Seroprevalence of toxoplasmosis in the residents of Cheju island, Korea

This study was performed to evaluate the epidemiological status of toxoplasmosis among the residents of Cheju island. The sera of local students from 18 high schools (boys 2110, girls 2460) and those of adults (474 admitted to Cheju Chungang General Hospital) were collected and checked for the IgG antibody titers against Toxoplasma gondii. Serum samples collected from both the students and adults showed sero-positive rate of 5.5% and 12.9%, respectively. Although the rates were not significantly different between the sexes (5.4% for the boys and 5.5% for the girls attending school), the geographical difference showed a significant difference between the urban (4.6-6.9%) and rural areas (5.6-8.8%) (p < 0.05). Based on the high positive rates, it should be necessary to control toxoplasmosis in Cheju island.     

Orexins: a role in medullary sympathetic outflow

Orexin A and B, also known as hypocretin 1 and 2, are two recently isolated hypothalamic peptides. As orexin-containing neurons are strategically located in the lateral hypothalamus, which has long been suspected to play an important role in feeding behaviors, initial studies were focused on the involvement of orexins in positive food intake and energy metabolism. Recent studies implicate a more diverse biological role of orexins, which can be manifested at different level of the neuraxis. For example, canine narcolepsy, a disorder with close phenotypic similarity to human narcolepsy, is caused by a mutation of hypocretin receptor 2 gene. Results from our immunohistochemical and functional studies, which will be summarized here, suggest that the peptide acting on neurons in the rostral ventrolateral medulla augment sympathoexcitatory outflow to the spinal cord. This finding is discussed in the context of increased sympathetic activity frequently associated with obesity.     

Wastewater Treatment in a Packed-Bed Reactor with Immobilized Cells onto a New Ceramic Carrier

A new porous ceramic carrier based on casting sand, sawdust and zeolite was used to immobilize microorganisms for continuous wastewater treatment in a packed-bed bioreactor. Scanning electron microscopy showed that the ceramic carrier has a high porosity with diameters of around 5~300µm. Immobilization capacity of the carrier was 62 mg/ g dry carrier, in terms of mixed liquor volatile suspended solids. When synthetic wastewater (chemical oxygen demand 250 mg/l) as a model feed solution and 30 mg kaolin/l as a suspended solid (SS) were used, the removal efficiency as measured by the total organic carbon and SS were 91% and 71%, respectively, at a hydraulic retention time of 3 h.     

Sister chromatid cohesion in mitosis

Sister chromatid cohesion is essential for accurate chromosome segregation during the cell cycle. Newly identified structural proteins are required for sister chromatid cohesion and there may be a link in some organisms between the processes of cohesion and condensation. Proteins that induce and regulate the separation of sister chromatids have also been recently identified.

为研究糜酶基因在体内的结构与功能以及与心肌肥厚的关系,并提高糜酶基因在小鼠心脏中的表达,构建肌球蛋白轻链－２启动子（ｍｙｏｓｉｎｌｉｇｈｔｃｈａｉｎ－２ｐｒｏｍｏｔｅｒ,ＭＬＣ２）－糜酶融合基因并产生转基因小鼠．通过删除糜酶基因启动子序列,构建结构基因克隆,然后与大鼠心脏肌球蛋白轻链－２启动子序列相拼接,构建ＭＬＣ２－糜酶融合基因克隆,回收并纯化融合基因片段,显微注射入小鼠受精卵产生转基因小鼠,经ＰＣＲ扩增、Ｓｏｕｔｈｅｒｎ印迹杂交和ＰＣＲ扩增产物的测序,筛选和确定转基因鼠．在新出生的４６只小鼠中有２只为转基因阳性鼠,且外源基因能稳定遗传给后代,从而获得了可用于研究糜酶基因在体内的结构与功能以及与心肌肥厚的关系的转基因小鼠模型．

A simple and sensitive ribonucleotide reductase assay

Ribonucleotide reductase (RR) is a key regulatory enzyme in the DNA synthesis pathway and is the target of the cancer chemotherapeutic agent hydroxyurea. The study of RR is significantly hindered by the tedious and labor-intensive nature of enzymatic assay. In this report, we present a novel RR assay in which detection of the deoxyribonucleotides produced by RR occurs via coupling to the DNA polymerase reaction, and is enhanced by using RNase to degrade endogenous RNA. Cell extracts from various cell lines were treated with RNase and then reacted with ATP and radioactive ribonucleotide diphosphate as the substrate. Incorporation of the radioactive substrate [¹⁴C]CDP into DNA was linear over 30 min and was linear with the amount of extract, which provided RR activity. The reaction was inhibited by hydroxyurea and required Mg²⁺ and ATP, suggesting that the assay is specific to RR activity. While RR activities determined by our method and by a conventional method were comparable, this novel method proved to be simpler, faster, more sensitive and less expensive. In addition, assay of the RR activity for multiple samples can easily be performed simultaneously. It is superior to other RR assays in all aspects.     

Optimization of cellulase production in batch fermentation byTrichoderma reesei

Maximum cellulase production was sought by comparing the activities of the cellulases produced by differentTrichoderma reesei strains andAspergillus niger. Trichoderma reesei Rut-C30 showed higher cellulase activity than otherTrichoderma reesei strains andAspergillus niger that was isolated from soil. By optimizing the cultivation condition during shake flask culture, higher cellulase production could be achieved. The FP (filter paper) activity of 3.7 U/ml and CMCase (Carboxymethylcellulase) activity of 60 U/ml were obtained from shake flask culture. When it was grown in 2.5L fermentor, where pH and DO levels are controlled, the Enzyme activities were 133.35 U/ml (CMCase) and 11.67 U./ml (FP), respectively. Ammonium sulfate precipitation method was used to recover enzymes from fermentation broth. The dried cellulase powder showed 3074.9 U/g of CMCase activity and 166.7 U/g of FP activity with 83.5% CMCase recovery.
