We report a series of potent and selective MC4R agonists based on spiroindane amide privileged structures for potential treatments of obesity. Among the synthetic methods used, Method C allows rapid synthesis of the analogs. The series of compounds can afford high potency on MC4R as well as good rodent pharmacokinetic profiles. Compound 1r (MK-0489) demonstrates MC4R mediated reduction of food intake and body weight in mouse models. Compound 1r is efficacious in 14-day diet-induced obese (DIO) rat models.  相似文献

Reverse cholesterol transport promoted by HDL-apoA-I is an important mechanism of protection against atherosclerosis. We have previously identified apoA-I mimetic peptides by synthesizing analogs of the 22 amino acid apoA-I consensus sequence (apoA-I(cons)) containing non-natural aliphatic amino acids. Here we examined the effect of different aliphatic non-natural amino acids on the structure-activity relationship (SAR) of apoA-I mimetic peptides. These novel apoA-I mimetics, with long hydrocarbon chain (C(5-8)) amino acids incorporated in the amphipathic α helix of the apoA-I(cons), have the following properties: (i) they stimulate in vitro cholesterol efflux from macrophages via ABCA1; (ii) they associate with HDL and cause formation of pre-β HDL particles when incubated with human and mouse plasma; (iii) they associate with HDL and induce pre-β HDL formation in vivo, with a corresponding increase in ABCA1-dependent cholesterol efflux capacity ex vivo; (iv) at high dose they associate with VLDL and induce hypertriglyceridemia in mice. These results suggest our peptide design confers activities that are potentially anti-atherogenic. However a dosing regimen which maximizes their therapeutic properties while minimizing adverse effects needs to be established.  相似文献

Aims

Banksia attenuata is a resprouting species growing in deep sand, while B. sessilis is a fire-killed species occurring in shallow sand over laterite or limestone. We aimed to discover the ecophysiological basis for their different distributions by exploring their investment in deep non-cluster roots and shallow cluster roots, and their cluster-root functioning.

Methods

Deep-pot (1 m), shallow-pot (400 mm), hydroponic experiments and phosphorus (P)-extraction experiment were carried out. Biomass allocation, cluster-root exudation, plant P and leaf manganese (Mn) concentrations were measured.

Results

Banksia attenuata allocated more biomass to deep roots and less biomass to cluster roots than B. sessilis did in deep pots. The two Banksias released similar carboxylates in all experiments, with similar carboxylate-exudation rates in hydroponics. The carboxylate amount per unit cluster root of B. sessilis grown in shallow pots was greater than that of B. attenuata, and B, sessilis acquired more P than B. attenuata did in limestone substrate.

Conclusions

Greater investment in deep roots for water uptake accounts for the presence of B. attenuata in deep sand, and vice versa for the absence of B. sessilis. A larger investment in cluster roots, which released greater amounts of carboxylates, likely accounts for B. sessilis occurring over limestone. Trade-offs in investment and cluster-root functioning support the species’ distribution patterns and life histories. Leaf Mn concentration was a good proxy for the plant capacity to acquire P.

Background

The unconventional motor protein, myosin Va, is crucial for the development of the mouse neuromuscular junction (NMJ) in the early postnatal phase. Furthermore, the cooperative action of protein kinase A (PKA) and myosin Va is essential to maintain the adult NMJ. We here assessed the involvement of myosin Va and PKA in NMJ recovery during muscle regeneration.

Methodology/Principal Findings

To address a putative role of myosin Va and PKA in the process of muscle regeneration, we used two experimental models the dystrophic mdx mouse and Notexin-induced muscle degeneration/regeneration. We found that in both systems myosin Va and PKA type I accumulate beneath the NMJs in a fiber maturation-dependent manner. Morphologically intact NMJs were found to express stable nicotinic acetylcholine receptors and to accumulate myosin Va and PKA type I in the subsynaptic region. Subsynaptic cAMP signaling was strongly altered in dystrophic muscle, particularly in fibers with severely subverted NMJ morphology.

Conclusions/Significance

Our data show a correlation between the subsynaptic accumulation of myosin Va and PKA type I on the one hand and NMJ regeneration status and morphology, AChR stability and specificity of subsynaptic cAMP handling on the other hand. This suggests an important role of myosin Va and PKA type I for the maturation of NMJs in regenerating muscle.  相似文献
