香菇B交配型位点的分子遗传学结构研究I.信息素受体和信息素前体编码基因的克隆和序列分析 总被引:3,自引:1,他引:3
本研究结合简并PCR和染色体步行两种方法研究了香菇135菌株的交配型B位点的分子遗传学结构。从135菌株的原生质体单核体1号菌株中获得了1个信息素受体编码基因LErcb1-B1和1个信息素前体编码基因LEphb1-B1。经序列比对分析,香菇的信息素受体LErcb1-B1序列与灰盖鬼伞和裂褶菌的信息素受体之间具有同源性,经SOSUI软件分析该序列具有7次跨膜结构特征。信息素前体LEphb1-B1具有CaaX基序特征。 相似文献

脊髓灰质炎病毒(Poliovirus)是一类无包膜单股正链RNA病毒,基因组长约7.5kb。其5′端非编码区由约742个核苷酸长,主要与病毒RNA复制、蛋白翻译起始、病毒颗粒的装配及病毒的细胞适应减毒及神经毒力密切相关。我们研究发现,脊髓灰质炎病毒(Sabin株)在人胚肺二倍体细胞(KME17)上致细胞病变较猴肾细胞慢,病毒产量亦明显低于恒河猴肾细胞培养,国内外也有类似的报道。 相似文献

β-淀粉样蛋白(β-AP)是阿尔茨海默氏病(Alzheimer’s disease)病人老年斑的主要成分,它是β-淀粉样前体蛋白(β-APP)剪切后的产物。β-APP基因在体内存在β-APP_(695),β-APP_(751)β-APP_(770)。等几种主要的转录物,它们的区别在于Kunize丝氨酸蛋白酶抑制区(KDI)编码序列的存在和缺失。通过RT-PCR技术,用针对KPI编码区两侧序列的一种特异性引物可由总RNA样品中扩增出反映以上三种转录物的cDNA片段。实验表明,胎鼠脑组织中未检测到β-APPmRNA;6月龄大鼠海马组织,大脑皮层中只检测到β-APP_(695);在神经胶质瘤细胞系C6中存在β-APP_(695),β-APP_(751),β-APP_(770),其中β-APP_(770)占优势。 相似文献

动态监测2011年、2013年和2016年我国不同地区医院内获得性血流感染病原菌分布及耐药进展趋势。从全国10个城市回顾性收集血流感染病原菌非重复性株,采用琼脂稀释法或微量肉汤稀释法进行药物敏感性试验,采用Whonet 5.6软件对药敏试验结果进行分析。收集的2 248株血流感染病原菌中革兰阴性杆菌为1 657株 (占73.7%),革兰阳性球菌为591株 (占26.3%)。分离率排名前五的病原菌依次为大肠埃希菌 (32.6%,733株/2 248株)、肺炎克雷伯菌 (14.5%,327株/2 248株)、金黄色葡萄球菌 (10.0%,225株/2 248株)、鲍曼不动杆菌 (8.7%,196株/ 2 248株) 和铜绿假单胞菌 (6.2%,140株/2 248株)。血流感染分离的革兰阴性杆菌对抗菌药物体外敏感率较高的抗菌药物依次为粘菌素 (96.5%,1 525株/1 581株,不包括天然耐药菌株)、替加环素 (95.6%,1 375株/1 438株,不包括天然耐药菌株)、头孢他啶/克拉维酸 (89.2%,1 112株/1 246株)、阿米卡星 (86.4%,1 382株/1 599株) 和美罗培南 (85.7%,1 376株/1 605株);革兰阳性球菌对抗菌药物体外敏感率较高的抗菌药物依次为替加环素、替考拉宁和达托霉素 (敏感率均为100.0%)、万古霉素和利奈唑胺 (敏感率均为99.7%)。2011年、2013年和2016年产超广谱β-内酰胺酶肠杆菌科细菌分离率分别为50.6% (206株/407株)、49.8% (136株/273株) 和38.9% (167株/429株);碳青霉烯不敏感肠杆菌科细菌分离率分别为2.2% (9株/408株)、4.0% (16株/402株) 和3.9% (17株/ 439株);多重耐药鲍曼不动杆菌分离率分别为76.4% (55株/72株)、82.7% (43株/52株) 和87.5% (63株/72株),多重耐药铜绿假单胞菌分离率分别为9.8% (5株/51株)、20.0% (7株/35株) 和13.0% (7株/54株);甲氧西林耐药金黄色葡萄球菌的分离率分别为51.9% (41株/79株)、29.7% (19株/64株) 和31.7% (26株/82株)。屎肠球菌和粪肠球菌中高水平庆大霉素耐药株分离率分别为43.2% (48株/111株) 和40.9% (27株/66株)。碳青霉烯不敏感肠杆菌科细菌中肺炎克雷伯菌居首位,占57.1% (24株/42株) 。肠杆菌科细菌中分离出30株替加环素不敏感株,其中肺炎克雷伯菌占76.7% (23株/30株);分离出粘菌素耐药肠杆菌科细菌39株,其中大肠埃希菌、阴沟肠杆菌和肺炎克雷伯菌分别占43.6% (17株/39株)、35.9% (14株/39株) 和15.4% (6株/39株)。医院获得性血流感染病原菌主要为革兰阴性杆菌 (以大肠埃希菌和肺炎克雷伯菌为主),其对替加环素、粘菌素和碳青霉烯类药物的敏感率较高;革兰阳性球菌中分离率最高的为金黄色葡萄球菌,其次为屎肠球菌,这两种细菌对替加环素、达托霉素、利奈唑胺、万古霉素和替考拉宁的敏感率较高。粘菌素耐药肠杆菌科细菌、替加环素不敏感肠杆菌科细菌、利奈唑胺或万古霉素不敏感革兰阳性球菌的分离,警示临床高度关注,仍需动态监测耐药进展趋势。 相似文献

