Background
A new subgroup of HIV-1, designated Group P, was recently detected in two unrelated patients of Cameroonian origin. HIV-1 Group P phylogenetically clusters with SIVgor suggesting that it is the result of a cross-species transmission from gorillas. Until today, HIV-1 Group P has only been detected in two patients, and its degree of adaptation to the human host is largely unknown. Previous data have shown that pandemic HIV-1 Group M, but not non-pandemic Group O or rare Group N viruses, efficiently antagonize the human orthologue of the restriction factor tetherin (BST-2, HM1.24, CD317) suggesting that primate lentiviruses may have to gain anti-tetherin activity for efficient spread in the human population. Thus far, three SIV/HIV gene products (vpu, nef and env) are known to have the potential to counteract primate tetherin proteins, often in a species-specific manner. Here, we examined how long Group P may have been circulating in humans and determined its capability to antagonize human tetherin as an indicator of adaptation to humans.Results
Our data suggest that HIV-1 Group P entered the human population between 1845 and 1989. Vpu, Env and Nef proteins from both Group P viruses failed to counteract human or gorilla tetherin to promote efficient release of HIV-1 virions, although both Group P Nef proteins moderately downmodulated gorilla tetherin from the cell surface. Notably, Vpu, Env and Nef alleles from the two HIV-1 P strains were all able to reduce CD4 cell surface expression.Conclusions
Our analyses of the two reported HIV-1 Group P viruses suggest that zoonosis occurred in the last 170 years and further support that pandemic HIV-1 Group M strains are better adapted to humans than non-pandemic or rare Group O, N and P viruses. The inability to antagonize human tetherin may potentially explain the limited spread of HIV-1 Group P in the human population. 相似文献52.

Sexual dimorphism in body size and canine weaponry is commonly associated with high levels of male-male competition. When
group living species do not rely heavily on male-male competition for access to females, sperm competition may represent a
viable alternative strategy. Unlike most haplorhine primates, lemurs are typically monomorphic in body weight and canine height.
We assessed variability of body mass dimorphism and canine size dimorphism in brown lemurs using morphometric data from 3
populations in southeastern Madagascar: Eulemur fulvus rufus, E. albocollaris, and hybrids of the species. We found significant male-biased canine dimorphism in E. albocollaris in conjunction with body-size monomorphism. We observed similar patterns in the hybrids, but E. fulvus rufus exhibited significant female-biased size dimorphism and canine monomorphism. Testes volume was relatively high across study
populations. Thus, sperm competition appears to be strong in brown lemurs. E. albocollaris males combine sperm competition with large canines, but not higher body mass, indicating a difference in sexual strategy

The ppk gene encodes polyphosphate kinase (Ppk), an enzyme that catalyses the polymerization of inorganic phosphate into long chains of polyphosphate (polyP). An insertion mutation in ppk causes a decrease in adaptive mutation in Escherichia coli strain FC40. Adaptive mutation in FC40 mostly results from error-prone DNA polymerase IV (Pol IV), encoded by dinB; most of the antimutagenic phenotype of the ppk mutant disappears in a dinB mutant strain. In addition, the ppk mutant causes a decrease in growth-dependent mutations produced by overexpressing Pol IV. However, the amount of Pol IV protein is unchanged in the ppk mutant strain, indicating that the activity or fidelity of Pol IV is altered. Adaptive mutation is inhibited both by the absence of Ppk, which results in low amounts of polyP, and by overproduction of Ppk, which results in high amounts of polyP, suggesting that an optimal level of polyP is necessary. Taken together, these results suggest a novel mechanism involving polyP that directly or indirectly regulates DNA polymerase activity or fidelity. 相似文献

