Botulinum neurotoxins (BoNTs), the most poisonous member of class A biothreat agent, cause neuroparalysis by blocking neurotransmitter release at the neuromuscular junctions. In its mechanism of action, the catalytic domain (light chain (LC) of BoNT) is transported to the cytosol by the heavy chain (HC) in order to reach its proteolytic substrates. The BoNT HC forms a membrane channel under acidic conditions encountered in endosomes to serve as a passageway for LC to enter into cytosol. We demonstrate here that BoNT/A LC undergoes unique structural changes under the low pH conditions, and adopts a molten globule state, exposing substantial number of hydrophobic groups. The flexibility of the molten globular structure combined with retention of the secondary structure and exposure of specific residues of LC for interaction with the HC, allows its translocation through the narrow endosomal membrane channel. 相似文献

1. In many animal populations a small proportion of individuals produce the majority of surviving offspring, but the underlying mechanisms are unclear. Behaviour may be an important determinant of variation in fitness: 'high-quality' individuals may have enhanced abilities in foraging or predator and parasite avoidance. 2. The role of behaviour in determining variation in quality was examined using the common guillemot Uria aalge, a monogamous seabird with biparental care. Using a novel mixed model approach, we analysed binary data on breeding success of each pair attempting to breed in each year with variables critical to breeding success (timing of breeding; inferred age; breeding experience and success; number of nest sites and partners) as fixed effects. Random effects for year, male, female and each distinct pairing of a male and a female were included in the model, allowing a quality estimate to be derived for each individual and pair. A range of behaviours associated with breeding were examined in relation to these quality estimates. 3. Breeding success declined with timing of breeding, and increased initially with age before declining in old age. It increased with previous successful experience, not breeding experience per se, until senescence effects became apparent. For males, breeding success declined with increasing numbers of mates. 4. The most important behavioural determinants of quality operated at the level of the pair, with the time mates spent together at the site and chick feeding rates both positively related to quality. At the individual level, trip durations and feeding rates were associated with female but not male quality, suggesting that pair quality was operating principally through the female. However, removal of laying date, the most important component in the binomial model, confirmed that the pair effect was much larger than the female effect. 5. This study demonstrates the potential of mixed modelling to determine quality estimates based on long-term breeding histories. The probability of a successful reproductive attempt was explained by the timing of breeding, age, successful breeding experience and number of mates. Behaviour was an important proximate mechanism underlying quality, in particular the foraging abilities of the pair, and the female's contribution to offspring provisioning. In species with biparental care, behavioural correlates of quality operate most strongly at the scale of the breeding pair, because contributions from both individuals are required for a successful outcome. 相似文献

Body size is a major component of fitness. However, the relative contributions of different factors to optimal size, and the determinants of spatial and temporal variation in size, have not been fully established empirically. Here, we use a mesocosm of a Drosophilidae assemblage inhabiting decaying nectarines to investigate the influence of spatial variation in temperature on adult body size in Drosophila simulans Sturtevant. Two treatments were established; one in the sun where developing larvae were exposed to high temperatures and the other in the shade where temperature conditions were milder. The simple developmental effects of temperature differences (i.e. larger flies are likely to emerge from cooler environments), or the simple effects of stressful temperatures (i.e. high temperatures yield wing abnormalities and smaller flies), were overridden by interactive effects between temperature and larval density. Emergences were lower in the sun than shade, probably as a result of temperature-induced mortality. However, flies attained the same final sizes in the shade and sun. In addition, abnormally winged flies were clustered in the shaded treatments. In the shade treatments, where emergences were higher than in the sun, stressful conditions as a result of high larval density likely resulted in wing abnormalities and small size. Consequently, there was little spatial variation in size across the mesocosm, but substantial spatial variation in abundance. Under natural conditions both mortality and non-lethal effects of temperature and/or crowding are likely to play a role in the evolution of body size. 相似文献

The wheat (Triticum aestivum) and rye (Secale cereale) beta-D-glucosidases hydrolyze hydroxamic acid-glucose conjugates, exist as different types of isozyme, and function as oligomers. In this study, three cDNAs encoding beta-D-glucosidases (TaGlu1a, TaGlu1b, and TaGlu1c) were isolated from young wheat shoots. Although the TaGlu1s share very high sequence homology, the mRNA level of Taglu1c was much lower than the other two genes in 48- and 96-h-old wheat shoots. The expression ratio of each gene was different between two wheat cultivars. Recombinant TaGlu1b expressed in Escherichia coli was electrophoretically distinct fromTaGlu1a and TaGlu1c. Furthermore, coexpression of TaGlu1a and TaGlu1b gave seven bands on a native-PAGE gel, indicating the formation of both homo- and heterohexamers. One distinctive property of the wheat and rye glucosidases is that they function as hexamers but lose activity when dissociated into smaller oligomers or monomers. The crystal structure of hexameric TaGlu1b was determined at a resolution of 1.8 A. The N-terminal region was located at the dimer-dimer interface and plays a crucial role in hexamer formation. Mutational analyses revealed that the aromatic side chain at position 378, which is located at the entrance to the catalytic center, plays an important role in substrate binding. Additionally, serine-464 and leucine-465 of TaGlu1a were shown to be critical in the relative specificity for DIMBOA-glucose (2-O-beta-D-glucopyranosyl-4-hydroxy-7-methoxy-1,4-benzoxazin-3-one) over DIBOA-glucose (7-demethoxy-DIMBOA-glucose). 相似文献

