of activity on the 33rd day after pollination. Bound GAs recovered from the precipitated protein of the buffer homogenate (“Protein-bound” GAs) were found in embryo and endosperm. Developing endosperm generally contains the major amount of the extractable gibberellin-like substances. In this tissue, the amount of all examined fractions (M-“free” GAs, B-“free” GAs and “protein-bound” GAs) increased after pollination to reach a maximum on the 21st day, before decreasing to a minimum at grain maturity. Moreover, the curves for dry weight increase and gibberellin like substances follow a remarkably similar course, with the latter reaching its maximim slightly earlier than the former one. This result indicates that gibberellines may participate in the regulation of the accumulation process in the endosperm of barley grain.  相似文献
