Abscisic acid is shown to enhance the permeability of crude egg lecithin and asolectin bilayers to water, urea and erythritol although it exhibits no effect on pure synthetic (phosphatidylethanolamine-free) dimyristoylphosphatidylcholine bilayers. Addition of dipalmitoylphosphatidylethanolamine to dimyristoylphosphatidylcholine bilayers at 10 or 20 membrane mole percent makes the membrane permeability responsive to abscisic acid. An abscisic acid-phosphatidylethanolamine interaction is also described for liposome aggregation. Both abscisic acid-induced permeability and aggregation changes are pH dependent with the undissociated form of the hormone exhibiting a greater effect than the dissociated, charged form. Enhancement of erythritol permeability is greater with the physiologically active cis-trans ABA isomer than with the inactive trans-trans isomer.  相似文献

Omega-3 polyunsaturated fatty acids (n-3 PUFA), enriched in fish oils, are increasingly recognized to have potential benefits for treating many human afflictions. Despite the importance of PUFA, their molecular mechanism of action remains unclear. One emerging hypothesis is that phospholipids containing n-3 PUFA acyl chains modify the structure and composition of membrane rafts, thus affecting cell signaling. In this study the two major n-3 PUFA found in fish oils, eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids, are compared. Using solid-state (2)H NMR spectroscopy we explored the molecular organization of 1-[(2)H(31)]palmitoyl-2-eicosapentaenoylphosphatidylcholine (PEPC-d(31)) and 1-[(2)H(31)]palmitoyl-2-docosahexaenoylphosphatidylcholine (PDPC-d(31)) in mixtures with sphingomyelin (SM) and cholesterol (chol). Our results indicate that whereas both PEPC-d(31) and PDPC-d(31) can accumulate into SM-rich/chol-rich raftlike domains, the tendency for DHA to incorporate into rafts is more than twice as great as for EPA. We propose that DHA may be the more bioactive component of fish oil that serves to disrupt lipid raft domain organization. This mechanism represents an evolution in the view of how PUFA remodel membrane architecture.  相似文献

Photooxidation of water-soluble chlorophyllin was measured in the presence of glycerol, phosphate, choline, phosphorylcholine, ethanolamine, phosphorylethanolamine and liposomes made from phosphatidylcholine and phosphatidylethanolamine. The choline containing compounds protected chlorophyllin from destruction while the ethanolamines enhanced degradation. The difference in activity between the two families of compounds is related to the availability of their protons. These experiments provide a further indication that in the thylakoid phosphatidylcholine may protect chlorophyll from photooxidation.  相似文献

Sexual dimorphism in body size (sexual size dimorphism) is common in many species. The sources of selection that generate the independent evolution of adult male and female size have been investigated extensively by evolutionary biologists, but how and when females and males grow apart during ontogeny is poorly understood. Here we use the hawkmoth, Manduca sexta, to examine when sexual size dimorphism arises by measuring body mass every day during development. We further investigated whether environmental variables influence the ontogeny of sexual size dimorphism by raising moths on three different diet qualities (poor, medium and high). We found that size dimorphism arose during early larval development on the highest quality food treatment but it arose late in larval development when raised on the medium quality food. This female-biased dimorphism (females larger) increased substantially from the pupal-to-adult stage in both treatments, a pattern that appears to be common in Lepidopterans. Although dimorphism appeared in a few stages when individuals were raised on the poorest quality diet, it did not persist such that male and female adults were the same size. This demonstrates that the environmental conditions that insects are raised in can affect the growth trajectories of males and females differently and thus when dimorphism arises or disappears during development. We conclude that the development of sexual size dimorphism in M. sexta occurs during larval development and continues to accumulate during the pupal/adult stages, and that environmental variables such as diet quality can influence patterns of dimorphism in adults.  相似文献

Free glycine was shown to diffuse very slowly across synthetic bimolecular lipid membranes, whereas several N-substituted derivatives of glycine were found to penetrate the membranes more readily. Pyridoxal, formaldehyde and acetaldehyde were shown to enhance the diffusion of glycine across the membranes, presumably the result of imine formation between the aldehyde and the α-amino group of glycine. Several N-substituted glycines were synthesized and their rates of efflux from liposomes were found to be related to their water-chloroform partition coefficients. This is the first demonstration of carrier-mediated diffusion of amino acids across bimolecular lipid membrane.  相似文献

