Lujo virus (LUJV), a new member of the family Arenaviridae and the first hemorrhagic fever–associated arenavirus from the Old World discovered in three decades, was isolated in South Africa during an outbreak of human disease characterized by nosocomial transmission and an unprecedented high case fatality rate of 80% (4/5 cases). Unbiased pyrosequencing of RNA extracts from serum and tissues of outbreak victims enabled identification and detailed phylogenetic characterization within 72 hours of sample receipt. Full genome analyses of LUJV showed it to be unique and branching off the ancestral node of the Old World arenaviruses. The virus G1 glycoprotein sequence was highly diverse and almost equidistant from that of other Old World and New World arenaviruses, consistent with a potential distinctive receptor tropism. LUJV is a novel, genetically distinct, highly pathogenic arenavirus.  相似文献

The objective of this study was to determine the effects of plant growth regulator (PGR) (no PGR, trinexapac-ethyl, and paclobutrazol) and N fertilizer (zero N, an average of 37 kg N ha?1 month?1, 6 and 12 kg N ha?1 week?1) on soil organic C (SOC) and soil N in creeping bentgrass (Agrostis stolonifera L.) fairway turf. After 4 years of field experiments soil samples were obtained from soil depths of 0–2.5, 2.5–5, 5–7.5, 7.5–10, 10–15, 15–20, and 20–30 cm. Soil bulk density, SOC, total N, NO 3 ? –N, and NH 4 + –N concentrations were determined. Paclobutrazol and trinexapac-ethyl application increased SOC. The 37 kg N ha?1 month?1 application increased SOC at the 0–2.5 cm depth with both PGRs. When paclobutrazol was used, N fertilizer always increased SOC; however, the greatest increase was observed with the 12 kg N ha?1 week?1 application when compared to other rates, inversely related to the NH 4 + –N concentration. Nitrogen application increased soil total N and NO 3 ? –N in the upper three depths. The application of PGRs and N fertilizer to creeping bentgrass fairway turf is an effective strategy for promoting C sequestration.  相似文献

Background  

Association mapping using abundant single nucleotide polymorphisms is a powerful tool for identifying disease susceptibility genes for complex traits and exploring possible genetic diversity. Genotyping large numbers of SNPs individually is performed routinely but is cost prohibitive for large-scale genetic studies. DNA pooling is a reliable and cost-saving alternative genotyping method. However, no software has been developed for complete pooled-DNA analyses, including data standardization, allele frequency estimation, and single/multipoint DNA pooling association tests. This motivated the development of the software, 'PDA' (Pooled DNA Analyzer), to analyze pooled DNA data.  相似文献

Dormant tumor cells resistant to ablative cancer therapy represent a significant clinical obstacle due to later relapse. Experimentally, the murine B cell lymphoma (BCL1) is used as a model of tumor dormancy in mice vaccinated with the BCL1 Ig. Here, we used this model to explore the cellular mechanisms underlying dormancy. Our previous studies have demonstrated that T cell-mediated immunity is an important component in the regulation of tumor dormancy because Id-immune T cells adoptively transferred into passively immunized SCID mice challenged with BCL1 cells significantly increased the incidence and duration of the dormant state. We have extended these observations and demonstrate that CD8+, but not CD4+, T cells are required for the maintenance of dormancy in BCL1 Ig-immunized BALB/c mice. In parallel studies, the transfer of Id-immune CD8+ cells, but not Id-immune CD4+ cells, conferred significant protection to SCID mice passively immunized with nonprotective levels of polyclonal anti-Id and then challenged with BCL1 cells. Furthermore, the ability of CD8+ T cells to induce a state of dormancy in passively immunized SCID mice was completely abrogated by treatment with neutralizing alpha-IFN-gamma mAbs in vivo. In vitro studies demonstrated that IFN-gamma alone or in combination with reagents to cross-link the surface Ig induced both cell cycle arrest and apoptosis in a BCL1 cell line. Collectively, these data demonstrate a role for CD8+ T cells via endogenous production of IFN-gamma in collaboration with humoral immunity to both induce and maintain a state of tumor dormancy.  相似文献

Controversy still exists regarding the biological function of granzyme serine proteases released with perforin from the cytotoxic granules of NK cells and CTLs. In particular, it is not clear whether the major granzymes, A and B, play an essential role in tumor rejection mediated by the perforin pathway. We have now examined the relative importance of perforin and granzyme A and B clusters in five different tumor models that stringently distinguish their importance. We conclude that granzyme A and B clusters are not essential for CTL- and NK cell-mediated rejection of spontaneous and experimental tumors, raising the likelihood that either perforin alone or in combination with an additional granzyme or granule component(s) mediates cytotoxicity of tumor cells in vivo.  相似文献

