Mares treated with subcutaneous deslorelin implants on the first postpartum estrus early in the breeding season had significant reductions in the number of large follicles at early pregnancy examinations and delayed return to estrus (in mares that failed to become pregnant); these adverse effects were attributed to a prolonged release of the drug from the implant. In 2003, an injectable short-term release (<24 h) deslorelin product became available. The objective of this study was to determine if this product would hasten ovulation in early foaling first postpartum estrus mares without reducing the number of large follicles at early pregnancy examination (14-15 days postovulation). Beginning 5-6 days postpartum, first postpartum estrus (foal-heat) mares were teased daily and examined thrice weekly (Tuesday, Thursday and Saturday) by transrectal ultrasonography. Mares in estrus with a follicle > or = 34 mm diameter on Tuesdays or Thursdays were alternately assigned to: Treatment 1, n = 17; 1.5 mg injectable short-term release deslorelin, or Treatment 2, n = 16; Control (no treatment). The schedule allowed accurate determination of the number of mares ovulating within 2 days of treatment (i.e., ovulations detected on Thursday or Saturday). Mares were mated on the day of treatment and at 2-day intervals until either ovulation was confirmed or until behavioral estrus ceased. Transrectal ultrasonography was done 14-15 days after ovulation to assess ovarian follicles and pregnancy status. Fewer covers were required and more mares ovulated within 2 days of treatment in deslorelin-treated versus Control mares (P < 0.01). Pregnancy rates were normal (69%) in deslorelin-treated mares. The number of large follicles 14-15 days after ovulation did not differ between deslorelin-treated and Control mares (P > 0.10), suggesting follicular suppression did not occur with this formulation of deslorelin.  相似文献

Rapeseed (Brassica napus L.) is the leading European oilseed crop serving as source for edible oil and renewable energy. The objectives of our study were to (i) examine the population structure of a large and diverse set of B. napus inbred lines, (ii) investigate patterns of genetic diversity within and among different germplasm types, (iii) compare the two genomes of B. napus with regard to genetic diversity, and (iv) assess the extent of linkage disequilibrium (LD) between simple sequence repeat (SSR) markers. Our study was based on 509 B. napus inbred lines genotyped with 89 genome-specific SSR primer combinations. Both a principal coordinate analysis and software STRUCTURE revealed that winter types, spring types, and swedes were assigned to three major clusters. The genetic diversity of winter oilseed rape was lower than the diversity found in other germplasm types. Within winter oilseed rape types, a decay of genetic diversity with more recent release dates and reduced levels of erucic acid and glucosinolates was observed. The percentage of linked SSR loci pairs in significant (r 2 > Q 95 unlinked loci pairs) LD was 6.29% for the entire germplasm set. Furthermore, LD decayed rapidly with distance, which will allow a relatively high mapping resolution in genome-wide association studies using our germplasm set, but, on the other hand, will require a high number of markers.  相似文献

