The genes of nicotine dehydrogenase (NDH) were identified, cloned and sequenced from the catabolic plasmid pA01 of Arthrobacter nicotinovorans. In immediate proximity to this gene cluster is the beginning of the 6-hydroxy-L-niotine oxidase (6-HLNO) gene. NDH is composed of three subunits (A, B and C) of Mr 30011, 14924 and 87677. It belongs to a family of bacterial hydroxylases with a similar subunit structure; they have molybdopterin dinucleotide, FAD and Fe-S clusters as cofactors. Here the first complete primary structure of a bacterial hydroxylase is provided. Sequence alignments of each of the NDH subunits show similarities to the sequences of eukaryotic xanthine dehydrogenase (XDH) but not to other known molybdenum-containing bacterial enzymes. Based on alignment with XDH it is inferred that the smallest subunit (NDHB) carries an iron-sulphur cluster, that the middle-sized subunit (NDHA) binds FAD, and that the largest NDH subunit (NDHC) corresponds to the molybdopterin-binding domain of XDH. Expression of both the ndh and the 6-hlno genes required the presence of nicotine and molybdenum in the culture medium. Tungsten inhibited enzyme activity but not the synthesis of the enzyme protein. The enzyme was found in A. nicotinovorans cells in a soluble form and in a membrane-associated form. In the presence of tungsten the fraction of membrane-associated NDH increased.  相似文献

Abstract An arg 7 mutant of the green alga Chlamydomonas reinhardtii was transformed with pARG7.8, a plasmid bearing the wild-type ARG 7 gene. Out of 4100 arg+ transformants selected on an arginine-free medium supplemented with acetate, nine failed to grow on acetate-free medium (ac mutants). The results of the genetic and molecular analysis of several ac mutants are in agreement with the hypothesis that they originated from insertion of the incoming plasmid into the nuclear genome. These mutants should constitute valuable tools for isolating the corresponding wild-type genes after plasmid rescue into Escherichia coli .  相似文献

In a phytotron experiment four rice varieties (Pokkali, IR 28, IR 50, IR 31785-58-1-2-3-3) grown in individual pots were subjected to low (40/55% day/night) and high (75/90%) air humidity (RH), while soil salinity was gradually increased by injecting 0, 30, 60 or 120 mM NaCl solutions every two days. Bulk root and stem base water potential (SWP), abscisic acid (ABA) content of the xylem sap and stomatal resistance (rs) of the youngest fully expanded leaf were determined two days after each salt application. The SWP decreased and xylem ABA and rs increased throughout the 8 days of treatment. The effects were amplified by low RH. A chain of physiological events was hypothesized in which high soil electric conductivity (EC) reduces SWP, followed by release of root-borne ABA to the xylem and eventually resulting in stomatal closure. To explain varietal differences in stomatal reaction, supposed cause and effect variables were compared by linear regression. This revealed strong differences in physiological reactions to the RH and salt treatments among the test varieties. Under salt stress roots of IR 31785-58-1-2-3-3 produced much ABA under low RH, but no additional effect of low RH on rs could be found. By contrast, Pokkali produced little ABA, but rs was strongly affected by RH. RH did not affect the relationships EC vs. SWP and SWP vs. ABA in Pokkali, IR 28, and IR 50, but the relationship ABA vs. rs was strongly affected by RH. In IR 31785-58-1-2-3-3 RH strongly affected the relationship SWP vs. ABA, but had no effect on ABA vs. rs and EC vs. rs. The results are discussed regarding possible differences in varietal stomatal sensitivity to ABA and their implications for varietal salt tolerance.  相似文献

Abstract The natural nutritional environments of most fungi are spatially non-uniform, yet the majority of studies of fungal growth take no account of this fact. An experimental system is described which permits the growth responses of eucarpic fungi to heterogeneously distributed nutrient resources to be studied. The system comprises tesselations of agar tiles of contrasting nutrient status separated by air gaps. Growth responses in such systems of Alternaria alternata, Mucor sp., Phoma foveata , Rhizoctonia solani and Trichoderma viride are described. Generally, the growth of the fungi reflected the nutrient status of the underlying substrate. There was evidence for growth in low-nutrient tiles being greater when high-nutrient tiles were included in the tessellation. Reproductive structures tended to be formed only in low nutrient tiles with Trichoderma and Rhizoctonia and only high nutrient tiles with Alternaria . Growth responses of Rhizoctonia were strongly asymmetric in nutritionally symmetric, but heterogeneous, tesselations. The consequences of the observations for fungal growth in heterogeneous environments such as soil is discussed.  相似文献

We consider two-state automata playing repeatedly the Prisoner's Dilemma (or any other 2 × 2-game). The 16 × 16-payoff matrix is computed for the limiting case of a vanishingly small noise term affecting the interaction. Mass spectra of underivatized hexa- and heptapeptide amides related to Substance P have been obtained with a conventional electron ionization mass spectrometer using sample vaporization from a tungsten wire by the technique of rapid heating, proton transfer ionization using ammonia, and photoplate recording of spectra. These spectra exhibit little evidence of sample pyrolysis and are readily interpreted to yield amino acid sequences.  相似文献

From the leaves of Myrica gale 2′,4′-dihydroxy-6′-methoxy-3′,5′-dimethylchalcone has been isolated. The fruits yielded 2′-hydroxy-4′,6′-dimethoxy-3′-methyldihydrochalcone. The constitutions were deduced from spectroscopic data and confirmed by synthesis.  相似文献

The intial phases of auxin-induced growth in coleoptile segments of Avena sativa L. were investigated using a high resolution growth recording technique, based on an angular position sensing transducer. The first response to the hormone is a slight, transient reduction of the growth rate lasting about 5 min. After this phase growth rate increases to a maximum. The duration of the increase and the maximum clearly depend on the concentration of the hormone. With increasing auxin concentration the duration of the growth rate increase is reduced from about 80 min in 10-9 M indoleacetic acid (IAA) to about 14 min in 10-4 M IAA. After the maximum the growth rate declines. Looking at the maximum of the growth rate, we obtained a dose-response curve with a sharp increase between 10-9 M and 10-6 M IAA and a slight decline between 10-6 M and 10-4 M IAA. The effect of reducing agents on the nitrosation of methylguanidine (MG) and on the in vitro activation of dimethylnitrosamine (DMN) was examined by measuring DNA-repair synthesis (unscheduled incorporation of [3h]TdR), shifts in alkaline sucrose gradients, frequency of chromosome aberrations, and clone-forming capacity of cultured human fibroblasts. The reducing agents examined were sodium ascorbate, cysteine, cysteamine, and propyl gallate. Since the short-term bioassays used can be quantitated, it has become relatively easy to detect the inhibitory action of reducing compounds on the nitrosation reaction of MG and metabolic activation (with S-9 preparation) of the precarcinogen DMN, to measure their effective dose range, and to establish the most effective ratios between inhibitory agent and reactant. The results indicate that DNA-repair synthesis is a suitable short-term test for studying the numerous combinations and premutations between several carcinogenic or non-carcinogenic agents, and for estimating the capacity of inhibitory agents to affect formation and activation of chemical carcinogens.  相似文献
