The additive main effects and multiplicative interaction (AMMI) model has emerged as a powerful analytical tool for genotype x environment studies. The objective of the present study was to assess its value in quantitative trait locus (QTL) mapping. This was done through the analysis of a large two-way table of genotype-by-environment data of barley (Hordeum vulgare L.) grain yields, where the genotypes constituted a genetic population suitable for mapping studies. Grain yield data of 150 doubled haploid lines derived from the Steptoe x Morex cross, and the two parental lines, were taken by the North American Barley Genome Mapping Project (NABGMP) at 16 environments throughout the barley production areas of the USA and Canada. Four regions of the genome were responsible for most of the differential genotypic expression across environments. They accounted for approximately 50% of the genotypic main effect and 30% of the genotype x environment interaction (GE) sums of squares. The magnitude and sign of AMMI scores for genotypes and sites facilitate inferences about specific interactions. The parallel use of classification (cluster analysis of environments) and ordination (principal component analysis of GE matrix) techniques allowed most of the variation present in the genotype x environment matrix to be summarized in just a few dimensions, specifically four QTLs showing differential adaptation to four clusters of environments. Thus, AMMI genotypic scores, when the genotypes constituted a population suitable for QTL mapping, could provide an adequate way of resolving the magnitude and nature of QTL x environment interactions.Ignacio Romagosa was on sabbatical leave from the University of Lleida and the Institut de Recerca i Tecnologia Agroalimentàries, Lleida, Spain, when this study was conducted  相似文献

Quantitative information on large scale spatial patterns of biodiversity remains poor, especially for pelagic systems. In this paper the regional diversity of procellariiforms is mapped worldwide at the species level. These seabirds do not display a conventional latitudinal gradient of decreasing species richness towards high latitudes, but rather are most speciose between 37° and 59°S in all ocean basins. Based on data for foraging ranges, areas with the highest species richness and the most species with smaller range sizes are all found in the vicinity of New Zealand and its sub-Antarctic islands. In contrast, data for breeding ranges show islands in the southern Indian and Atlantic oceans to have the highest number of breeding species, while these islands and New Zealand have the most species with smaller range sizes. No northern hemisphere regions are amongst the top ten grid cells for foraging and breeding species richness, although Hawaii has the highest species richness of procellariilforms north of the equator. Northern Baja California. Madeira, the Canary islands, and the west coast of South America are all important sites of narrow endemism in the northern hemisphere. High species richness and narrow endemism coincide with areas of significant longline fishing activity in the southern hemisphere. Near-minimum sets based on one and three representations demonstrate that if all procellariiform species are to be retained, large areas of the ocean and almost all breeding sites require conservation.  相似文献

We have used top-down metabolic control analysis to investigate the control of carbon flux through potato (Solanum tuberosum) plants during tuberisation. The metabolism of the potato plant was divided into two blocks of reactions (the source and sink blocks) that communicate through the leaf apoplastic sucrose pool. Flux was measured as the transfer of 14C from CO2 to the tuber. Flux and apoplastic sucrose concentration were varied either by changing the light intensity or using transgenic manipulations that specifically affect the source or sink blocks, and elasticity coefficients were measured. We have provided evidence in support of our assumption that apoplastic sucrose is the only communicating metabolite between the source and sink blocks. The elasticity coefficients were used to calculate the flux control coefficients of the source and sink blocks, which were 0.8 and 0.2, respectively. This work suggests that the best strategy for the manipulation of tuber yield in potato will involve increases in photosynthetic capacity, rather than sink metabolism.  相似文献

