Suppression subtractive hybridisation was used to isolate heavy metal-induced genes from Pisum sativum roots hydroponically exposed to 5 microM HgCl2 and 10 microM EDTA. Six genes were induced out of which one, PsHMIP6B, was novel. The other genes (PsSAMT, PsI2'H, PsNDA, PsAPSR, PsPOD) had not previously been isolated from pea and sequenced. All six genes were also induced after exposure to 5 microM HgCl2 in the absence of EDTA. The induction pattern was in some cases different for the two Hg species, demonstrating a quicker response to-free Hg2+ than Hg-EDTA. The stress-specificity of the gene regulation was investigated by hydroponically adding 5 microM Cd2+. Most Hg-induced cDNAs were also induced by Cd2+ but to a smaller extent than after Hg exposure. In addition, the gene expression was also probed for tissue specificity, which showed that all six genes were expressed in roots and not in leaves. 相似文献

Phagocytosis of microbial pathogens is essential for the host immune response to infection. Our previous work has shown that lipooligosaccharide (LOS) expression on the surface of Neisseria meningitidis (Nm) is essential for phagocytosis, but the receptor involved remained unclear. In this study, we show that human CR3 (CD11b/CD18) and CR4 (CD11c/CD18) are phagocytic receptors for Nm as illustrated by the capacity of CR3- and CR4-transfected Chinese hamster ovary (CHO) cells to facilitate Nm uptake. A CR3-signalling mutant failed to internalize Nm, showing that the ability of CR3 to signal is essential for phagocytosis. Internalization of Nm by CR3-transfected CHO cells could be inhibited by the presence of CR3-specific antibodies. Furthermore, dendritic cells from leukocyte adhesion deficiency-1 patients, who have diminished expression of β2 integrins, showed markedly reduced phagocytosis of Nm. The CR3-mediated phagocytosis required the presence of lipopolysaccharide-binding protein (LBP). Furthermore, the expression of LOS by Nm was essential for LBP binding and phagocytosis via CR3. These results reveal a critical role of CR3 and LBP in the phagocytosis of Nm and provide important insights into the initial interaction meningococci have with the immune system. 相似文献

The effect of the electrical potential on the H+-ATPase of Rhodospirillum rubrum is examined. It is shown that the forward reaction rate (ATP synthesis) is increased by a factor of 10 during illumination while the reversed rate is only slightly decreased. This indicates that the electrical potential across the membrane affects the rate constants mainly by increasing the forward rate constants rather than decreasing the reversed rate constants in order to go from net hydrolysis to net synthesis. 相似文献

PMS-dependent photophosphorylation in bundle sheath chloroplasts isolated from Zea mays was monitored by using a continuous method. Carbonyl cyanide p-trifluoromethoxyphenylhydrazone (FCCP) and venturicidin were shown to inhibit the ATP-synthesis. Venturicidin has been shown to inhibit ATP-formation in both mesophyll and bundle sheath chloroplasts. In contrast to the case in mesophyll chloroplasts, FMN was not able to promote photophosphorylation in bundle sheath chloroplasts. The effects of other cofactors and inhibitors on the ATP-synthesis in bundle sheath chloroplasts are shown. No photoinduced synthesis of inorganic pyrophosphate was seen, neither in bundle sheath chloroplasts, nor in mesophyll chloroplasts. 相似文献

Manifestations of aluminium (Al) toxicity in two cultivars of wheat ( Triticum aestivum L. cvs Kadett [relatively Al-resistant] and WW 20299 [relatively Al-sensitive]) were investigated at two root zone temperatures (RZT) that may occur in the field. The plants were grown for 9 days at 10 or 25°C RZT. Mineral nutrients other than CaSO4 were supplied daily in exponentially increasing amounts to meet the demand of the plants. Al was added as Al2 (SO4 )3 at the beginning of the culture period at concentrations ranging from 0 to 100 μ M . pH was kept constant at 4.1. Experimental data were analysed for interactions between Al and RZT on a fresh weight basis by the nonlinear Weibull function. Cultivar Kadett, when grown at 25°C RZT, was more resistant to Al than when grown at 10°C RZT. Cultivar WW 20299 was equally sensitive to Al at 10 and 25°C RZT but generally more sensitive to Al than cv. Kadett. It is suggested that cv. Kadett, in contrast to cv. WW 20299, possesses a mechanism for Al resistance that is less effective at 10°C than at 25°C RZT and therefore may be metabolically dependent. In roots, the concentrations of K, P, Mg and Ca were not negatively affected by Al or by RZT. In shoots of both cultivars the concentrations of Ca and Mg became comparatively low when the plants were treated with Al or at low RZT, the effect being larger for Ca than for Mg. At 10°C RZT under Al stress, the Ca concentrations in shoots approached the critical concentration where growth may be inhibited. As no Al was detected in the shoots, it is suggested that Al in the roots inhibits shoot growth by reducing transport of Ca from roots to shoots. 相似文献
