Variation in upper limb long bone cross‐sectional properties may reflect a phenotypically plastic response to habitual loading patterns. Structural differences between limb bones have often been used to infer past behavior from hominin remains; however, few studies have examined direct relationships between behavioral differences and bone structure in humans. To help address this, cross‐sectional images (50% length) of the humeri and ulnae of university varsity‐level swimmers, cricketers, and controls were captured using peripheral quantitative computed tomography. High levels of humeral robusticity were found in the dominant arms of cricketers, and bilaterally among swimmers, whereas the most gracile humeri were found in both arms of controls, and the nondominant arms of cricketers. In addition, the dominant humeri of cricketers were more circular than controls. The highest levels of ulnar robusticity were also found in the dominant arm of cricketers, and bilaterally amongst swimmers. Bilateral asymmetry in humeral rigidity among cricketers was greater than swimmers and controls, while asymmetry for ulnar rigidity was greater in cricketers than controls. The results suggest that more mechanically loaded upper limb elements––unilaterally or bilaterally––are strengthened relative to less mechanically loaded elements, and that differences in mechanical loading may have a more significant effect on proximal compared to distal limb segments. The more circular humerus in the dominant arm in cricketers may be an adaptation to torsional strain associated with throwing activities. The reported correspondence between habitual activity patterns and upper limb diaphyseal properties may inform future behavioral interpretations involving hominin skeletal remains. Insulin resistance, as well as vascular disease, both share a relevant genetic background taking the influence of a positive family history of these disorders. On the other hand, insulin resistance is associated with a proatherosclerotic disturbance in nitric oxide dependent vasodilation, probably contributing to the link between these two disorders. We examined the association between nitric oxide dependent vasodilation (measured with high resolution ultrasound at 13 MHz) and three relevant NO-synthase (eNOS)-polymorphisms in 200 insulin resistant subjects participating in the Tuebinger Lifestyle Intervention Program (TULIP). This study revealed that carriers of the eNOS intron 4 polymorphism (aa 2.16%; ab 24.2%; bb 73.2%) show significantly worse endothelial, and thereby eNOS dependent vasodilation (p=0.03, multivariate ANOVA), as compared to wildtype carriers. The 5' UTR T-786C and the G894 T polymorphism did not show any influence on eNOS-activity. In subjects at increased risk to develop type 2 diabetes, the eNOS intron 4 polymorphism is independently associated with endothelial function as indicated by disturbed endothelial NO production. Due to the high prevalence and the relatively strong effect, this polymorphism might help to identify subjects at increased risk for atherosclerosis associated with overweight and insulin resistance.  相似文献

Simulated data were used to determine the properties of multivariate prediction of breeding values for categorical and continuous traits using phenotypic, molecular genetic and pedigree information by mixed linear-threshold animal models via Gibbs sampling. Simulation parameters were chosen such that the data resembled situations encountered in Warmblood horse populations. Genetic evaluation was performed in the context of the radiographic findings in the equine limbs. The simulated pedigree comprised seven generations and 40 000 animals per generation. The simulated data included additive genetic values, residuals and fixed effects for one continuous trait and liabilities of four binary traits. For one of the binary traits, quantitative trait locus (QTL) effects and genetic markers were simulated, with three different scenarios with respect to recombination rate (r) between genetic markers and QTL and polymorphism information content (PIC) of genetic markers being studied: r = 0.00 and PIC = 0.90 (r0p9), r = 0.01 and PIC = 0.90 (r1p9), and r = 0.00 and PIC = 0.70 (r0p7). For each scenario, 10 replicates were sampled from the simulated horse population, and six different data sets were generated per replicate. Data sets differed in number and distribution of animals with trait records and the availability of genetic marker information. Breeding values were predicted via Gibbs sampling using a Bayesian mixed linear-threshold animal model with residual covariances fixed to zero and a proper prior for the genetic covariance matrix. Relative breeding values were used to investigate expected response to multi- and single-trait selection. In the sires with 10 or more offspring with trait information, correlations between true and predicted breeding values ranged between 0.89 and 0.94 for the continuous traits and between 0.39 and 0.77 for the binary traits. Proportions of successful identification of sires of average, favourable and unfavourable genetic value were 81% to 86% for the continuous trait and 57% to 74% for the binary traits in these sires. Expected decrease of prevalence of the QTL trait was 3% to 12% after multi-trait selection for all binary traits and 9% to 17% after single-trait selection for the QTL trait. The combined use of phenotype and genotype data was superior to the use of phenotype data alone. It was concluded that information on phenotypes and highly informative genetic markers should be used for prediction of breeding values in mixed linear-threshold animal models via Gibbs sampling to achieve maximum reduction in prevalences of binary traits.  相似文献

