Salt stress is considered one of the main abiotic factors to limit crop growth and productivity by affecting morpho-physiological and biochemical processes. Genetically, a number of salt tolerant Brassica varieties have been developed and introduced, but breeding of such varieties is time consuming. Therefore, current focus is on transgenic technology, which plays an important role in the development of salt tolerant varieties. Various salt tolerant genes have been characterized and incorporated into Brassica. Therefore, such genetic transformation of Brassica species is a significant step for improvement of crops, as well as conferring salt stress resistance qualities to Brassica species. Complete genome sequencing has made the task of genetically transforming Brassica species easier, by identifying desired candidate genes. The present review discusses relevant information about the principles which should be employed to develop transgenic Brassica species, and also will recommend tools for improved tolerance to salinity.  相似文献

This paper reports the successful excitation of surface plasmon polaritons (SPPs) through 1D metallic grating on higher refractive index GaP substrate. Coupling efficiency (η) of a free-space transverse-magnetic (TM) plane-wave mode into a SPP mode is crucial for many plasmonic devices. This η predominantly depends on the fabrication (milling) parameters and the factors (under- and over-milling) affecting the η is investigated experimentally and numerically. First of all, η is estimated by measuring the transmission spectra obtained through the plasmonic grating structures by varying the slit width (a) for a fixed period (Λ) and the thickness (t) of the gold (Au) film in which the grating is formed. The wave vector of the incident light is tuned to match the wave vector of the SPP, to get maximum η. For an optimum Au film thickness, a slit width of half of the periodicity of 770 nm in the grating device yields a maximum η. Such grating devices support only a fundamental plasmonic mode because the profile/shape of the slit in the grating device is more like a sinusoidal nature. Furthermore, such grating offers intermediate scattering to the incident light and the SPP as well which in-truns couple more incident energy to the SPPs. Moreover, over-milling results in decreased η where the crystalline plane of the substrate is disturbed. Finite element method (FEM) in COMSOL modeling is used to understand the underlying physics. Plant growth-promoting endophytic fungi with gibberellin-producing ability were isolated from the roots of Carex kobomugi Ohwi, a common sand-dune plant, and bioassayed for plant growth-promotion. A new strain, Arthrinium phaeospermum KACC43901, promoted growth of waito-c rice and Atriplex gemelinii. Analysis of its culture filtrate showed the presence of bioactive GA1 (0.5 ng/ml), GA3 (8.8 ng/ml), GA4 (4.7 ng/ml) and GA7 (2.2 ng/ml) along with physiologically inactive GA5 (0.4 ng/ml), GA9 (0.6 ng/ml), GA12 (0.4 ng/ml), GA15 (0.4 ng/ml), GA19 (0.9 ng/ml) and GA24 (1.8 ng/ml). The fungal isolate was identified through sequence homology and phylogenetic analysis of 18S rDNA (internal transcribed region). Plant growth-promoting bacteria-mediated biocontrol of plant pathogens is renowned to enhance the growth of the plants using different direct or indirect mechanisms. The goal of the present investigation was the evaluation of Pseudomonas aeruginosa Z5 isolated from cotton grown in Pakistani soils for the suppression of Fusarium oxysporum associated with cotton seedling disease. In dual culturing techniques, four bacterial strains inhibited fungal pathogens, i.e. F. oxysporum, Fusarium moniliforme, Fusarium solani and Rhizoctonia solani, significantly with percent inhibition ranging from 25% to 91.5%. P. aeruginosa Z5 showed maximum suppression of all the tested pathogens. Net-house experiments showed that the application of P. aeruginosa Z5 both separately and in combination with Bacillus fusiformis S10 significantly reduced the disease incidence by suppressing F. oxysporum (the causal agent of cotton seedling disease) up to 64–65% and improved the percent germination as compared to the infected control plants. The production of antibiotics, proteases and siderophores may be the contributing factors for its antagonistic properties. Highest bacterial population (8.9 CFU/g root) observed on roots of cotton plants inoculated with P. aeruginosa Z5 showed its good colonisation aptitudes even in the presence of high inoculation of soil with F. oxysporum. Confocal laser scanning microscopy supported the root colonisation of cotton plants with fluorescently labelled P. aeruginosa Z5. Because of innate fungicidal potential, growth promoting P. aeruginosa Z5 can be used as a bioinoculant and an antagonist to suppress the growth of cotton root-associated fungal pathogen.  相似文献

