Nematode resistance protein HSPRO2 of Arabidopsis thaliana has important roles in defence response to the bacterium (Pseudomonas syringae) and shows response against oxidative stress and salicylic acid. We have explored the gene encoding it from Rorippa indica (L.) Hiern, a wild relative of cultivated crucifers experimentally challenged with mustard aphid Lipaphis erysimi. The gene seems to be a strong candidate for aphid tolerance in cultivated mustards. Presently we are reporting the results of a time-course quantitative relative expression analysis of HSPRO2 of R. indica where we have observed?~?tenfold increase in its expression in R. indica at 12 hours post infestation with L. erysimi. We have also isolated the full-length gene (1314 bp) by both 5′ and 3′ RACE (Rapid Amplification of cDNA Ends). Genome walking experiment identified the promoter sequence. Expression, purification and characterisation of RiHSPRO2 showed that it encodes for a 437 amino acid peptide. The 66 kDa recombinant RiHSPRO2 protein was purified for studying its efficacy against L. erysimi in an artificial diet-based insect bioassay that revealed LC50 (Lethal Concentration 50) values of RiHSPRO2 significantly within limits against L. erysimi. The present study might have a significant implication in future towards aphid management program of Brassica juncea through the development of aphid-tolerant transgenic plants. 相似文献

Human ovarian mucinous cystadenocarcinoma-associated antigen recognized by murine monoclonal antibody 1D3 (Bhattacharya et al., 1982) was characterized. Gel filtration and sodium dodecylsulfate polyacrylamide gel electrophoresis, followed by Western-blot analysis showed that 1D3 is a high molecular weight glycoprotein. Isoelectric focusing of 1D3 antigen showed 2 overlapping antigenic components with PI 2.5 and 2.6. 1D3 antigen was extremely stable (10 min at 100 degrees C) to heating. The antigenic activity was slightly stimulated by treatment with galactosidases, but neuraminidase treatment enhanced the antigenic activity about 3-fold. Antigen activity was completely stable to periodate oxidation. Pronase and trypsin treatment completely destroyed the antigenic activity. Properties of 1D3 antigen suggest that this is a high molecular weight (approximately 5-20 x 10(6) Dalton), sialomucin. Monoclonal antibody 1D3 recognizes only the protein part of this molecule. 相似文献

We report the design, synthesis and photophysical properties of highly solvatochromic donor/acceptor substituted naphthalimide based fluorophores. The synthesized naphthalimides containing propargyl ends showed highly solvatochromic intramolecular charge transfer (ICT) feature as was revealed from the UV–visible, fluorescence photophysical properties of these fluorophores and DFT/TDDFT calculation. Fluorescence life times for the imide fluorophores were also measured in different solvents. The solid state photophysical property of donor substituted naphthalimide 1 showed promising for future application in material sciences. Furthermore, both the donor/acceptor substituted naphthalimide fluorophores 1–2 were exploited in sensing calf-thymus DNA via switch-on fluorescence response. The propargyl linker containing naphthalimides can further be exploited for the synthesis of labeled biomolecular building blocks. 相似文献

Point mutation in the human dystrophin gene: identification through western blot analysis 总被引:17,自引:0,他引:17
Using antibodies directed against the amino-terminus of dystrophin, we identified a truncated protein in a Duchenne muscular dystrophy patient. Antibodies directed against the carboxy-terminus failed to identify any cross-reactive material, a result consistent with premature termination of dystrophin translation. The estimated molecular mass of 126 kDa predicted the approximate location of the mutation in the mRNA and in the gene. Sequencing of cloned PCR products from patient muscle cDNA revealed a nonsense mutation, which was confirmed by direct sequencing of amplified patient genomic DNA. The mutation, a G to T transversion, at position 3714 changes a glutamic acid codon to an Amber stop codon. Translation of mRNA containing this mutation would be expected to result in a truncated protein with a molecular mass of 133 kDa, in close agreement with the 126 kDa estimated by Western blot analysis. This is the first reported case of a point mutation in this very large human gene. 相似文献

achieved using a novel encapsulation-based, antibiotic selection procedure. The Agrobacterium-infected micro-shoots derived from callus explants survived selection in high concentration of hygromycin (60 mg l−1 and beyond) in encapsulated alginate beads. The present study aimed at ergonomic evaluation of the prevalence of work-related musculoskeletal disorders among construction labourers working in unorganized sectors in West Bengal, India. A modified Nordic questionnaire was applied to one hundred forty male and ninety female construction labourers to acquire information about musculoskeletal symptoms like pain in different body parts. Work-rest schedules of the labourers in different work activities were studied. Working postures were analyzed by means of the Rapid Entire Body Assessment (REBA) tool. Body part discomfort (BPD) scale was used to assess the intensity of feeling of discomforts in the different body parts. It was revealed that the labourers performed repetitive, stressful work for a long period of time in a single work-rest cycle and the load lifted and carried by them were more than the NIOSH recommended weight limit. The analyses of working postures revealed that most of their working postures were unsafe and ranked under REBA action level 3 and 4. The results obtained by applying the Nordic questionnaire and BPD scale revealed that the prevalence of pain in various regions of the body, especially low-back pain, was alarmingly high in both male and female labourers. Training for safe lifting of materials, proper work-rest schedule, modifications of some working procedures and the use of ergonomically designed equipment may certainly reduce the work-related musculoskeletal disorders and improve the health status of construction labourers working in unorganized sectors. 相似文献
