The rhythm generated by the peribladder area was found to be slower than that generated by the basis perirenal pacemaker in cats and rats. The antiperistaltic waves' frequency of the peribladder area was the lowest within the range of all the recorded rhythms.  相似文献

Extracts of pig kidneys or germinated soya beans after preliminary steps were affinity chromatographied on Sepharose containing cyanogen bromide immobilized monoclonal antibodies to pig insulin. The material bound to the affinity column was separated by HPLC resulting in one homogeneous protein from each source. Both proteins have been shown to inhibit DNA synthesis in cultured embryonic human fibroblasts and VERO fibroblasts. The effect of pig kidney protein was potentiated by insulin. Soya and pig proteins were characterized by the following parameters: molecular weights of 8.5 and 10.3 kD, apparent constants of dissociation with rat liver plasma membranes of 4.7 x 10(-8) M and 9.8 X 10(-8) M, respectively. The soya proteins competed for the binding sites on plasma membranes with insulin whereas the pig protein did not. The N-terminal amino acid sequences of 20 residues were determined for both proteins. Comparison of these sequences with known protein sequences was performed. A 30-40% primary structure homology of the studied fragment of soya bean protein with the fragments of some oncogenic viruses proteins and transforming proteins was revealed.  相似文献

We tried to answer the question to what extent neurons in the tectum of the superior colliculus (SC) of the cat and in regions of the mesencephalic reticular formation (MRF) localized more ventrally are involved in the control of movements of the limbs. The impulse activity of neurons of the above-mentioned structures was recorded in animals performing targeted operated food-procuring movements by their forelimbs (pressing the pedal). As was found, neurons with impulse reactions correlating with forelimb movements are rather numerous in the SC and adjacent MRF, and there are several groups of neurons, whose impulse responses reached their maxima within different phases of the movements. These were neurons with peaks of the discharge frequency coinciding with the target-reaching movement, with the moment of touching the pedal, with pressing the pedal, and with the development of the muscle force counteracting the forced withdrawal of the limb toward the initial position. Such specific patterns of the responses of different neurons of the SC and neighboring MRF are indicative of a rather specific involvement of the above structures in the control of forelimb movements in cats. Poly(acrylonitrile)chitosan composite membranes for urease immobilization   总被引:1,自引:0,他引:1  
(Poly)acrylonitrile/chitosan (PANCHI) composite membranes were prepared. The chitosan layer was deposited on the surface as well as on the pore walls of the base membrane. This resulted in the reduction of the pore size of the membrane and in an increase of their hydrophilicity. The pore structure of PAN and PANCHI membranes were determined by TEM and SEM analyses. It was found that the average size of the pore under a selective layer base PAN membrane is 7 microm, while the membrane coated with 0.25% chitosan shows a reduced pore size--small or equal to 5 microm and with 0.35% chitosan--about 4 microm. The amounts of the functional groups, the degree of hydrophilicity and transport characteristics of PAN/Chitosan composite membranes were determined. Urease was covalently immobilized onto all kinds of PAN/chitosan composite membranes using glutaraldehyde. Both the amount of bound protein and relative activity of immobilized urease were measured. The highest activity (94%) was measured for urease bound to PANCHI2 membranes (0.25% chitosan). The basic characteristics (pH(opt), pH(stability), T(opt), T(stability), heat inactivation and storage stability) of immobilized urease were determined. The obtained results show that the poly(acrylonitrile)chitosan composite membranes are suitable for enzyme immobilization.  相似文献

Two strains (KM3 and KM5) of halophilic methylobacteria isolated from Red Sea algae do not require vitamin B12 for growth and can use methanol, methylamine, dimethylamine, trimethylamine, dimethyl sulfide, and fructose as sources of carbon and energy. The cells of these strains are gram-negative motile monotrichous (strain KM3) or peritrichous (strain KM5) rods. The strains are strictly aerobic and require Na+ ions but not growth factors. They are oxidase-and catalase-positive and reduce nitrates to nitrites. Both strains can grow in a temperature range of 4 to 37°C (with optimal growth at 29–34°C), at pH between 5.5 and 8.5 (with optimal growth at pH 7.5–8.0), and in a range of salt concentrations between 0.5 and 15% NaCl (with optimal growth at 5–9% NaCl). The phospholipids of these strains are dominated by phosphatidylethanolamine and phosphatidylglycerol and also include phosphatidylcholine, phosphatidylserine, and cardiolipin. The dominant fatty acids are C16:1ω7c and C16:0. The major ubiquinone is Q8. The cells accumulate ectoin, glutamate, and sucrose as intracellular osmoprotectants. The strains implement the 2-keto-3-deoxy-6-phosphogluconate-dependent variant of the ribulose monophosphate pathway. The G+C content of the DNA is 44.4–44.7 mol%. Analysis of the 16S rRNA genes showed that both strains belong to Gammaproteobacteria and have a high degree of homology (99.4%) to Methylophaga marina ATCC 35842T. Based on the data of polyphasic taxonomy, isolates KM3 and KM5 are identified as new strains M. marina KM3 (VKM B-2386) and M. marina KM5 (VKM B-2387). The ability of these strains to produce auxins (indole-3-acetic acid) suggests their metabolic association with marine algae.  相似文献