Uhl and Daniel reported in this journal in 1999 (Extremophiles 3:263-267) the characterization of the first archaeal hemicellulase with a unique N-terminal sequence showing no homology with any xylanase or other protein from the databases. A genomic library of the chromosomal DNA of Thermococcus zilligii strain AN1 was screened by using a degenerate probe deduced from the N-terminal sequence. A positive clone was identified, and an amino acid sequence analysis revealed that the N-terminal sequence from this protein and the N-terminal sequence from the putative xylanase of T. zilligii were identical. However, the comparison of the amino acid sequence of the protein with sequences in the main protein databases revealed significant similarities with maltodextrin phosphorylases. In conclusion, it is likely that the N-terminal sequence proposed by Uhl and Daniel is not that of the T. zilligii xylanase, but corresponds to an archaeal T. zilligii maltodextrin phosphorylase.  相似文献

The alpha3 fucosyltransferase, FucT-VII, is one of the key glycosyltransferases involved in the biosynthesis of the sialyl Lewis X (sLex) antigen on human leukocytes. The sialyl Lewis X antigen (NeuAcalpha(2-3)Galbeta(1-4)[Fucalpha(1-3)]GlcNAc-R) is an essential component of the recruitment of leukocytes to sites of inflammation, mediating the primary interaction between circulating leukocytes and activated endothelium. In order to characterize the enzymatic properties of the leukocyte alpha3 fucosyltransferase FucT-VII, the enzyme has been expressed in Trichoplusia ni insect cells. The enzyme is capable of synthesizing both sLexand sialyl-dimeric-Lexstructures in vitro , from 3'-sialyl-lacNAc and VIM-2 structures, respectively, with only low levels of fucose transfer observed to neutral or 3'-sulfated acceptors. Studies using fucosylated NeuAcalpha(2-3)-(Galbeta(1- 4)GlcNAc)3-Me acceptors demonstrate that FucT-VII is able to synthesize both di-fucosylated and tri-fucosylated structures from mono- fucosylated precursors, but preferentially fucosylates the distal GlcNAc within a polylactosamine chain. Furthermore, the rate of fucosylation of the internal GlcNAc residues is reduced once fucose has been added to the distal GlcNAc. These results indicate that FucT-VII is capable of generating complex selectin ligands, in vitro , however the order of fucose addition to the lactosamine chain affects the rate of selectin ligand synthesis.   相似文献

Sunopsis Monoamine oxidase (MAO) activity has been demonstrated histochemically in rat hypothalamic ependyma using the sulphate-tetrazolium and coupled peroxidatic techniques with tryptamine, tyramine, 5-hydroxytryptamine and benzylamine as substrates. Both methods were applied to cryostat sections with and without exposure to selective amine oxidase inhibitors, including the selective A-MAO inhibitor clorgyline, and the B-MAO inhibitor deprenyl. Our results show that both cuboidal-columnar and tanycyte ependyma contain one or more forms of MAO not generally present in the hypothalamus. It is suggested that ependymal MAO may form an amine-barrier system modulating the movement and effect within the hypothalamus of specific cerebrospinal fluid or blood monoamines.  相似文献

Estimation of heterozygosity for single-probe multilocus DNA fingerprints   总被引:8,自引:0,他引:8  
In spite of the increasing application of DNA fingerprinting to natural populations and to the genetic identification of humans, explicit methods for estimation of basic population genetic parameters from DNA fingerprinting data have not been developed. Contributing to this omission is the inability to determine, for multilocus fingerprinting probes, relatively important genetic information, such as the number of loci, the number of alleles, and the distribution of these alleles into specific loci. One of the most useful genetic parameters that could be derived from such data would be the average heterozygosity, which has traditionally been employed to measure the level of genetic variation within populations and to compare genetic variation among different loci. We derive here explicit formulas for both the estimation of average heterozygosity at multiple hypervariable loci and a maximum value for this estimate. These estimates are based upon the DNA restriction-pattern matrices that are typical for fingerprinting studies of humans and natural populations. For several empirical data sets from our laboratory, estimates of average and maximal heterozygosity are shown to be relatively close to each other. Furthermore, variances of these statistics based on simulation studies are relatively small. These observations, as well as consideration of the effect of missing alleles and alternate numbers of loci, suggest that the average heterozygosity can be accurately estimated using phenotypic DNA fingerprint patterns, because this parameter is relatively insensitive to the lack of certain genetic information.   相似文献
