Summary Some basic parameters of the life history of Alsodes montanus and Alsodes tumultuosus (Anura-Leptodactylidae), were studied from 1977 to 1980 by periodic field observations at Farellones and La Parva (33–34° south lat.; 2,700–3,000 m above sea level). Special attention was paid to strategies of resource partitioning in relation to gross features of the environment. The latter was unstable with a relative short period favorable for activity of the animals. Physical environmental differences between the first and second season of this study, resulted in a decrease in total number of active adults, a reduction in the duration of larval activity and a shift in microhabitat preferences of larvae.During the favorable season, October to May, adults of both species showed spatial and temporal segregation, related to different physical features of the environment; larvae did not show temporal segregation. Larvae of both species were found in seven different microhabitats; only in one of these did they show significant difference in microhabitat preference, A. tumultuosus was found more often in crevices. Microhabitat dimensions were more important than time and food resources in the separation of the niches of the two species. The segregation of niche dimensions, microhabitat, diel and annual activity and food were not complementary.Coexistence was therefore observed with the species tending to use different resources. We have identified a 1-kb genomic sequence that represents the major class of 5S rRNA genes in the nematode Caenorhabditis elegans. This 1-kb sequence is tandemly repeated 110 times in the haploid genome forming a single homogeneous gene family. Other nematode genomic sequences, distinct from the major 1-kb repeat class but homologous to it, may represent dispersed 5S rRNA genes or the ends of a gene cluster. One such fragment shows a restriction fragment length difference between two C. elegans strains. This should allow the genetic analysis of 5S rRNA-coding DNA (5S X rDNA) and its flanking regions in C. elegans. 相似文献

The phosphoenolpyruvate-D-glucose phosphotransferase system of Enterobacteriaceae is thought to regulate the synthesis and activity of a number of catabolite uptake systems, including those for maltose, lactose, and glycerol, via the phosphorylation state of one of its components, IIIGlc. We have investigated the proposal by Kornberg and co-workers (FEBS Lett. 117(Suppl.):K28-K36, 1980) that not IIIGlc, but an unknown protein, the product of the iex gene, is responsible for the exclusion of the above-mentioned compounds from the cell. The iex mutant HK738 of Escherichia coli contains normal amounts of IIIGlc as measured by specific antibodies, in contrast to crr mutants that lack IIIGlc. The IIIGlc of the iex strain functions normally in glucose and methyl alpha-glucoside transport, and the specific activity in in vitro phosphorylation is approximately 60% of that of the parent. The IIIGlc activity of the iex strain is, however, heat labile, in contrast to the parental IIIGlc, suggesting that the mutant contains an altered IIIGlc. This is supported by the observation that IIIGlc from the iex strain cannot bind to the lactose carrier. Thus it cannot inhibit the carrier, and this explains why the uptake of non-phosphotransferase system compounds in an iex strain is resistant to phosphotransferase system sugars. The introduction of a plasmid containing a wild-type crr+ allele into the iex strain restores the iex phenotype to that of the iex+ parent. The IIIGlc produced from the plasmid in the iex strain is heat stable and binds normally to the lactose carrier. These results lead to the conclusion that the iex mutation is most likely allelic with crr and results in an altered, temperature-sensitive IIIGlc that is still able to function D-glucose and methyl alpha-glucoside uptake and phosphorylation and in the activation of adenylate cyclase, but is unable to bind to and inhibit the lactose carrier. 相似文献

Changes in peroxidase activity during pregnancy were examined in CD-1 mice. Peroxidase activity was measured with guaiacol as the substrate in uterine extracts of nonpregnant mice and in uterine, placental, and fetal extracts of pregnant mice on days 9, 12, 14, 16, and 18 of gestation. Uterine peroxidase activity in nonpregnant mice was high, but declined logarithmically to only 0.2% by day 18 of pregnancy. In contrast to this decline, a concomitant 50-fold logarithmic increase in fetal peroxidase activity was observed between day 12 and 18. Activity in placental extracts did not change significantly throughout the gestational period examined. These results suggest that membrane bound peroxidase in mouse uterus and fetus undergoes major shifts during pregnancy. 相似文献

A flow-dialysis apparatus suitable for the study of high-affinity metal-binding proteins has been utilized to study calmodulin-metal exchange as a measure of relative calmodulin-metal association constants. Calmodulin labelled with radioactive 153Gd was dialysed against buffer containing various competing metal ions. The rate of label exchange was monitored by a gamma-ray scintillation detector. Competing metals used were Ca2+ and Cd2+, and the lanthanides Gd3+, Eu3+, La3+ and Lu3+. All exchange processes were first-order, and two categories of metal were found: Ca2+ and Cd2+ in one, the lanthanides comprising the other. In addition calmodulin-metal complexes with radioactive 109Cd and 45Ca released the bound label without any competing metal being added to the buffer. The kinetics of this metal loss can be described by two consecutive first-order processes, and the fraction of label associated with each rate can be determined. Studies of phosphodiesterase activation by calmodulin show Cd2+ and calmodulin to cause 80% of the maximum activation found when Ca2+ and calmodulin are used. 相似文献