Microbiological technology for the enhancement of oil recovery based on the activation of the stratal microflora was tested in the high-temperature horizons of the Kongdian bed (60 degrees C) of the Dagang oil field (China). This biotechnology consists in the pumping of a water-air mixture and nitrogen and phosphorus mineral salts into the oil stratum through injection wells in order to stimulate the activity of the stratal microflora which produce oil-releasing metabolites. Monitoring of the physicochemical, microbiological, and production characteristics of the test site has revealed large changes in the ecosystem as a result of the application of biotechnology. The cell numbers of thermophilic hydrocarbon-oxidizing, fermentative, sulfate-reducing, and methanogenic microorganisms increased 10-10 000-fold. The rates of methanogenesis and sulfate reduction increased in the near-bottom zone of the injection wells and of some production wells. The microbial oil transformation was accompanied by the accumulation of bicarbonate ions, volatile fatty acids, and biosurfactants in the formation waters, as well as of CH4 and CO2 both in the gas phase and in the oil. Microbial metabolites promoted the additional recovery of oil. As a result of the application of biotechnology, the water content in the production liquid from the test site decreased, and the oil content increased. This allowed the recovery of more than 14000 tons of additional oil over 3.5 years.  相似文献

The ultrastructure of neurons, synapses and astrocytes of hippocampal CA1 area in rats was investigated 14 days after: systemic injection of kainic acid and kainic acid and myo-Inositol. After injection of kainic acid numerous neurons with superficial and deep ultrastructural changes of cytoplasmic organelles were described. Among synapses numerous forms with osmiophilic active zone and single synaptic vesicles, also presynaptic terminals with core vesicles were often seen. After kainic acid + myo-Inositol injection the cells with superficial changes of organelles dominated and the synapsoarchitectonics of the area was close to normal. Thus, electrono-microscopic data indicate possible neuroprotective (antiepileptic?) features of myo-Inositol.  相似文献

The haloalkaliphilic methylotrophic bacterium (strain Kr3) isolated from material scraped off the deteriorating marble of the Moscow Kremlin masonry has been found to be able to utilize methanol, methylamine, trimethylamine, and fructose as carbon and energy sources. Its cells are gram-negative motile rods multiplying by binary fission. Spores are not produced. The isolate is strictly aerobic and requires vitamin B12 and Na+ ions for growth. It is oxidase- and catalase-positive and reduces nitrates to nitrites. Growth occurs at temperatures between 0 and 42 degrees C (with the optimum temperatures being 20-32 degrees C), pH values between 6 and 11 (with the optimum at 8-9), and NaCl concentrations between 0.05 and 3 M (with the optimum at 0.5-1.5 M). The dominant cellular phospholipids are phosphatidylethanolamine, phosphatidylglycerol, and cardiolipin. The major cellular fatty acids are palmitic (C16:0), palmitoleic (C16:1), and octadecenoic (C18:1) acids. The major ubiquinone is Q8. The isolate accumulates ectoine and glutamate, as well as a certain amount of sucrose, to function as osmoprotectants and synthesizes an exopolysaccharide composed of carbohydrate and protein components. It is resistant to heating at 70 degrees C, freezing, and drying; utilizes methanol, with the resulting production of formic acid, which is responsible for the marble-degrading activity of the isolate; and implements the 2-keto-3-deoxy-6-phosphogluconate variant of the ribulose monophosphate pathway. The G+C content of its DNA is 44.6 mol%. Based on 16S rRNA gene sequencing and DNA-DNA homology levels (23-41%) with neutrophilic and alkaliphilic methylobacteria from the genus Methylophaga, the isolate has been identified as a new species, Methylophaga murata (VKM B-2303T = NCIMB 13993T).  相似文献

Changes in the kinematic and electromyographic characteristics that occur while learning to move as fast as possible have been studied experimentally. Experimental investigation of what happens to the individual motor units (MUs) is more difficult. Access to each MU is impossible, and the recruitment and force developing properties of all individual MUs cannot be known. Thus, what is currently known about MU firing is based on experiments that have recorded relatively few MUs compared to what exists in the entire muscle. A recently developed muscle model (Raikova and Aladjov, 2002, J. Biomechanics, 35, 1123-1135) composed of MUs with different properties can be used for such investigation. The process of learning fast elbow flexion in the horizontal plane was simulated and the results were compared with experimentally measured data. Comparing the simulation results of the very first trial of a particular subject with those of the last trail (at the end of the learning process), it can be concluded that the speed of limb motion and muscle forces increase initially as a result of the more synchronous MUs activation and the increase of firing rate of active MUs. Further improvement necessitated an appreciable reduction in the motor task requirements (i.e. less muscle force and less MUs' activity) set in the computational algorithm by optimization criteria. This forced the next process-inclusion of additional MUs.  相似文献
