Group Y incompatibility and copy control of P1 prophage   总被引:4,自引:0,他引:4  
We have identified a restriction fragment (EcoRI-5) of bacteriophage P1 that, when cloned in a λ prophage, expresses incompatibility characteristic of the unit copy P1 plasmid prophage. Lysogens of λ-P1 chimeras in which the P1 fragment is EcoRI-5 fail to maintain P1 or P7 plasmids. In order to study the nature of this incompatibility, we isolated P1 mutants that overcome it. These mutants exhibit an elevated copy number. We provide evidence that the increased copy number results from a defect in a repressor of replication that can be furnished in trans by a chromosomally integrated P1, but not by EcoRI-5 itself. We, therefore, suggest that the incompatibility exerted by EcoRI-5 is not attributable to the represser of replication involved in the above copy control defect. Instead, it could be attributed to the presence of a DNA site required for proper plasmid partition at cell division. The elevated copy number of the P1 mutants would then enable them to compete favorably with the single copy of the cloned EcoRI fragment for a cellular component of the partition apparatus. Carbon isotope ratios (13C/12C) were measured for the leaves of the seagrass Thalassia testudinum Banks ex König and carbonates of shells collected at the seagrass beds from seven sites along the coast of southern Florida, U.S.A. The δ13C values of seagrass leaves ranged from −7.3 to −16.3‰ among different study sites, with a significantly lower mean value for seagrass leaves from those sites near mangrove forests (−12.8 ± 1.1‰) than those far from mangrove forests (−8.3 ± 0.9‰; P < 0.05). Furthermore, seagrass leaves from a shallow water area had significantly lower δ13C values than those found in a deep water area (P < 0.01). There was no significant variation in δ13C values between young and mature leaves (P = 0.59) or between the tip and base of a leaf blade (P = 0.46). Carbonates of shells also showed a significantly lower mean δ13C value in the mangrove areas (−2.3 ± 0.6‰) than in the non-mangrove areas (0.6 ± 0.3‰; P <0.025). In addition, the δ13C values of seagrass leaves were significantly correlated with those of shell carbonates (δ13C seagrass leaf = −9.1 + 1.3δ13C shell carbonate (R2 = 0.83, P < 0.01)). These results indicated that the input of carbon dioxide from the mineralization of mangrove detritus caused the variation in carbon isotope ratios of seagrass leaves among different sites in this study.  相似文献

The purpose of this paper is to assess the Georgetown University Family Center Biofeedback Clinic in its first 2 years of operation. The clinic used Budzynski's and Stoyva's training routines as well as family systems theory. Details of the treatment methods and the patients seen are described, along with an ongoing procedure developed to assess results. Preliminary results on 93 patients show that 80% report symptom improvement. Self-report of improvement is associated with decreases in frontal EMG levels and with changes in life-style.  相似文献

Caenorhabditis elegans male spicule morphogenesis requires the coordinated cellular behaviors of several types of cells. We found that the spicule neurons and sheath cells, although important for spicule function, are dispensable for spicule morphology. In contrast, the spicule socket cells are essential for both spicule elongation and formation of spicule cuticle. The socket cells are not only necessary but also sufficient to produce spicule cuticle. This functional aspect of socket cells is genetically separable from their function in mediating spicule elongation: elongated spicules with defective spicule cuticle can be formed. During spicule morphogenesis, the expression of an egl-17::GFP reporter gene is found in the spicule socket cells and its expression appears to be regulated in the socket cells. Mutants defective in TGF-beta signaling display a crumpled spicules phenotype as a result of failure of socket cell movement during spicule morphogenesis. These observations suggest that both the FGF and the TGF-beta signaling pathways might be involved in spicule elongation.  相似文献
