Hook1 is a member of a family of microtubule-binding proteins. Studies on the Drosophila homolog of Hook1 have suggested a role in the maturation and trafficking of internalized proteins to the late endosome. A weak interaction between Hook1 and the lysosomal/late endosomal protein, CLN3, was recently reported. Mutations in CLN3 result in the neurological disorder Batten disease. Here we show a novel interaction between Hook1 and Ankyrin G, an adaptor protein that binds the spectrin-actin cytoskeleton and targets proteins to the peripheral membrane. Although we demonstrate co-localization of Hook1 and Ankyrin G, Hook1 also localizes to additional regions of the cell devoid of Ankyrin G where it likely interacts with other proteins. There is no disruption of the Hook1-Ankyrin G interaction or localization in tissue derived from a Cln3-knockout mouse despite a nearly threefold increase in the expression of Hook1. However, mutation of CLN3 could lead to alterations in the functioning and positioning of organelles and membrane proteins through this Hook1-Ankyrin G interaction. 相似文献

Transition metal-mediated oxidation of hydroquinones is an important physiologic reaction, and copper(II) effectively catalyzes the reaction in phosphate-buffered saline (PBS). Studies reported herein in phosphate buffer alone demonstrate that copper(II) is an ineffective catalyst in the absence of coordinating ligands, but that 1,10-phenanthroline and histamine facilitate the copper(II)-mediated oxidation of hydroquinone and its 2,5- and 2,6-di-tert-butyl analogs to the corresponding benzoquinones. The high concentration of chloride in PBS is the key element that allows copper(II) to work in this system. Although the bis-bathocuproine disulfonate complex of Cu(II), (BC)2Cu(II), is a strong stoichiometric oxidant, stoichiometric amounts of copper(II) in the presence of ligands other than BC oxidize hydroquinones very slowly under anaerobic conditions. Thus, the rapid copper(II)-catalyzed reaction operating aerobically does not involve a simple ping-pong reduction of copper(II) to copper(I) by hydroquinone and reoxidation of copper(I) by O2. 相似文献

The expression of mu-opioid receptors in the developing rat spinal cord (Postnatal days 7, 14, 30) was studied by autoradiography using [3H]DAMGO. When compared to camera lucida drawings, the receptor was noted over the entire gray matter and dorsal root ganglia at postnatal days 7 and 14. At postnatal day 30, the receptor expression decreased over the gray matter except the superficial laminae (laminae I and II). At all age groups studied, a higher expression of the receptor was noted over the superficial laminae. The study shows that micro-opioid receptors appears early in postnatal development and attains mature receptor distribution relatively late in ontogeny, suggesting a possible role in the normal development of nervous system. Although conventional breeding and propagation contributed significantly for last several decades for varietal improvement, due to the limitations of conventional breeding coupled with the demand for increasing productivity with lower cost of production, application of biotechnology becomes an alternative approach. Though morphine remains the most powerful drug for treating pain, its effectiveness is limited by the development of tolerance
and dependence. The mechanism underlying development of tolerance to morphine is still poorly understood. One of the factors
could be an alteration in the number of Μ-receptors within specific parts of the nervous system. However, reports on changes
in the Μ-opioid receptor density in the spinal cord after chronic morphine administration are conflicting. Most of the studies
have used subcutaneously implanted morphine pellets to produce tolerance. However, it does not simulate clinical conditions,
where it is more common to administer morphine at intervals, either by injections or orally. In the present study, rats were
made tolerant to morphine by injecting increasing doses of morphine (10-50 mg/kg, subcutaneously) for five days.In vitro tissue autoradiography for localization of Μ-receptor in the spinal cord was done using [3H]-DAMGO. As compared to the spinal cord of control rats, the spinal cord of tolerant rats showed an 18.8% increase or up-regulation
in the density of Μ-receptors in the superficial layers of the dorsal horn. This up-regulation of Μ-receptors after morphine