These experiments show that 7.3 and 3.65 per cent ether solutions cause an increase in respiration followed by a decrease. The results agree with those of Haas on Laminaria, of Gustafson on higher fungi, and of Mrs. Brooks on bacteria. It is commonly argued that inbred individuals should be more sensitive to environmental stress than are outbred individuals, presumably because stress increases the expression of deleterious recessive alleles. However, the degree to which inbreeding depression is dependent on environmental conditions is not clear. We use two populations of the seed-feeding beetle, Callosobruchus maculatus, to test the hypotheses that (a) inbreeding depression varies among rearing temperatures, (b) inbreeding depression is greatest at the more stressful rearing temperatures, (c) the degree to which high or low temperature is stressful for larval development varies with inbreeding level, and (d) inbreeding depression is positively correlated between different environments. Inbreeding depression (δ) on larval development varied among temperatures (i.e., there was a significant inbreeding-environment interaction). Positive correlations for degree of inbreeding depression were consistently found between all pairs of temperatures, suggesting that at least some loci affected inbreeding depression across all temperatures examined. Despite variation in inbreeding depression among temperatures, inbreeding depression did not increase consistently with our proxy for developmental stress. However, inbreeding changed which environments are benign versus stressful for beetles; although 20°C was not a stressful rearing temperature for outbred beetles, it became the most stressful environment for inbred larvae. The finding that inbreeding-environment interactions can cause normally benign environments to become stressful for inbred populations has important consequences for many areas of evolutionary genetics, artificial breeding (for conservation or food production), and conservation of natural populations.  相似文献

Plasma high (HDL) and low (LDL) density lipoproteins were isolated from rats fed a diet supplemented with either fish (menhaden) oil or hydrogenated coconut oil (control). Fluorescence polarization and electron spin resonance of labelled fatty acid probe molecules incorporated into the outer amphiphilic monolayer of HDL indicate molecular motion is restricted in the upper portion of the acyl chain following the fish oil diet, which is consistent with a 'hook' conformation predicted by preliminary molecular model calculations for n-3 fatty acids (the predominant component of fish oil). Negligible dependence on diet was observed in LDL. Thus, a HDL specific effect of dietary fish oil on molecular fluidity and order in the outer monolayer of rat lipoproteins is suggested.  相似文献

The role of cattle in the volatile loss of nitrogen from a shortgrass steppe   总被引:10,自引:7,他引:3  
The cycling and volatile loss of N derived from cattle urine at upland and lowland sites within the shortgrass steppe of eastern Colorado was studied, using15N-labelled urea as an N source. Losses of NH 0 3 were determined by direct measurement and by difference. Losses were higher from coarse (27% summer, 12% winter) than from fine textured (0–2%) soils. Immobilization and plant uptake of N accounted for significant amounts of added N. Extrapolating our plot measurements to a typical pasture, using spatially and temporally stratified urine deposition data, losses from upland sites were calculated to be 0.016 g N · m-2 · y-1, while losses from lowland sites were negligible. This resulted in an average loss of 0.011 g N · m-2 · y-1 for a pasture divided 70:30 between uplands and lowlands. The loss of urine N calculated assuming no spatial stratification would be sevenfold higher (0.076 g N · m-2 · y-1). Losses of NH 0 3 from urine, animal biomass removal, and NH2O loss totaled only 0.07 g N · m-2 · y-1 , or about 25% of wet deposition input. We calculated a potential loss of NH 0 3 from senescing vegetation of 0.26 g N · m-2 · y-1, an order of magnitude larger than all other losses combined.  相似文献

Retinol at levels above 0.5 n moles/mg protein and retinoic acid at levels above 0.25 n moles/mg protein are shown to uncouple mitochondrial oxidative phosphorylation. Uncoupling is measured with the substrates ascorbate plus N,N,N',N'-tetra-methyl-p-phenylenediamine, succinate and beta-hydroxybutyrate. These experiments indicate that the uncoupling effects of retinol and retinoic acid are not site specific but likely are the result of the vitamin being a general membrane bilayer disrupting agent.  相似文献