Lignin mineralization represents a critical flux in the terrestrial carbon (C) cycle, yet little is known about mechanisms and environmental factors controlling lignin breakdown in mineral soils. Hypoxia is thought to suppress lignin decomposition, yet potential effects of oxygen (O2) variability in surface soils have not been explored. Here, we tested the impact of redox fluctuations on lignin breakdown in humid tropical forest soils during ten‐week laboratory incubations. We used synthetic lignins labeled with 13C in either of two positions (aromatic methoxyl or propyl side chain Cβ) to provide highly sensitive and specific measures of lignin mineralization seldom employed in soils. Four‐day redox fluctuations increased the percent contribution of methoxyl C to soil respiration relative to static aerobic conditions, and cumulative methoxyl‐C mineralization was statistically equivalent under static aerobic and fluctuating redox conditions despite lower soil respiration in the latter treatment. Contributions of the less labile lignin Cβ to soil respiration were equivalent in the static aerobic and fluctuating redox treatments during periods of O2 exposure, and tended to decline during periods of O2 limitation, resulting in lower cumulative Cβ mineralization in the fluctuating treatment relative to the static aerobic treatment. However, cumulative mineralization of both the Cβ‐ and methoxyl‐labeled lignins nearly doubled in the fluctuating treatment relative to the static aerobic treatment when total lignin mineralization was normalized to total O2 exposure. Oxygen fluctuations are thought to be suboptimal for canonical lignin‐degrading microorganisms. However, O2 fluctuations drove substantial Fe reduction and oxidation, and reactive oxygen species generated during abiotic Fe oxidation might explain the elevated contribution of lignin to C mineralization. Iron redox cycling provides a potential mechanism for lignin depletion in soil organic matter. Couplings between soil moisture, redox fluctuations, and lignin breakdown provide a potential link between climate variability and the biochemical composition of soil organic matter.  相似文献

Ca3SiO4Cl2 co‐doped with Ce3+,Eu2+ was prepared by high temperature reaction. The structure, luminescent properties and the energy transfer process of Ca3SiO4Cl2: Ce3+,Eu2+ were investigated. Eu2+ ions can give enhanced green emission through Ce3+ → Eu2+ energy transfer in these phosphors. The green phosphor Ca2.9775SiO4Cl2:0.0045Ce3+,0.018Eu2+ showed intense green emission with broader excitation in the near‐ultraviolet light range. A green light‐emitting diode (LED) based on this phosphor was made, and bright green light from this green LED could be observed by the naked eye under 20 mA current excitation. Background

Heredity and environmental exposures may contribute to a predisposition to allergic rhinitis (AR). Autoimmunity may also involve into this pathologic process. FCRL3 (Fc receptor-like 3 gene), a novel immunoregulatory gene, has recently been reported to play a role in autoimmune diseases.

Objective

This study was performed to evaluate the potential association of FCRL3 polymorphisms with AR in a Chinese Han population.

Methods

Five single-nucleotide polymorphisms of FCRL3, rs945635, rs3761959, rs7522061, rs10489678 and rs7528684 were genotyped in 540 AR patients and 600 healthy controls using a PCR-restriction fragment length polymorphism assay. Allele, genotype and haplotype frequencies were compared between patients and controls using the χ2 test. The online software platform SHEsis was used to analyze their haplotypes.

Results

This study identified three strong risk SNPs rs7528684, rs10489678, rs7522061 and one weak risk SNP rs945635 of FCRL3 in Chinese Han AR patients. For rs7528684, a significantly increased prevalence of the AA genotype and A allele in AR patients was recorded. The frequency of the GG genotype and G allele of rs10489678 was markedly higher in AR patients than those in controls. For rs7522061, a higher frequency of the TT genotype, and a lower frequency of the CT genotype were found in AR patients. Concerning rs945635, a lower frequency of the CC genotype, and a higher frequency of G allele were observed in AR patients. According to the analysis of the three strong positive SNPs, the haplotype of AGT increased significantly in AR cases (AR = 38.8%, Controls = 24.3%, P = 8.29×10-14, OR [95% CI] 1.978 [1.652~2.368]).

Conclusions

This study found a significant association between the SNPs in FCRL3 gene and AR in Chinese Han patients. The results suggest these gene polymorphisms might be the autoimmunity risk for AR.  相似文献

Background and Objectives

Influenza A viruses cause highly contagious diseases in a variety of hosts, including humans and pigs. To develop a vaccine that can be broadly effective against genetically divergent strains of the virus, in this study we employed molecular breeding (DNA shuffling) technology to create a panel of chimeric HA genes.