Proteolytic cleavage of amyloid precursor protein by beta-secretase (BACE-1) and gamma-secretase leads to formation of beta-amyloid (A beta) a key component of amyloid plaques, which are considered the hallmark of Alzheimer's disease. Small molecule inhibitors of BACE-1 may reduce levels of A beta and thus have therapeutic potential for treating Alzheimer's disease. We recently reported the identification of a novel small molecule BACE-1 inhibitor N-[2-(2,5-diphenyl-pyrrol-1-yl)-acetyl]guanidine (3.a.1). We report here the initial hit-to-lead optimization of this hit and the SAR around the aryl groups occupying the S(1) and S(2') pockets leading to submicromolar BACE-1 inhibitors.  相似文献

Astronauts on exploratory missions will experience a complex environment, including microgravity and radiation. While the deleterious effects of unloading on bone are well established, fewer studies have focused on the effects of radiation. We previously demonstrated that 2 Gy of ionizing radiation has deleterious effects on trabecular bone in mice 4 months after exposure. The present study investigated the skeletal response after total doses of proton radiation that astronauts may be exposed to during a solar particle event. We exposed mice to 0.5, 1 or 2 Gy of whole-body proton radiation and killed them humanely 117 days later. Tibiae and femora were analyzed using microcomputed tomography, mechanical testing, mineral composition and quantitative histomorphometry. Relative to control mice, mice exposed to 2 Gy had significant differences in trabecular bone volume fraction (-20%), trabecular separation (+11%), and trabecular volumetric bone mineral density (-19%). Exposure to 1 Gy radiation induced a nonsignificant trend in trabecular bone volume fraction (-13%), while exposure to 0.5 Gy resulted in no differences. No response was detected in cortical bone. Further analysis of the 1-Gy mice using synchrotron microCT revealed a significantly lower trabecular bone volume fraction (-13%) than in control mice. Trabecular bone loss 4 months after exposure to 1 Gy highlights the importance of further examination of how space radiation affects bone.  相似文献

Recent development of the phosphate chelator, Phos-tag, together with Phos-tag pendant reagents, has provided new methods for detection of phosphorylated serine, threonine, tyrosine, and histidine residues in phosphoproteins. We have investigated the use of Phos-tag for detection and quantification of phospho-aspartate in response regulator proteins that function within two-component signaling systems. Alternative methods are especially important, because the labile nature of the acylphosphate bond in response regulator proteins has restricted the application of many traditional methods of phosphoprotein analysis. We demonstrate that Phos-tag gel stain can be used to detect phospho-Asp in response regulators and that Phos-tag acrylamide gel electrophoresis can be used to separate phosphorylated and unphosphorylated forms of response regulator proteins. The latter method, coupled to Western blot analysis, enables detection of specific phosphorylated proteins in complex mixtures such as cell lysates. Standards of phosphorylated proteins can be used to correct for hydrolysis of the labile phospho-Asp bond that invariably occurs during analysis. We have employed Phos-tag methods to characterize the phosphorylation state of the Escherichia coli response regulator PhoB both in vitro, using purified protein, and in vivo, by analyzing lysates of cells grown under different conditions of induction of the PhoR/PhoB phosphate assimilation pathway.  相似文献

Many populations of native Acacia raddiana trees in ephemeral riverbeds in the Negev desert are suffering from high mortality. Road-building techniques that cut off water to trees in downstream populations and the pumping of aquifers for agriculture have been mooted as causes of this mortality. We studied the water relations of nine isolated populations of these trees using a pressure chamber and stable carbon isotopes in order to determine whether current water stress could be the cause of this mortality. High mortality populations had more negative water potentials, lower relative water contents, and lower cell water volumes at full turgor than low mortality populations. Thus, mortality is correlated with current inter-population differences in water stress. However, there was no difference in water potentials between trees upstream and downstream of roads. Furthermore, greater water stress in trees after a dry winter than in the previous summer indicates that these trees are largely drawing on surface floods for water. Thus, road-building practices and aquifer pumping are unlikely causes of current water stress. Hence, water stress is linked to mortality in these trees, although our results indicate that irregular water stress (perhaps in drought years only) is probably the cause of this mortality.  相似文献