Plant germplasm collections invariably contain varying levels of genetic redundancy, which hinders the efficient conservation
and utilization of plant germplasm. Reduction of genetic redundancies is an essential step to improve the accuracy and efficiency
of genebank management. The present study targeted the assessment of genetic redundancy and genetic structure in an international
cacao (Theobroma cacao L.) collection maintained in Costa Rica. A total of 688 cacao accessions maintained in this collection were genotyped with
15 simple sequence repeat (SSR) loci, using a capillary electrophoresis genotyping system. The SSR markers provided a high
resolution among the accessions. Thirty-six synonymously labeled sets, involving 135 accessions were identified based on the
matching of multilocus SSR profiles. After the elimination of synonymous sets, the level of redundancy caused by closely related
accessions in the collection was assessed using a simulated sampling scheme that compared allelic diversity in different sample
sizes. The result of the simulation suggested that a random sample of 113 accessions could capture 90% of the total allelic
diversity in this collection. Principal Coordinate Analysis revealed that the Trinitario hybrids from Costa Rica shared a
high similarity among groups as well as among individual accessions. The analysis of the genetic structure illustrated that
the within-country/within-region difference accounted for 84.6% of the total molecular variation whereas the among-country/among-region
difference accounted for 15.4%. The Brazilian germplasm contributed most to this collection in terms of total alleles and
private alleles. The intercountry/interregion relationship by cluster analysis largely agreed with the geographical origin
of each germplasm group and supported the hypothesis that the Upper Amazon region is the center of diversity for cacao. The
results of the present study indicated that the CATIE International Cacao Collection contains a high level of genetic redundancy.
It should be possible to rationalize this collection by reducing redundancy and ensuring optimal representation of the genetic
diversity from distinct germplasm groups. The results also demonstrated that SSR markers, together with the statistical tools
for individual identification and redundancy assessment, are technically practical and sufficiently informative to assist
the management of a tropical plant germplasm collection. 相似文献

Na+- Ca2 + exchanger (NCX) has been proposed to play a role in refilling the sarco/endoplasmic reticulum (SER) Ca2 + pool along with the SER Ca2 + pump (SERCA). Here, SERCA inhibitor thapsigargin was used to determine the effects of SER Ca2 + depletion on NCX–SERCA interactions in smooth muscle cells cultured from pig coronary artery. The cells were Na+-loaded and then placed in either a Na+-containing or in a Na+-substituted solution. Subsequently, the difference in Ca2 + entry between the two groups was examined and defined as the NCX mediated Ca2 + entry. The NCX mediated Ca2 + entry in the smooth muscle cells was monitored using two methods: Ca2 +sensitive fluorescence dye Fluo-4 and radioactive Ca2 +. Ca2 +-entry was greater in the Na+-substituted cells than in the Na+-containing cells when measured by either method. This difference was established to be NCX-mediated as it was sensitive to the NCX inhibitors. Thapsigargin diminished the NCX mediated Ca2 + entry as determined by either method. Immunofluorescence confocal microscopy was used to determine the co-localization of NCX1 and subsarcolemmal SERCA2 in the cells incubated in the Na+-substituted solution with or without thapsigargin. SER Ca2 + depletion with thapsigargin increased the co-localization between NCX1 and the subsarcolemmal SERCA2. Thus, inhibition of SERCA2 leads to blockade of constant Ca2 + entry through NCX1 and also increases proximity between NCX1 and SERCA2. This blockade of Ca2 + entry may protect the cells against Ca2 +-overload during ischemia–reperfusion when SERCA2 is known to be damaged. 相似文献

Competitive repair by naturally dispersed repetitive DNA during non-allelic homologous recombination

Genome rearrangements often result from non-allelic homologous recombination (NAHR) between repetitive DNA elements dispersed throughout the genome. Here we systematically analyze NAHR between Ty retrotransposons using a genome-wide approach that exploits unique features of Saccharomyces cerevisiae purebred and Saccharomyces cerevisiae/Saccharomyces bayanus hybrid diploids. We find that DNA double-strand breaks (DSBs) induce NAHR-dependent rearrangements using Ty elements located 12 to 48 kilobases distal to the break site. This break-distal recombination (BDR) occurs frequently, even when allelic recombination can repair the break using the homolog. Robust BDR-dependent NAHR demonstrates that sequences very distal to DSBs can effectively compete with proximal sequences for repair of the break. In addition, our analysis of NAHR partner choice between Ty repeats shows that intrachromosomal Ty partners are preferred despite the abundance of potential interchromosomal Ty partners that share higher sequence identity. This competitive advantage of intrachromosomal Tys results from the relative efficiencies of different NAHR repair pathways. Finally, NAHR generates deleterious rearrangements more frequently when DSBs occur outside rather than within a Ty repeat. These findings yield insights into mechanisms of repeat-mediated genome rearrangements associated with evolution and cancer. 相似文献
