PHA production, from bacteria to plants.   总被引:5,自引:0,他引:5  
The genes encoding the polyhydroxyalkanoate (PHA) biosynthetic pathway in Ralstonia eutropha (3-ketothiolase, phaA or bktB; acetoacetyl-CoA reductase, phaB; and PHA synthase, phaC) were engineered for plant plastid targeting and expressed using leaf (e35S) or seed-specific (7s or lesquerella hydroxylase) promoters in Arabidopsis and Brassica. PHA yields in homozygous transformants were 12-13% of the dry mass in homozygous Arabidopsis plants and approximately 7% of the seed weight in seeds from heterozygous canola plants. When a threonine deaminase was expressed in addition to bktB, phaB and phaC, a copolyester of 3-hydroxybutyrate and 3-hydroxyvalerate was produced in both Arabidopsis and Brassica.  相似文献

The shift in marine resource management from a compartmentalized approach of dealing with resources on a species basis to an approach based on management of spatially defined ecosystems requires an accurate accounting of energy flow. The flow of energy from primary production through the food web will ultimately limit upper trophic-level fishery yields. In this work, we examine the relationship between yield and several metrics including net primary production, chlorophyll concentration, particle-export ratio, and the ratio of secondary to primary production. We also evaluate the relationship between yield and two additional rate measures that describe the export of energy from the pelagic food web, particle export flux and mesozooplankton productivity. We found primary production is a poor predictor of global fishery yields for a sample of 52 large marine ecosystems. However, chlorophyll concentration, particle-export ratio, and the ratio of secondary to primary production were positively associated with yields. The latter two measures provide greater mechanistic insight into factors controlling fishery production than chlorophyll concentration alone. Particle export flux and mesozooplankton productivity were also significantly related to yield on a global basis. Collectively, our analyses suggest that factors related to the export of energy from pelagic food webs are critical to defining patterns of fishery yields. Such trophic patterns are associated with temperature and latitude and hence greater yields are associated with colder, high latitude ecosystems.  相似文献

The relationship between changes in soil nutrient characteristics and fynbos community boundaries was investigated near Cape Agulhas, South Africa. Soil characteristics relating to total nutrient content (pH, total N and total P, organic carbon, and various cations) were assessed at sites along three transects crossing the boundaries between five plant communities. Dynamics of available N and P in soils of three communities were studied in the field over one year, using ion-exchange resins. There was a wide range in the degree of change in soil nutrient content across different community boundaries. The characteristics that varied most were pH, total N, Ca and total P. Differences in available nutrients among soils indicated that the communities in this landscape were associated with a mosaic of N and P availability. It is proposed that spatial variation in soil nutrient availability rather than total soil nutrient contents may be important in explaining landscape-level species distributions and community composition in nutrient-poor mediterranean-climate ecosystems.  相似文献

Breeding snow petrels (Pagodroma nivea) may be a major source of nutrients for the ecosystems on the inland nunataks (mountain outcrops) of continental Antarctica. To test this theory, the δ15N and δ13C values of soil and plants (moss, lichen and algae) from 11 nunataks with and without breeding snow petrels in northwestern Dronning Maud Land were compared. High δ15N values in all samples from nunataks with breeding birds provide a strong indication of the presence of breeding birds, as well as the use of bird-derived nitrogen by the plants of these ecosystems. Resting oxygen consumption was measured in anaesthetized male and female common marmosets (Callithrix jacchus) at 29 degrees C. Injection of noradrenaline caused a marked increase in oxygen consumption (63%), but this response was lower in males. Large deposits (750 mg) of brown adipose tissue (BAT) were dissected from the cervical, subscapular, axillary and perirenal areas. High levels of guanosine diphosphate binding to isolated brown fat mitochondria were observed, indicating that the thermogenic proton conductance pathway is very active in brown fat from the marmoset. High levels of brown adipose tissue thermogenesis in these animals may be related to a requirement for diet-induced thermogenesis.  相似文献

Recently, two mandibular traits--ramus flexure and gonial eversion--have come under close scrutiny (Loth & Henneberg 1996, 2000). The present study investigates the reliability of these two traits when each is applied as a single and independent indicator of sex, including the question of repeatability. The investigation was designed to give insights into possible confounding factors such as age and remodeling after tooth loss. Two samples, one of forensic (N = 153) and one of archaeological provenance (N = 80), were examined. The forensic sample was evaluated by a single observer while the archaeological sample was independently scored by three different observers. The results document that age and localized tooth loss seriously reduce the accuracy of these traits. For ramus flexure, male accuracy was only 66%, while female accuracy was even lower (32%). Overall accuracy was 59%. It is believed that the original scoring system devised by Loth and Henneberg (1996) creates an inherent bias in favor of males. For gonial eversion, a similar picture emerged (75.4% for males, 45.2% for females and 69.3% overall accuracy). Furthermore, both indicators are prone to intra- as well as inter-observer bias. While both possess some merit as sex indicators, they show marked functional and adaptive responses and may not be suitable for all samples.  相似文献