Entomopathogenic fungi have great potential to control agricultural and horticultural insect pests, however optimizing conidial production systems to demonstrate high productivity and stability still needs additional efforts for successful field application and industrialization. Although many virulent entomopathogenic fungal isolates have been viewed as potential candidates in a laboratory environment, very few of the isolates are being used in practice for application in agricultural fields as commercial products. I. javanicus is an entomopathogenic fungus that is parasitic to various diverse coleopteran and lepidopteran insects and thought good candidate as biopesticdes. In this work, the basic characteristics of two entomopathogenic fungi, I. javanica FG340 and Pf04, were investigated in morphological examinations, genetic identification, and virulence against Thrips palmi, and then the feasibility of various grains substrates for conidial production was assessed, particularly focusing on conidial productivity and thermotolerance. Isaria javanica FG340 and Pf04 conidia were solid-cultured on 12 grains for 14?days in a Petri dish. Of the tested Italian millet, perilla seed, millet and barley-based cultures showed high conidial production. The four-grain media yielded >1?×?109 conidia/g of I. javanica FG340 and Pf04. Pf04 strain had enhanced thermotolerance up to 45?°C when cultured on Italian millet. In application, it was easy to make a conidial suspension using the cultured grains, and several surfactants were tested to release the conidia. This work suggests several possible inexpensive grain substrates by which to promote conidial production combined with enhanced stability against exposure to high temperature. 相似文献

Ultra Performance Liquid Chromatography-Electrospray ionization-Quadrupole-Time of Flight Mass Spectrometry (UPLC-ESI-Q-TOF–MS) is a powerful lipidomic tool. In this study, we developed a UPLC/Q-TOF–MS based method to investigate the lipid metabolomic changes in different growth phases of Nitzschia closterium f. minutissima. The data classification and biomarker selection were carried out by using multivariate statistical analysis, including principal components analysis (PCA), projection to latent structures with discriminant analysis (PLS-DA), and orthogonal projection to latent structures with discriminant analysis (OPLS-DA). We discovered that the intercellular lipid metabolites were significantly different among exponential, early stationary and late stationary phases. Thirty-one lipid molecules were selected and identified as putative biomarkers, including free fatty acid, Harderoporphyrin, phosphatidylglycerol, 1,2-diacyglycerl-3-O-4′-(N,N-trimethy)-homoserine, triacylglycerol, cholesterol, sulfoquinovosyldiacylglycerol, lyso-sulfoquinovosyldiacylglycerol, monogalactosyldiacylglycerol, digalactosyldiacylglycerol and lyso-digalactosyldiacylglycerol. These lipids have been shown previously to function in energy storage, membrane stability and photosynthesis efficiency during the growth of diatoms. Further analysis on the putative biomarkers demonstrated that nitrate starvation played critical role in the transition from exponential phase to stationary phase in N. closterium. This study is the first one to explore the lipidomic changes of microalgae in different growth phases, which promotes better understanding of their physiology and ecology. 相似文献

利用大肠杆菌研究番茄红素的合成,不仅可以获得副产物少的高产菌株,而且可以探讨基因或基因簇的功能。文中将番茄LeGGPS2和LePSY1的cDNA序列,及欧文氏菌crtI的编码序列分别添加上核糖体结合位点后,以单独或组合的方式受控于T7启动子和终止子,在大肠杆菌菌株BL21(DE3)中进行表达和诱导番茄红素合成。结果显示,仅T7::crtI-LeGGPS2-LePSY1三价基因共表达时才能合成番茄红素,且将种子液以1∶50接种于含3%蔗糖的LB培养基(pH 6.8)中,于37℃摇8 h左右的对数生长后期加IPTG至80μmol/L,30℃诱导表达5 h的发酵条件下,获得2.124 mg/g DCW的番茄红素。该结果既验证了原核化的番茄LeGGPS2和LePSY1基因及与crtI基因协同作用的功能,又为在番茄质体中建立独立的番茄红素合成途径奠定了基础。 相似文献

The aggregation and accumulation of amyloid-β (Aβ) plays a significant role in the pathogenesis of Alzheimer’s disease. Aβ is known to increase free radical production in neuronal cells, leading to oxidative stress and cell death. Diazoxide (DZ), a highly selective drug capable of opening mitochondrial ATP-sensitive potassium channels, has neuroprotective effects against neuronal cell death. However, the mechanism through which DZ protects cholinergic neurons against Aβ-induced oxidative injury is still unclear. The present study was designed to investigate the effects of DZ pretreatment against Aβ1–42 induced oxidative damage and cytotoxicity. Through measures of DZ effects on Aβ1–42 induced cellular damage, reactive oxygen species (ROS) and MDA generation and expressions of gp91phox and p47phox in cholinergic neurons, new insights into the neuroprotective mechanisms can be derived. Aβ1–42 significantly decreased 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide levels and increased ROS and MDA production; all effects were attenuated by pretreatment with DZ or diphenyleneiodonium chloride (a NOX2 inhibitor). Pretreatment with DZ also attenuated the upregulation of NOX2 subunits (gp91phox and p47phox) induced by Aβ1–42. Since NOX2 is one of the main sources of free radicals, these results suggest that DZ can counteract Aβ1–42 induced oxidative stress and associated cell death by reducing the level of ROS and MDA, in part, by alleviating NOX2 expression. 相似文献
