The behaviour of endogenous Mn2+ was studied by electron spin resonance spectro-scopy during benzyladenine-induced growth of excised cucumber ( Cucumis sativis L. cv. Long green) cotyledons. The level of endogenous Mn2+ was decreased by ben-zyladenine treatment, most pronounced after 96 h. MnCl2 applied alone promoted chlorophyll synthesis at relatively low concentrations but in the presence of ben-zyladenine higher concentrations of MnCl2 were required for stimulation of chlorophyll synthesis. A pronounced increase in growth was observed when Mn2+ was applied with benzyladenine at 96 h, when the decline in the endogenous level of paramagnetic Mn2+ was maximal.  相似文献

Quantitative Real-Time PCR (qPCR) is a preferred and reliable method for accurate quantification of gene expression to understand precise gene functions. A total of 25 candidate reference genes including traditional and new generation reference genes were selected and evaluated in a diverse set of chickpea samples. The samples used in this study included nine chickpea genotypes (Cicer spp.) comprising of cultivated and wild species, six abiotic stress treatments (drought, salinity, high vapor pressure deficit, abscisic acid, cold and heat shock), and five diverse tissues (leaf, root, flower, seedlings and seed). The geNorm, NormFinder and RefFinder algorithms used to identify stably expressed genes in four sample sets revealed stable expression of UCP and G6PD genes across genotypes, while TIP41 and CAC were highly stable under abiotic stress conditions. While PP2A and ABCT genes were ranked as best for different tissues, ABCT, UCP and CAC were most stable across all samples. This study demonstrated the usefulness of new generation reference genes for more accurate qPCR based gene expression quantification in cultivated as well as wild chickpea species. Validation of the best reference genes was carried out by studying their impact on normalization of aquaporin genes PIP1;4 and TIP3;1, in three contrasting chickpea genotypes under high vapor pressure deficit (VPD) treatment. The chickpea TIP3;1 gene got significantly up regulated under high VPD conditions with higher relative expression in the drought susceptible genotype, confirming the suitability of the selected reference genes for expression analysis. This is the first comprehensive study on the stability of the new generation reference genes for qPCR studies in chickpea across species, different tissues and abiotic stresses.  相似文献

Salt stress is a major environmental factor in arid and semi-arid regions and influences many aspects of plant development. Salinity results in generation of various free radicals that can potentially damage the cellular constituents in plants. Plants were able to effectively reduce the damage caused by these free radicals by a way of enzymatic and non enzymatic defenses for better survival. Enhanced efficacy of antioxidative enzyme systems such as superoxide dismutase, catalase and ascarbate peroxidase was well documented in several plants subjected to salinity stress. Aldose reductase, an important enzyme is also known to detoxify free toxic aldehydes like HNE (4-hydroxynon-2-enal, a hydroxyalkenal) generated during oxidative damage of cellular components. However, the role of aldose reductase to impart tolerance to the plants under salt stress has not been studied in any detail. Therefore, we were interested to study the aldose reductase activity and its expression to gain an insight into the role of aldose reductase in imparting tolerance to foxtail millet cultivars (viz., Cv. Prasad and Lepakshi) subjected to NaCl stress. We observed that subjecting foxtail millets to increasing levels of stress significantly increased aldose reductase activity and in a way that correlated positively with elevated levels of sorbitol, an osmotic solute involved in osmotic balance. This suggests the involvement of aldose reductase in sorbitol biosynthesis in foxtail millet. Additionally, we observed higher levels of 4-hydroxynon-2-enal, a major lipid peroxidation product, in the susceptible than the tolerant cultivar indicating a higher proportion of cellular damage in former than in the latter. This high content of 4-hydroxynon-2-enal in the susceptible cultivar was negatively correlated with its aldose reductase activity, indicating the involvement of aldose reductase in detoxification of 4-hydroxynon-2-enal. 4-hydroxynon-2-enal is also known to be a catalyzed by glutathione-S-transferase. Glutathione-S-transferase activity was found higher in the tolerant foxtail millet than the sensitive cultivar: the tolerant cultivar showed a low 4-hydroxynon-2-enal content compared to the susceptible cultivar, demonstrating a possible mechanism for detoxification of 4-hydroxynon-2-enal by two enzymes, glutathione-S-transferase and aldose reductase in plants under stressful conditions.  相似文献

Background

Glucose restriction in cells increases the AMP/ATP ratio (energetic stress), which activates the AMPK/p53 pathway. Depending upon the energetic stress levels, cells undergo either autophagy or cell death. Given that the activated p53 induces the expression of IFI16 protein, we investigated the potential role of the IFI16 protein in glucose restriction-induced responses.

Methodology/Principal Findings

We found that glucose restriction or treatment of human diploid fibroblasts (HDFs) with the activators of the AMPK/p53 pathway induced the expression of IFI16 protein. The induced levels of IFI16 protein were associated with the induction of autophagy and reduced cell survival. Moreover, the increase in the IFI16 protein levels was dependent upon the expression of the functional ATM protein kinase. Importantly, the knockdown of the IFI16 expression in HDFs inhibited the activation of the ATM/AMPK/p53 pathway in response to glucose restriction and also increased the survival of HDFs.