We tested the hypothesis that the cystic fibrosis transmembrane conductance regulator (CFTR) could be involved in the volume regulation of human red blood cells (RBC). Experiments were based on two gadolinium (Gd(3+)) sensitive mechanisms, i.e. inhibition of ATP release (thetaATP(i)) and membrane destabilization. RBC of either cystic fibrosis (CF) patients or healthy donors (non-CF) were exposed to KCl buffer containing Gd(3+). A significantly larger quantity of non-CF RBC (2.55 %) hemolyzed as compared to CF RBC (0.89 %). It was found that both of the Gd(3+) mechanisms simultaneously are needed to achieve hemolysis, since either overriding thetaATP(i) by exogenous ATP addition prevented Gd(3+) induced hemolysis, or mimicking thetaATP(i) by apyrase in absence of Gd(3+) could not trigger hemolysis. Additionally, ion driven volume uptake was found to be a prerequisite for Gd3+ induced hemolysis as chloride and potassium channel blockers reduced the Gd(3+) response. The results show that in non-CF RBC Gd(3+) exerts its dual effect leading to hemolysis. On the contrary, in CF RBC, lacking CFTR dependent ATP release, the sole Gd(3+) effect of membrane destabilization is not sufficient to induce hemolysis similar to non-CF. After a 20-year absence, severe cyanobacterial blooms have returned to Lake Erie in the last decade, in spite of negligible change in the annual load of total phosphorus (TP). Medium-spectral Resolution Imaging Spectrometer (MERIS) imagery was used to quantify intensity of the cyanobacterial bloom for each year from 2002 to 2011. The blooms peaked in August or later, yet correlate to discharge (Q) and TP loads only for March through June. The influence of the spring TP load appears to have started in the late 1990 s, after Dreissenid mussels colonized the lake, as hindcasts prior to 1998 are inconsistent with the observed blooms. The total spring Q or TP load appears sufficient to predict bloom magnitude, permitting a seasonal forecast prior to the start of the bloom. 相似文献

About a century ago, Conrad Röentgen discovered X-rays, and Henri Becquerel discovered a new phenomenon, which Marie and Pierre Curie later coined as radio-activity. Since their seminal work, we have learned much about the physical properties of radiation and its effects on living matter. Alas, the more we discover, the more we appreciate the complexity of the biological processes that are triggered by radiation exposure and eventually lead (or do not lead) to disease. Equipped with modern biological methods of high-throughput experimentation, imaging, and vastly increased computational prowess, we are now entering an era where we can piece some of the multifold aspects of radiation exposure and its sequelae together, and develop a more systemic understanding of radiogenic effects such as radio-carcinogenesis than has been possible in the past. It is evident from the complexity of even the known processes that such an understanding can only be gained if it is supported by mathematical models. At this point, the construction of comprehensive models is hampered both by technical inadequacies and a paucity of appropriate data. Nonetheless, some initial steps have been taken already and the generally increased interest in systems biology may be expected to speed up future progress. In this context, we discuss in this article examples of relatively small, yet very useful models that elucidate selected aspects of the effects of exposure to ionizing radiation and may shine a light on the path before us. 相似文献

The increasingly widespread use of parametric mathematical models to describe biological systems means that the ability to infer model parameters is of great importance. In this study, we consider parameter inferability in nonlinear ordinary differential equation models that undergo a bifurcation, focusing on a simple but generic biochemical reaction model. We systematically investigate the shape of the likelihood function for the model's parameters, analyzing the changes that occur as the model undergoes a Hopf bifurcation. We demonstrate that there exists an intrinsic link between inference and the parameters’ impact on the modeled system's dynamical stability, which we hope will motivate further research in this area. 相似文献

Blooms of the toxic dinoflagellate, Karenia brevis, have had detrimental impacts on the coastal Gulf of Mexico for decades. Detection of Karenia brevis blooms uses an ecological approach based on anomalies derived from ocean color imagery. The same anomaly product used in Florida produces frequent false positives on the Texas coast. These failures occurred during wind-driven resuspension events. During these events resuspension of benthic algae significantly increases chlorophyll concentrations in the water, resulting in confusion with normal water column phytoplankton, such as Karenia. A method was developed to separate the resuspended chlorophyll from the water column chlorophyll, decreasing the false positives used with the detection method. 相似文献

The new satellite ocean color sensors offer a means of detecting and monitoring algal blooms in the ocean and coastal zone. Beginning with SeaWiFS (Sea Wide Field-of-view Sensor) in September 1997, these sensors provide coverage every 1 to 2 days with 1-km pixel view at nadir. Atmospheric correction algorithms designed for the coastal zone combined with regional chlorophyll algorithms can provide good and reproducible estimates of chlorophyll, providing the means of monitoring various algal blooms. Harmful algal blooms (HABs) caused by Karenia brevis in the Gulf of Mexico are particularly amenable to remote observation. The Gulf of Mexico has relatively clear water and K. brevis, in bloom conditions, tends to produce a major portion of the phytoplankton biomass. A monitoring program has begun in the Gulf of Mexico that integrates field data from state monitoring programs with satellite imagery, providing an improved capability for the monitoring of K. brevis blooms. 相似文献
