An integrated analysis of a terrestrial-aquatic ecosystem, the North Branch of the Moose River in the Adirondack region of New York, was conducted. This basin contains a large number of interconnected surface waters that exhibit marked gradients in pH and acid neutralizing capacity (ANC). As a result, the basin has been the focus of research activity, including the Regional Integrated Lake-Watershed Acidification Study (RILWAS). The objective of the current analysis was to use the North Branch of the Moose River as a case study to:
  1. Evaluate processes regulating the acid-base chemistry of surface waters.
  2. To assess the effects of surface water acidification on fish populations.
The observations of this study were consistent with the model of surface water acidification developed during the Integrated Lake-Watershed Acidification Study (ILWAS). The processes depicted in the original ILWAS simulation model were adequate to describe the acid-base chemistry of surface waters in the North Branch of the Moose River. However, the reduction of SO 4 2? in lake sediments, a process not represented in the original model, proved to be a significant source of acid neutralizing capacity (ANC) for some of these waters. As a result, reduction processes were added to the model. Analysis of in-situ bioassay and survey data indicate that acid-sensitive fish species have disappeared from the more acidic areas of the basin over the last half century. Paleoecological analyses indicate that pH has decreased from the high 5's to about 5 in Big Moose Lake during this period. ILWAS model simulations indicate that the pH of Big Moose Lake would increase by at least 0.1 to 0.5 pH units (depending on the season) in response to a 50% reduction in total atmospheric S deposition. Considerable variability in processes regulating acid/base chemistry was evident in the North Branch of the Moose River. Therefore, regional assessments of past or possible future effects of acidic deposition require widespread application of ILWAS theory within the Adirondack region and other potentially acid-sensitive areas.  相似文献

The ILWAS model has been enhanced for application to multiple-lake hydrologic basins. This version of the model has been applied to the Big Moose basin, which includes Big Moose Lake and its tributary streams, lakes, and watersheds. The basin, as defined, includes an area of 96 km2, with over 20 lakes and ponds, and 70 km of streams. Hydrologic and chemical calibrations have been made using data from seven sampling stations. When total atmospheric sulfur loading to the basin is halved, the model predicts, after four years of simulation, a decreasing sulfate concentration and to a lesser extent a rising alkalinity at Big Moose Lake outlet. At the end of four years, the results show an increase in pH of 0.1 to 0.5 pH units depending upon season.  相似文献

Intense interest in advanced squamous cell cancers of the head and neck (SCC of H&N) has resulted from the recent progress made in tumor responses with chemotherapy and radiotherapy. Unfortunately, the response patterns and clinical outcome of such patients are not adequately predicted on an individual patient basis using clinical parameters or conventional morphology. The study of flow cytometrically determined cellular parameters in such tumors is therefore of interest, but is hindered by inadequate tumor preparative technology. The previous article (10) in this journal describes the use of a murine SCC tumor, LC12, which was employed for comparative testing and determination of optimum techniques of preparation for this tumor. This report describes the application of these techniques to 144 specimens of human SCC of H&N. The mean total yield for these specimens is 7.4 X 10(7) cells/g of tissue. The mean viable enzymatic yield (3.3 X 10(7) cells/g) was higher than the mean viable mechanical yield (2.0 X 10(7) cells/g) except when lymph nodes were the source of the specimen (5.4 X 10(7) cells/g). The mean dye exclusion viability from enzymatically dissociated specimens were above 90%. Significant aneuploidal subpopulation losses were evident in mechanically dissociated and enucleated specimens. 65% of the enzymatically dissociated specimens were successfully cultured with a mean cloning efficiency of 2.1 X 10(-3). Preparative techniques derived from comparative testing with a murine standard tumor have been successfully applied to 144 specimens of SCC of H&N with resultant high yields and excellent viability. Technical problems detected during the preliminary testing with LC12 were confirmed in the human tumors.  相似文献

When detergent-derived photosystem II (PSII) membranes are treated with CaCl2 to remove the three extrinsic proteins associated with the O2-evolving complex, the resulting membranes (CaPSII) can still catalyze water oxidation if sufficient Ca2+ and Cl- are present. When CaPSII membranes are exposed to single turnover flashes on an O2 rate electrode, anomalous O2 is produced by the first two flashes. The addition of catalase to the membrane suspension completely inhibits O2 produced by the first two flashes, but not by subsequent flashes. Exogenous H2O2 stimulates anomalous O2 production by the first few flashes in CaPSII membranes, but not in control PSII membranes. Diuron (DCMU) does not inhibit H2O2-stimulated O2 production by the first flash. However, it does inhibit the O2 yield of all subsequent flashes, indicating that all flash-induced O2 signals in CaPSII membranes are dependent on photosystem II electron transport. H2O2 stimulation of O2 yields is inhibited in Tris-, heat-, and EDTA-(ethylenediaminetetraacetic acid)-treated CaPSII. In the presence of high salt, H2O2 (but not EDTA) treatment of CaPSII, extracts Mn functional in normal photosynthetic O2 evolution. The addition of exogenous Mn2+ reconstitutes anomalous O2 production in Tris-and H2O2/EDTA-treated CaPSII preparations but only in the presence of H2O2. Anomalous H2O2-stimulated O2 production can be observed both with a Clark electrode (steady state) and an O2 rate electrode (flash sequence). The mechanism involves electron donation from H2O2, mediated by free Mn2+, to PSII, and the 33-kDa extrinsic protein under some conditions can block this process. Since H2O2 can remove functional Mn from CaPSII membranes, its presence can convert functional Mn to the Mn2+ mediator state required for anomalous O2 production. We have cloned and characterized three distinct alpha-globin haplotypes obtained from inbred strains of the mouse, Mus domesticus. We report here the complete nucleotide sequence of the six alpha-globin genes that the haplotypes contain. Our analysis of these genes and those from one other previously described haplotype indicates that recurrent gene conversion events have played a major role in their history. The pattern of nucleotide substitutions suggests that conversions have occurred both within and between haplotypes. Limited segments of coding and noncoding DNA have been involved in these gene conversion events. In two of the haplotypes, the nonallelic genes of each maintain DNA sequence identity over discrete intervals and encode the same alpha-globin polypeptide. On the other hand, the coding regions of some genes have accumulated replacement changes that result in distinct alpha-globins. In one instance, these changes appear to reflect positive selection of advantageous mutations.  相似文献