Conclusions/Significance

Our observations demonstrate a role for the IFI16 protein in the energetic stress-induced regulation of autophagy and cell survival. Additionally, our findings also indicate that the loss of IFI16 expression, as found in certain cancers, may provide a survival advantage to cancer cells in microenvironments with low glucose levels.  相似文献

Secondary salinization and its relationship to irrigation are strong incentives to improve the tolerance of crops to salinity and to drought. Achieving this through the pyramiding of physiological traits (phenotypic selection without knowledge of genotype) is feasible. However, wide application of this approach is limited by the practicalities of assessing not only the parents, but also large numbers of individuals and families in segregating generations. Genotypic information is required in the form of markers for any quantitative trait loci involved (marker-assisted selection) or of direct knowledge of the genes. In the absence of adequate candidate genes for salt tolerance, a quantitative trait locus/marker-assisted selection approach has been used here. Putative markers for ion transport and selectivity, identified from analysis of amplified fragment length polymorphism, had been discovered within a custom-made mapping population of rice. Here it is reported that none of these markers showed any association with similar traits in a closely related population of recombinant inbred lines or in selections of a cultivar. Whilst markers will be of value in using élite lines from the mapping population in backcrossing, this has to be considered alongside the effort required to develop and map any given population. This result cautions against any expectation of a general applicability of markers for physiological traits. It is concluded that direct knowledge of the genes involved is needed. This cannot be achieved at present by positional cloning. The elucidation of candidate genes is required. Here the problem lies not in the analysis of gene expression but in devising protocols in which only those genes of interest are differentially affected by the experimental treatments.  相似文献

Studies evaluated the effect of rhizobacterial interactions on a virus under natural conditions of high levels of vector–virus pressure on tomato plants. The rhizobacterium Bacillus subtilis was isolated from the rhizosphere soil of healthy tomato plant and the strain is referred to as BS3A25. The BS3A25 treatment of seeds significantly enhanced tomato seed germination rates to 99% compared to the control seeds at 78%. Combination of both seed and foliar spray treatments significantly protected the plants from cucumber mosaic virus (CMV) and also significantly enhanced plant growth. Strain BS3A25 possesses many positive traits such as phosphate solubilization ability, ACC utilization as the sole source of nitrogen and production of significantly higher concentrations of indoles, IAA and the cytokinin, IPA. The growth parameters of the aphid Aphis gossypii Glover such as development time, time from birth to adult (tD) and prereproduction time (td) were longer, whereas the relative growth rate (RGR) and intrinsic rate of natural increase (rm), were found to be lower in the BS3A25 supernatant treatment compared to commercial insecticide imidacloprid treatment. Noticeably, the present study recorded higher activity of phenylalanine ammonia lyase, Peroxidase, total phenols, and polyphenol oxidase in plants treated with BS3A25 generating the speculation of induced defence responses in plants. The results obtained in the field study showed that treatment by BS3A25 formulation as seed and foliar treatment proved highly effective in reducing the aphid population and CMV incidence compared to a commercially available insecticide. The highest cost:benefit ratio (1:2.5) was obtained in BS3A25 formulation treatment followed by imidacloprid (1:2.0) and control plants (1:1.4). These constitute an important step towards the production of BS3A25 as a commercially-available induced resistance agent.  相似文献

We developed a practical and efficient gene transfer system for indica rice utilizing mature-seed derived explants and a simple bombardment device which uses compressed helium for accelerating DNA-coated metal particles. Unlike instruments which have been described in the literature previously, this new bombardment device, which is an improvement of the particle inflow concept, does not require vacuum. This attribute simplifies the transformation procedure significantly and it makes rice transformation technology accessible to laboratories which may not have the resources to invest in more expensive particle bombardment instruments. We determined experimentally that we could recover transgenic rice plants utilizing three different particle bombardment instruments at comparable frequencies.  相似文献

ABSTRACT: INTRODUCTION: Osteoporosis is an asymptomatic disease characterized by bone weakening and predisposition to fragility (insufficiency) fractures and can have devastating effects on individual life and great financial impact on the economy. Bisphosphonates are used worldwide for the primary and secondary prevention of osteoporotic fractures. However, increasing evidence raises concern that bisphosphonates can be associated with atypical fractures. CASE PRESENTATION: A 65-year-old Caucasian woman on long-term steroid treatment for polymyalgia rheumatica was admitted with severe and constant pain in the right hip, radiating to the right knee. She had a history of steroid-induced osteoporosis, for which she was started on risedronate four years earlier. She had no history of trauma. Her blood results were unremarkable. Her X-rays confirmed that she had an incomplete right subtrochanteric femoral fracture. A bone scan confirmed the diagnosis and also ruled out any other associated fractures. Our patient successfully underwent internal nail fixation of the fracture. She was reviewed by a rheumatology team, which stopped the risedronate. She was started on treatment with denosumab injection. CONCLUSIONS: Previous case series have reported that long-term bisphosphonate use is associated with atypical fractures of the femur, and certain criteria have been established to help identify such rare fractures. Delayed union or non-union is expected in such fractures following definitive orthopedic treatment because of the long half life of bisphosphonates. In this case report, we try to raise questions related to this important subject, like the duration and safety of bisphosphonate use and the alternative medications used in osteoporosis in this rare condition. We consider this case report not only interesting but also important and unusual because it is about a patient who developed a potentially rare and serious side effect of long-term bisphosphonate use, estimated to affect 2.3 in every 10,000 patients, and who presented with a pelvic X-ray that showed the characteristic features of atypical fractures secondary to risedronate use. In addition, most of the documented cases have been associated with many years of bisphosphonate use whereas our patient had been on risedronate for only four years.  相似文献

Background  

Clustering techniques are routinely used in gene expression data analysis to organize the massive data. Clustering techniques arrange a large number of genes or assays into a few clusters while maximizing the intra-cluster similarity and inter-cluster separation. While clustering of genes facilitates learning the functions of un-characterized genes using their association with known genes, clustering of assays reveals the disease stages and subtypes. Many clustering algorithms require the user to specify the number of clusters a priori. A wrong specification of number of clusters generally leads to either failure to detect novel clusters (disease subtypes) or unnecessary splitting of natural clusters.  相似文献