We recently developed a procedure for the molecular analysis of specific mammalian chromosomal fragments. This procedure allows for the transfer of contiguous chromosomal fragments, varying in size from a fraction to several centimorgans in length, from the donor cell of one species into a recipient cell of a different species. Specifically, we inserted a neor gene, encoded by a recombinant retrovirus, into the murine major histocompatibility complex (MHC). Metaphase chromosome transfers with this neor-tagged chromosome into recipient hamster, primate, and canine fibroblasts produced a panel of primary neor transferents, each containing a portion of, or all of, the murine MHC. A cosmid library was made from one such transferent, CHMD(D)B1. Cosmid clones were divided, using species-specific repeat probes, into those containing murine (donor) DNA sequences and those containing sequences derived from the recipient cell. The murine-specific cosmids were clustered into overlapping DNA segments by restriction enzyme digest analysis of the cosmid DNAs coupled with Southern blot analysis with, as probes, murine-specific repeat sequences and nick-translated murine genomic DNA. These cosmid clusters were analyzed for their position within or outside of the MHC, using recombinant mouse strains, and for the presence within them of known murine MHC genes. 相似文献

The incidental catch of northern sea lions ( Eumetopias jubatus ) in the walleye pollock ( Theragra chalcogramma ) joint-venture fishery in Shelikof Strait, Alaska, was studied during 1982–1984 to assess the nature and magnitude of the catch. Data were obtained by placing U.S. observers on foreign processing vessels. Dead sea lions recovered from trawl nets were counted, sexed and measured, teeth were removed for age determination by dental laminae; and stomach contents were analyzed. Although the fishery has continued to expand both in number of boats and estimated total catch (74,136 metric tons [t] in 1982 to 171,539 t in 1984), the estimated incidental catch of northern sea lions has declined (ranging from 958 to 1,436 in 1982, 216 to 324 in 1983 and 237 to 355 in 1984). Of the sea lions processed, 73 percent were caught between 2000 and 0500 h, probably during net retrieval. Most caught sea lions were females ranging in age from 1–25 yr with a mean age of 6.43 yr; 79 percent of the females were sexually mature and probably part of the reproducing population. Males had a mean age of 4.8 yr and only 12 percent were old enough to obtain and defend territories. Analysis of stomach contents showed that the sea lions consumed pollock the same size as that taken by the commercial fishery. The impact of the incidental catch on the Gulf of Alaska sea lion population is unknown. 相似文献

Increased longevity and recently intensified emphasis on gerodontology mandate that dental students'exposure to clinical care of the elderly be enhanced. The extent to which individual students can be exposed to geriatric dental care depends on the availability of elderly patients to dental school clinics and the treatment needs of these patients. The purposes of this study were (1) to determine the dental treatment needs of geriatric patients who seek care at a dental school, and (2) to ascertain if differences exist between the needs of older versus relatively young geriatric patients. Data collected on 128 elderly patients during a three month period indicate that 57% of the aged were either edentulous at examination or treatment planned to become edentulous and receive two full dentures. The remaining 43% were treatment planned to remain dentulous and receive therapies other than full upper and lower dentures. More older geriatric patients required full dentures than their younger cohorts. More young elderly required prophylaxes, scalings, root planing therapy, dental restorations, and partial dentures. Additional to their denture requirements, aged patients appear to have sufficient non-prosthetic needs to allow for meaningful gerodontic experience by students. 相似文献

Neurospora crassa mutants deficient in asparagine synthetase were selected by using the procedure of inositol-less death. Complementation tests among the 100 mutants isolated suggested that their alterations were genetically allelic. Recombination analysis with strain S1007t, an asparagine auxotroph, indicated that the mutations were located near or within the asn gene on linkage group V. In vitro assays with a heterokaryon indicated that the mutation was dominant. Thermal instability of cell extracts from temperature-sensitive strains in an in vitro asparagine synthetase assay determined that the mutations were in the structural gene(s) for asparagine synthetase. 相似文献

SWR/J mice possess two- to threefold higher 4-methylumbelliferyl sulfate (4MUS), dehydroepiandrosterone sulfate (DHEAS) and estrone sulfate (E1S) sulfatase activities in liver and kidney extracts than do A/J mice. These interstrain activity differences are maintained throughout the 6- to 45-day postnatal period. Characteristics of the hepatic activities of SWR/J mice suggest that all three activities reside in the same enzyme. Biochemical properties of the SWR/J and A/J enzyme were not significantly different. Expression of hepatic enzyme activity is subject to regulation by an autosomal locus possessing two alleles with additive effects. Postnuclear E1S- and DHEAS-sulfatase activities are primarily microsomal. Although postnuclear hepatic 4MUS-sulfatase activity is predominantly microsomal, renal activity is primarily nonmicrosomal. Only that portion of 4MUS-sulfatase occurring in cell membranes appears capable of hydrolyzing E1S and DHEAS. The hepatic- and renal-specific subcellular distributions of 4MUS-sulfatase activity may reflect tissue differences in enzyme processing. Renal 4MUS-sulfatase activity is also controlled by an autosomal gene with two alleles having additive effects. Positive correlation between hepatic and renal 4MUS-sulfatase activities indicates that both activities are most likely influenced by the same gene. 相似文献