Methods and Results

Each chimeric HA gene contained genetic elements from parental swine influenza A viruses that had a history of zoonotic transmission, and also from a 2009 pandemic virus. Each parental virus represents a major phylogenetic clade of influenza A H1N1 viruses. Nine shuffled HA constructs were initially screened for immunogenicity in mice by DNA immunization, and one chimeric HA (HA-129) was expressed on both a A/Puerto Rico/8/34 backbone with mutations associated with a live, attenuated phenotype (PR8LAIV-129) and a A/swine/Texas/4199-2/98 backbone (TX98-129). When delivered to mice, the PR8LAIV-129 induced antibodies against all four parental viruses, which was similar to the breadth of immunity observed when HA-129 was delivered as a DNA vaccine. This chimeric HA was then tested as a candidate vaccine in a nursery pig model, using inactivated TX98-129 virus as the backbone. The results demonstrate that pigs immunized with HA-129 developed antibodies against all four parental viruses, as well as additional primary swine H1N1 influenza virus field isolates.

Conclusion

This study established a platform for creating novel genes of influenza viruses using a molecular breeding approach, which will have important applications toward future development of broadly protective influenza virus vaccines.  相似文献

Life cycle assessment (LCA) was combined with primary data from nine forest harvesting operations in New York, Maine, Massachusetts, and Vermont, from 2013 to 2019 where forest biomass (FB) for bioenergy was one of several products. The objective was to conduct a data‐driven study of greenhouse gas emissions associated with FB feedstock harvesting operations in the Northeast United States. Deterministic and stochastic LCA models were built to simulate the current FB bioenergy feedstock supply chain in the Northeast US with a cradle‐to‐gate scope (forest harvest through roadside loading) and a functional unit of 1.0 Mg of green FB feedstock at a 50% moisture content. Baseline LCA, sensitivity analysis, and uncertainty analyses were conducted for three different FB feedstock types—dirty chips, clean chips, and grindings—enabling an empirically driven investigation of differences between feedstock types, individual harvesting process contributions, and literature comparisons. The baseline LCA average impacts were lower for grindings (4.57 kg CO2eq/Mg) and dirty chips (7.16 kg CO2eq/Mg) than for clean chips (23.99 kg CO2eq/Mg) under economic allocation, but impacts were of similar magnitude under mass allocation, ranging from 24.42 to 27.89 kg CO2eq/Mg. Uncertainty analysis showed a wider range of probable results under mass allocation compared to economic allocation. Sensitivity analysis revealed the impact of variations in the production masses and total economic values of primary products of forest harvests on the LCA results due to allocation of supply chain emissions. The high variability in fuel use between logging contractors also had a distinct influence on LCA results. The results of this study can aid decision‐makers in energy policy and guide emissions reductions efforts while informing future LCAs that expand the system boundary to regional FB energy pathways, including electricity generation, transportation fuels, pellets for heat, and combined heat and power.  相似文献

In this study, we investigated the influence of zearalenone (ZEA) on the dextran sulfate sodium (DSS)‐induced colitis model both in vitro and in vivo. Our results show that the mRNA levels of IL‐1β, IL‐18, NLRP3, ASC, and caspase‐1 in the DSS+ZEA‐treated group are lower than those in either the DSS or ZEA group, and the protein expression trends are similar. Furthermore, colitis, which is characterized by body weight loss, stool consistency, and the presence of bloody feces, was significantly alleviated in the DSS+ZEA group when compared with that in the DSS group. In addition, histological analysis showed that inflammatory cell infiltration and tissue damage of the colon in the DSS+ZEA group were recovered compared with that in the DSS‐treated group. These results suggest that, instead of aggravating DSS‐induced colitis, ZEA relieves the inflammatory reaction in colon tissue, which may be related to its estrogenic activity.  相似文献
