The synthesis of 1-O-β-lactosyl-(R,S)-glycerols was achieved by three methods: (a) in 25% yield by the trimethylsilyl trifluoromethanesulfonate-promoted reaction of octa-O-acetyl-β-lactose (11) with 0.5 mol-equiv. of 2-O-benzylglycerol (4), (b) in 34% yield by the coupling of 4 with an equimolar amount of hepta-O-acetyl--lactosyl bromide (12) in the presence of mercury(II) cyanide, and (c) in 50% yield by the coupling of equimolar amounts of 12 and 1,2-di-O-benzyl-(R,S)-glycerols in the presence of mercury(II) cyanide, followed in each case by the removal of the blocking groups. 1,3-Di-O-β-lactosylglycerols were prepared in 21% yield by the coupling of 11 with 0.5 mol-equivalent of 4 by method (a), and in 38% yield by the coupling of 12 with 0.5 mol-equiv. of 4 by method (b), followed by the removal of the blocking groups. The configuration of the glycosidic linkage between the lactose units and the glycerol residue was established by high-resolution, two-dimensional 1H-n.m.r. spectroscopy. 相似文献

The first crystal structure of mannose 1-phosphate is described. The dipotassium hydrate salt crystallizes in the P2(1)2(1)2 space group. There are two independent dianions (I and II) in the asymmetric unit, which are alpha anomers adopting the 4C(1) chair conformation. The main difference between the two mannose 1-phosphate dianions is the orientation of the phosphate group with relation to the pyranosyl ring. In I, one of the phosphate oxygen atoms is antiperiplanar positions with respect to carbon atom C-1, whereas the two others are situated synclinally. The corresponding orientations of the terminal phosphate oxygen atoms in II are synperiplanar and anticlinal. The potassium cations are six- and seven-coordinate, mainly with O atoms of hydroxyl groups and water molecules. There are potassium channels extending along the c-axis. In the packing arrangement, water molecules and mannose phosphate groups also define two different types of layers parallel to a-axis. Within water channels there are extensive hydrogen-bonding networks. 相似文献

Halogen-substituted iminosulfuranes are transdermal penetration enhancers (TPEs) in permeation studies using hairless mouse or human cadaver skin. The interaction of N--(4--R-benzoyl)-S,S-dimethyliminosulfuranes 1--4, where R=H, Cl, Br, and I, with l-alpha-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) has been studied using differential scanning calorimetry, isothermal titration calorimetry, nuclear Overhauser effect spectroscopy (NOESY), and NMR spectroscopy, and by calculation of the iminosulfurane polarizabilities in order to elucidate the molecular basis of the TPE activity. The active compounds reduce the melting temperature of the gel-to-liquid-crystal phase transition and induce multiple components in the transition excess heat capacity profile. The partitioning of the bromo derivative 3, the most active compound, into DMPC is unique in that 3 may be trapped in the bilayer, affording an enhanced residence time and a reason for its high TPE activity. The entropy decrease associated with the transfer of 3 to the bilayer is much lower than that for the other compounds, indicating that 3 occupies or induces sites that afford it considerable local motional freedom. Correlations between the iminosulfurane TPE activities, the partition coefficients, and NOESY crosspeak volume were observed. Molecular polarizabilities are not consistent with a TPE mode of action involving interaction of these agents with protein side chains. 相似文献

The effect of acidification on ecosystems is reviewed. About 30% of the world land area is acidified mostly by natural processes. Only a small part of it is acidified from pollution which leads to extensive forest and surface water deterioration in Europe and North America. The damage is especially visible in areas of poor soil quality. The acid deposition pattern is characterized, with a special emphasis on Poland. It was shown that strategies for mitigation of ecosystem acidification lead to a decrease of SO2 emissions due to the use of lower sulphur content fuels and application of desulphurisation methods. Also, a decrease in NOx emissions is observed due to use of catalysts and application of NOx removal technologies. However, since about 50% of NOx emission is from vehicles, this improvement is offset by a continuously increasing vehicle population. The application of liming, forest fertilisation and suitable land use for mitigation of ecosystem acidification is also reviewed. 相似文献