Growth is an integrative trait that responds to environmental factors and is crucial for plant fitness. A major environmental factor influencing plant growth is nutrient supply. In order to explore this relationship further, we quantified growth-related traits, ion content, and other biochemical traits (protein, hexose, and chlorophyll contents) of a recombinant inbred line population of Arabidopsis (Arabidopsis thaliana) grown on different levels of potassium and phosphate. Performing an all subsets multiple regression analyses revealed a link between growth-related traits and mineral nutrient content. Based on our results, up to 85% of growth variation can be explained by variation in ion content, highlighting the importance of ionomics for a broader understanding of plant growth. In addition, quantitative trait loci (QTLs) were detected for growth-related traits, ion content, further biochemical traits, and their responses to reduced supplies of potassium or phosphate. Colocalization of these QTLs is explored, and candidate genes are discussed. A QTL for rosette weight response to reduced potassium supply was identified on the bottom of chromosome 5, and its effects were validated using selected near isogenic lines. These lines retained over 20% more rosette weight in reduced potassium supply, accompanied by an increase in potassium content in their leaves.Plants in natural environments face abiotic constraints limiting growth and ultimately affecting their fitness. In response to such constraints, flowering time (Korves et al., 2007) and seed dormancy (Donohue et al., 2005) as well as vegetative growth (Barto and Cipollini, 2005; Milla et al., 2009) are the main traits controlling fitness (for review, see Alonso-Blanco et al., 2009). These traits are under the control of complex networks integrating genetic (G) and environmental (E) factors as well as their interaction (G × E). Due to the implications for food and renewable energy sources, dissecting the genetic architecture that underlies plant growth is becoming a priority for plant science (Rengel and Damon, 2008; Carroll and Somerville, 2009; Gilbert, 2009).Plant growth is highly dependent on mineral nutrient uptake (Clarkson, 1980; Sinclair, 1992). Minerals can be distinguished into two categories based on the amount required by plants: micronutrients, which are found in relatively small amounts in the plant (such as copper and iron), and macronutrients, which constitute between 1,000 and 15,000 μg g−1 plant dry weight (such as potassium and phosphate; Marschner, 1995, Buchanan et al., 2002). Phosphate is an important structural and signaling molecule with an essential role in photosynthesis, energy conservation, and carbon metabolism. Its deficiency leads to a reduction of growth and an increase of pathogen susceptibility (Marschner, 1995; Williamson et al., 2001; Abel et al., 2002; López-Bucio et al., 2005; Poirier and Bucher, 2008; Vijayraghavan and Soole, 2010). Potassium is not incorporated into any organic substances but acts as the major osmoticum of the cell, controlling cell expansion, plasma membrane potential and transport, pH value, and many other catalytic processes (Maathuis and Sanders, 1996; Armengaud et al., 2004; Christian et al., 2006; Di Cera, 2006). Potassium deficiency leads to reduced plant growth, a loss of turgor, increased susceptibility to cold stress and pathogens, and the development of chlorosis and necrosis (Marschner, 1995; Véry and Sentenac, 2003; Ashley et al., 2006; Amtmann et al., 2008). To cope with changes in nutrient availability, plants have evolved different mechanisms of adaptation, such as changes in ion transporter expression and activity (Ashley et al., 2006; Jung et al., 2009), morphological changes, such as an increase in root growth to explore more soil volume (Marschner, 1995; Shirvani et al., 2001; Jiang et al., 2007; Jordan-Meille and Pellerin, 2008), or acidification of the surrounding soil in order to mobilize more mineral nutrients (for review, see Ryan et al., 2001). Although these adaptations are well known, the mechanisms involved in sensing and signaling low mineral nutrient status are less well understood, despite significant progress in this area being made (Doerner, 2008; Jung et al., 2009; Luan et al., 2009; Wang and Wu, 2010).One approach to identify genes that are involved in plant responses to environmental factors is to perform a quantitative trait locus (QTL) analysis on a mapping population grown in contrasting environments, allowing the identification of QTL-environment (QTL × E) interactions. Some QTLs for growth-related traits in response to environmental changes were cloned already. For example, the differential response of root growth of some Arabidopsis (Arabidopsis thaliana) accessions to phosphate starvation led to the identification of allelic differences responsible for this phenotype (Reymond et al., 2006; Svistoonoff et al., 2007). Other studies have identified QTLs for shoot dry matter under changing nitrogen supply (Rauh et al., 2002; Loudet et al., 2003). In parallel to natural variation for growth, natural variation for ion content has also been reported. In Arabidopsis, considerable variation in the content of mineral nutrients exists both in seeds (Vreugdenhil et al., 2004; Waters and Grusak, 2008) and in leaves (Harada and Leigh, 2006; Rus et al., 2006; Baxter et al., 2008a; Morrissey et al., 2009). Furthermore, changes in mineral nutrient homeostasis have also been reported to be associated with characteristic multivariate changes in the leaf ionome, the mineral nutrient and trace element composition of an organism or an organ (Baxter et al., 2008b). Due to higher throughput and lower costs, such “omics” analyses examining alterations of large numbers of certain molecules at once have recently become available for mapping purposes. Some QTL studies have linked the variations of these omics data to variation of growth or other physiological traits. For instance, Meyer et al. (2007) and Schauer et al. (2008) linked plant growth or morphological traits to a synergistic network of metabolomic compounds in Arabidopsis and tomato (Solanum lycopersicum), respectively. In addition, Sulpice et al. (2009) associated differences in growth with starch content using a set of Arabidopsis accessions. Compiling the importance of ions in the process of cell division (Lai et al., 2007; Sano et al., 2007) or cell expansion (Philippar et al., 1999; Elumalai et al., 2002), ionomics appears to be a major unexplored field for understanding growth.In this study, we focus on variation in plant growth, the root and leaf ionomes, and their response to varying supplies of potassium and phosphate. Studying variations for these traits among recombinant inbred lines (RILs) in Arabidopsis enabled us to detect QTL and QTL × E interactions for all of these traits. To understand the observed variation in plant growth, predictors that explained a high percentage of variation of growth-related traits have been selected especially among the root and leaf ionomes. The colocalization between growth-related trait QTLs and QTLs for their predictors allowed us to point out genetic regions of possible causality. In addition, the effect of a growth-response QTL on reduced potassium supply was validated with selected near isogenic lines (NILs) that maintained a higher rosette weight when grown in reduced potassium supply. This growth advantage went along with significant changes in ion contents that further emphasize the impact of the ionome in plant growth variations.  相似文献