We report a simple 1D grating device fabrication on ~50 nm gold (Au) film deposited on glass, which is employed as a high performance refractive index (RI) sensor by exploiting the surface plasmon polaritons (SPP) excited by the grating device along the Au/analyte interface. A finite element analysis (FEA) method is employed to maximize the sensitivity of the sensor for a fixed period and thickness of a gold film and its close correspondence with experiment has given the insight for high sensitivity and enhanced transmission. Significantly, in the context of economic design and performance, it is shown that an optimally designed and fabricated 1D grating can be as sensitive as 524 nm/RIU (linearity RI?=?1.33303 to 1.47399), which is remarkably higher than existing reports operating in a similar wavelength region.  相似文献

Desmoplastic malignancies such as cholangiocarcinoma (CCA) are characterized by a dense stroma containing an abundance of myofibroblasts termed cancer-associated fibroblasts (CAF). The CAF phenotype represents an “activated state” in which cells are primed for cell death triggered by BH3 mimetics. Accordingly, this primed state may be therapeutically exploited. To elucidate the mechanisms underlying this poorly understood apoptotic priming, we examined the role of platelet-derived growth factor (PDGF) in CAF priming for cell death given its prominent role in CAF activation. PDGF isomers PDGF-B and PDGF-D are abundantly expressed in CCA cells derived from human specimens. Either isomer sensitizes myofibroblasts to cell death triggered by BH3 mimetics. Similar apoptotic sensitization was observed with co-culture of myofibroblasts and CCA cells. Profiling of Bcl-2 proteins expressed by PDGF-primed myofibroblasts demonstrated an increase in cellular levels of Puma. PDGF-mediated increases in cellular Puma levels induced proapoptotic changes in Bak, which resulted in its binding to Bcl-2. Short hairpin RNA-mediated down-regulation of Puma conferred resistance to PDGF-mediated apoptotic priming. Conversely, the BH3 mimetic navitoclax disrupted Bcl-2/Bak heterodimers, allowing Bak to execute the cell death program. Treatment with a Bcl-2-specific BH3 mimetic, ABT-199, reduced tumor formation and tumor burden in a murine model of cholangiocarcinoma. Collectively, these findings indicate that apoptotic priming of CAF by PDGF occurs via Puma-mediated Bak activation, which can be converted to active full-blown apoptosis by navitoclax or ABT-199 for therapeutic benefit.  相似文献

This paper reports the excitation of surface plasmon polaritons (SPPs) and associated plasmonic band gap (PBG) while using TM plane wave interacting with 1D metallic grating on higher refractive index GaP substrate. A simple method is introduced to estimate the PBG which is crucial for many plasmonic devices. The PBG is estimated by measuring the transmission spectra obtained through the plasmonic grating structures when slit width is varied while periodicity and the thickness of the gold (Au) film remained fixed. The PBG is observed for the grating devices whose slit width is less than one third of the periodicity which is caused by the presence of a higher plasmonic mode. The PBG is absent for the grating device whose slit width is slightly less than half and greater than one third of the periodicity. Such grating devices support only a fundamental plasmonic mode because the profile/shape of the slit in the grating device is more like a sinusoidal nature. Furthermore, such grating offers intermediate scattering to the incident light and the SPP as well which in turn couple more incident energy to the SPPs. Far-field modelling results also support the results obtained through experiment.