This article presents a comprehensive review of large and highly diverse superfamily of nucleotidyltransferase fold proteins by providing a global picture about their evolutionary history, sequence-structure diversity and fulfilled functional roles. Using top-of-the-line homology detection method combined with transitive searches and fold recognition, we revised the realm of these superfamily in numerous databases of catalogued protein families and structures, and identified 10 new families of nucleotidyltransferase fold. These families include hundreds of previously uncharacterized and various poorly annotated proteins such as Fukutin/LICD, NFAT, FAM46, Mab-21 and NRAP. Some of these proteins seem to play novel important roles, not observed before for this superfamily, such as regulation of gene expression or choline incorporation into cell membrane. Importantly, within newly detected families we identified 25 novel superfamily members in human genome. Among these newly assigned members are proteins known to be involved in congenital muscular dystrophy, neurological diseases and retinal pigmentosa what sheds some new light on the molecular background of these genetic disorders. Twelve of new human nucleotidyltransferase fold proteins belong to Mab-21 family known to be involved in organogenesis and development. The determination of specific biological functions of these newly detected proteins remains a challenging task. 相似文献

The heterogeneous nuclear ribonucleoprotein K is an ancient RNA/DNA-binding protein that is involved in multiple processes that compose gene expression. The pleiotropic action of K protein reflects its ability to interact with different classes of factors, interactions that are regulated by extracellular signals. We used affinity purification and MS to better define the repertoire of K protein partners. We identified a large number of new K protein partners, some typically found in subcellular compartments, such as plasma membrane, where K protein has not previously been seen. Electron microscopy showed K protein in the nucleus, cytoplasm, mitochondria, and in vicinity of plasma membrane. These observations greatly expanded the view of the landscape of K protein-protein interaction and provide new opportunities to explore signal transduction and gene expression in several subcellular compartments. 相似文献

9-Nickelafluorenyllithium (1) reacted with iodine and then with various CpxM complexes (Cpx = Cp, Cp∗, M = Na, Mg, Li) in different solvents. The products (π-allyl complexes), isolated from these reactions, were formed via addition of the biphenyl ligand to the Cp ring originally bonded to the Ni atom. Apigenin (5,7,4′-trihydroxyflavone) is a cancer chemopreventive agent and a member of the family of plant flavonoids. Apigenin interaction with liposomes formed with dipalmitoylphosphatidylcholine (DPPC) was investigated by means of FTIR spectroscopy, 1H NMR and EPR techniques. Fluorescent microscopy and electron microscopy were applied to study the apigenin effects on colon myofibroblasts and human skin fibroblasts. The strong rigidifying effect of apigenin with respect to polar head groups was concluded on the basis of the action of the flavone on partition coefficient of Tempo spin label between the water and lipid phases. The ordering effect was also found in hydrophobic region at the depth monitored by 5-SASL and 16-SASL spin labels. The inclusion of apigenin to the membrane restricted the motional freedom of polar head groups lowering penetration of Pr3 + ions to the membranes. The 1H NMR technique supported also the restriction of motional freedom of the membrane in the hydrophobic region, especially in the zone of CH2 groups of alkyl chains. FTIR analysis showed that apigenin incorporates into DPPC liposomes via hydrogen bonding between its own hydroxyl groups and lipid polar head groups in the COPOC segment. It is also very likely that hydroxyl groups of apigenin link with polar groups of DPPC by water bridges. Electron and fluorescence microscopic observations revealed changes in the internal membrane organization of the examined cells. In conclusion, the changes of the structural and dynamic properties of membranes can be crucial for processes involving tumor suppression signal transduction pathways and cell cycle regulation. 相似文献

The aim of present study was to determine the changes of some biochemical and physiological processes, which occurred in seeds and seedlings of white lupine and faba bean after pre-sowing treatment with laser beams. It was found this treatment of seeds considerably increased the activity of amylolytic enzymes in seeds of both plants. The greatest differentiation of the enzymatic activity was noticed after 120?h from the time of sowing but the activity of these enzymes in the seeds of both tested plants was similar and it had the same course in time. The irradiated seeds of white lupine and faba bean had higher fresh weight at the time of imbibition than the seeds which were not treated with laser beams. It resulted in earlier and more uniform germination. The concentration of free radicals increased considerably in the seeds pre-treated with laser beams and the largest increase in seeds of both plant species was noticed after five exposures to laser beams. Treating seeds with laser beams considerably increased the amount of indole-3-acetic acid (IAA) in the germinating seeds. Three exposures of seeds caused the largest increase of this plant hormone content in the seeds. The activity of IAA in faba bean was slightly higher than in white lupine seeds. Pre-sowing stimulation with laser had a positive influence on the growth and development of seedlings, which had longer hypocotyl and roots in comparison to seedlings which grew from non irradiated seeds. 相似文献