中华穿山甲(Manis pentadactyla)属于全球极度濒危物种,也是我国一级保护动物。对中华穿山甲的非法捕杀曾导致其种群数量锐减。但是,近年来相关研究报道较少,穿山甲分布状况不明,极大地制约了对该物种的有效保护。搜集了近年来国内中华穿山甲的救护记录和救护新闻,甄别出67个记录分布点,利用最大熵模型软件(MaxEnt)进行因子筛选,结果表明最冷季度降水量、人口密度、年降水量、坡度、坡向、海拔等6个环境变量是与中华穿山甲分布显著相关的影响因子。基于6个主导环境变量构建的MaxEnt模型AUC平均值为0.961±0.014,预测结果达到极好标准。刀切法(Jackknife)表明,其中最冷季度降水量、年降水量、人口密度和海拔是影响中华穿山甲分布的主要因素。中华穿山甲适宜生境(出现概率大于0.498)具有以下特点:最冷季度降水量141.22-439.46 mm,年降水量1471.67-2386.56 mm,人口密度≥390人/km2,海拔<316.98 m。该模型预测中华穿山甲在我国的潜在分布适宜区主要位于我国长江以南地区,总面积约为74.27×104 km2,占国土面积的7.73%,主要集中在江西、广东、湖南和广西省,面积分别占该区域的97.58%,89.65%,76.90%和73.08%;其次是浙江、福建、台湾和安徽省。湖北、江苏、四川、云南、贵州等省份也有中华穿山甲的零星分布。湖北东南部、江苏南部、浙江西南部和福建西北部等与江西接壤的区域也是中华穿山甲的重要潜在分布适宜区。明确中华穿山甲的潜在分布适宜区,可为该物种的种群保护和栖息地管理提供科技支撑。  相似文献

Ozone and light effects on endophytic colonization by Apiognomonia errabunda of adult beech trees (Fagus sylvatica) and their putative mediation by internal defence compounds were studied at the Kranzberg Forest free-air ozone fumigation site. A. errabunda colonization was quantified by "real-time PCR" (QPCR). A. errabunda-specific primers allowed detection without interference by DNA from European beech and several species of common genera of plant pathogenic fungi, such as Mycosphaerella, Alternaria, Botrytis, and Fusarium. Colonization levels of sun and shade leaves of European beech trees exposed either to ambient or twice ambient ozone regimes were determined. Colonization was significantly higher in shade compared to sun leaves. Ozone exhibited a marginally inhibitory effect on fungal colonization only in young leaves in 2002. The hot and dry summer of 2003 reduced fungal colonization dramatically, being more pronounced than ozone treatment or sun exposure. Levels of soluble and cell wall-bound phenolic compounds were approximately twice as high in sun than in shade leaves. Acylated flavonol 3- O-glycosides with putatively high UV-B shielding effect were very low in shade canopy leaves. Ozone had only a minor influence on secondary metabolites in sun leaves. It slightly increased kaempferol 3- O-glucoside levels exclusively in shade leaves. The frequently prominent hydroxycinnamic acid derivative, chlorogenic acid, was tested for its growth inhibiting activity against Apiognomonia and showed an IC50 of approximately 8 mM. Appearance of Apiognomonia-related necroses strongly correlated with the occurrence of the stress metabolite, 3,3',4,4'-tetramethoxybiphenyl. Infection success of Apiognomonia was highly dependent on light exposure, presumably affected by the endogenous levels of constitutive phenolic compounds. Ozone exerted only minor modulating effects, whereas climatic factors, such as pronounced heat periods and drought, were dramatically overriding.  相似文献

A major goal of today's biology is to understand the genetic basis of quantitative traits. This can be achieved by statistical methods that evaluate the association between molecular marker variation and phenotypic variation in different types of mapping populations. The objective of this work was to evaluate the statistical power of quantitative trait loci (QTL) detection of various multi-parental mating designs, as well as to assess the reasons for the observed differences. Our study was based on an empirical data of 20 Arabidopsis thaliana accessions, which have been selected to capture the maximum genetic diversity. The examined mating designs differed strongly with respect to the statistical power to detect QTL. We observed the highest power to detect QTL for the diallel cross with random mating design. The results of our study suggested that performing sibling mating within subpopulations of joint-linkage mapping populations has the potential to considerably increase the power for QTL detection. Our results, however, revealed that using designs in which more than two parental alleles segregate in each subpopulation increases the power even more.  相似文献