Populations of the vernal cladoceranDaphniopsis ephemeralis are found in woodland ponds throughout southern Ontario. The species reproduces by cyclic parthenogenesis, and genotype frequencies at allozyme loci are ordinarily in good agreement with Hardy-Weinberg expectations. Occasional heterozygote deficiencies are apparently the consequence of admixture of ephippial hatchlings produced in temporally separated bouts of sexual reproduction. Considerable heterogeneity in genotypic frequencies exists among local populations in southwestern Ontario, indicating that gene flow among populations is restricted. Inbreeding coefficients suggest that populations receive an average of 0.3 migrants per generation. The completion of a sexual life cycle is made possible despite the brief persistence of populations by the emergence of males from ephippial eggs and by the production of equal numbers of male and female progeny in the first parthenogenetic brood.  相似文献

To understand the roles of individual amino acids in the folding and stability of globular proteins, a systematic structural analysis of mutants of the lysozyme of bacteriophage T4 has been undertaken. The isolation, characterization, crystallographic refinement and structural analysis of a temperature-sensitive lysozyme in which threonine 157 is replaced by isoleucine is reported here. This mutation reduces the temperature of the midpoint of the reversible thermal denaturation transition by 11 deg.C at pH 2.0. Electron density maps showing differences between the wild-type and mutant X-ray crystal structures have obvious features corresponding to the substitution of threonine 157 by isoleucine. There is little difference electron density in the remainder of the molecule, indicating that the structural changes are localized to the site of the mutation. High-resolution crystallographic refinement of the mutant lysozyme structure confirms that it is very similar to wild-type lysozyme. The largest conformational differences are in the gamma-carbon of residue 157 and in the side-chain of Asp159, which shift 1.0 A and 1.1 A, respectively. In the wild-type enzyme, the gamma-hydroxyl group of Thr157 participates in a network of hydrogen bonds. Substitution of Thr157 with an isoleucine disrupts this set of hydrogen bonds. A water molecule bound in the vicinity of Thr155 partially restores the hydrogen bond network in the mutant structure, but the buried main-chain amide of Asp159 is not near a hydrogen bond acceptor. This unsatisfied hydrogen-bonding potential is the most obvious reason for the reduction in stability of the temperature-sensitive mutant protein.  相似文献

Synopsis The silky shark, Carcharhinus falciformis, and scalloped hammerhead, Sphyrna lewini, represent >80% of the shark by-catch of the winter swordfish/tuna longline fishery of the northwestern Gulf of Mexico. This catch represents a potential supplemental fishery, yet little is known of the life histories of the two species. This report relates reproductive biology data to age and growth estimates for 135 C. falciformis and 78 S. lewini. Unlike other regional populations, C. falciformis in the Gulf of Mexico may have a seasonal 12 month gestation period. Males mature at 210–220 cm TL (6–7 yr); females at >225 cm TL (7–9 yr). Application of age at length data for combined sexes produced von Bertalanffy growth model parameter estimates of L = 291 cm TL, K = 0.153, t0 = −2.2 yr. Adult male S. lewini outnumbered adult females in catches because of differences in the distributions of the sexually segregated population. Males mature at 180 cm TL (10 yr); females at 250 cm TL (15 yr). von Bertalanffy parameter estimates for combined sexes of this species were L = 329 cm TL, K = 0.073, to = −2.2 yr.  相似文献

Chelation by citrate was found to promote the autoxidation of Fe2+, measured as the disapperance of 1,10-phenanthroline-chelatable Fe2+. The autoxidation of citrate---2+ could in turn promote the peroxidation of microsomal phospholipid liposomes, as judged by malondialdehyde formation. At low citrate---Fe2+ ratios the autoxidation of Fe2+ was slow and the formation of malondialdehyde was preceded by a lag phase. The lag phase evidence of this, linear initial rates of lipid peroxidation were obtained via the combination of citrate---Fe2+ and citrate---Fe3+, optimum activity occurring at a Fe3+---Fe2+ ratio of 1:1. Evidence is also presented to suggest that the superoxide and the hydrogen peroxide that are formed during the autoxidation of citrate---Fe2+ can either stimulate or inhibit lipid peroxidation by affecting the yield of citrate---Fe3+ from citrate---Fe2+. No evidence was obtained for the participation of the hydroxyl radical in the initiation of lipid peroxidation by citrate---Fe2+.  相似文献