This study was designed to assess whether physiological activation of the sympathetic nervous system induced by exercise changes adipose tissue responsiveness to catecholamines in humans. Lipid mobilization in abdominal subcutaneous adipose tissue was studied with the use of a microdialysis method in 11 nontrained men (age: 22. 3 +/- 1.5 yr; body mass index: 23.0 +/- 1.6). Adipose tissue adrenergic sensitivity was explored with norepinephrine, dobutamine (beta(1)-agonist), or terbutaline (beta(2)-agonist) perfused during 30 min through probes before and after 60-min exercise (50% of the maximal aerobic power). The increase in extracellular glycerol concentration during infusion was significantly lower after the exercise when compared with the increase observed before the exercise (P < 0.05, P < 0.02, and P < 0.01, respectively, for norepinephrine, dobutamine, and terbutaline). In a control experiment realized without exercise, no difference in norepinephrine-induced glycerol increase between the two infusions was observed. To assess the involvement of catecholamines in the blunted beta-adrenergic-induced lipolytic response after exercise, adipose tissue adrenergic sensitivity was explored with two 60-min infusions of norepinephrine or epinephrine separated by a 60-min interval. With both catecholamines, the increase in glycerol was significantly lower during the second infusion (P < 0.05). The findings suggest that aerobic exercise, which increased adrenergic activity, induces a desensitization in beta(1)- and beta(2)-adrenergic lipolytic pathways in human subcutaneous adipose tissue.  相似文献

Biparental incubation is a form of cooperation between parents, but it is not conflict‐free because parents trade off incubation against other activities (e.g. self‐maintenance, mating opportunities). How parents resolve such conflict and achieve cooperation remains unknown. To understand better the potential for conflict, cooperation and the constraints on incubation behaviour, investigation of the parents' behaviour, both during incubation and when they are off incubation‐duty, is necessary. Using a combination of automated incubation‐monitoring and radiotelemetry we simultaneously investigated the behaviours of both parents in the biparentally incubating Semipalmated Sandpiper Calidris pusilla, a shorebird breeding under continuous daylight in the high Arctic. Here, we describe the off‐nest behaviour of 32 off‐duty parents from 17 nests. Off‐duty parents roamed on average 224 m from their nest, implying that direct communication with the incubating partner is unlikely. On average, off‐duty parents spent only 59% of their time feeding. Off‐nest distance and behaviour (like previously reported incubation behaviour) differed between the sexes, and varied with time and weather. Males roamed less far from the nest and spent less time feeding than did females. At night, parents stayed closer to the nest and tended to spend less time feeding than during the day. Further exploratory analyses revealed that the time spent feeding increased over the incubation period, and that at night, but not during the day, off‐duty parents spent more time feeding under relatively windy conditions. Hence, under energetically stressful conditions, parents may be forced to feed more. Our results suggest that parents are likely to conflict over the favourable feeding times, i.e. over when to incubate (within a day or incubation period). Our study also indicates that Semipalmated Sandpiper parents do not continuously keep track of each other to optimize incubation scheduling and, hence, that the off‐duty parent's decision to remain closer to the nest drives the length of incubation bouts.  相似文献

CHO cells undergo proliferative arrest when incubated in medium deficient in the amino acid arginine (ADM). Cells arrested in this way can be released and resume mitotic activity after a brief lag period. The incidence of U.V.-induced sister chromatid exchanges (SCEs) induced in cells arrested in ADM was reduced when the cells were incubated in ADM after irradiation and prior to release. Periods of incubation in ADM of 24 and 48 h prior to release reduced the resulting SCE levels (relative to the SCE levels present in cells irradiated immediately prior to release) by an average of 35 and 45% respectively. A similar time-dependent decrease in the incidence of chromosome aberrations induced in CHO cells arrested in ADM was not observed. Despite the decrease in SCEs over time in ADM, the survival of ADM-arrested cells was not enhanced by a period of incubation in ADM after irradiation of 48 h. These observations are consistent with the hypothesis that the U.V.-induced lesions responsible for the induction of SCE are repaired in time in ADM-arrested CHO cells. Repair of those lesions resulting in chromosome aberrations was not detected in ADM-arrested CHO cells. This absence of repair of certain lesions was apparently reflected in the absence of any enhancement of cell survival.  相似文献
