Reproduction in mice: The fate of spermatozoa not involved in fertilization

Mechanisms were sought through which the control of preimplantation mouse embryo development by spermatozoa might be effected. A potential route for the transmission of sperm-dependent stimuli to C3HeB/FeJ females was uncovered. It was found that within 24–48 hr after artificial insemination with spermatozoa, in which the DNA had been labeled with tritiated thymidine, a minimum of 9% of the radioactivity was transported across the uterine walls. It was deposited among the maternal tissues in a pattern that differed from the patterns of isotope distribution obtained when either free tritiated thymidine or Escherichia coli cells containing DNA labeled with tritiated thymidine were used instead of labeled sper-matozoa. In sperm-treated animals the ovaries, the adrenals, and a mesenteric lymph node exhibited strikingly large accumulations of radioactivity. The heart, spleen, and uterus manifested lesser accumulations of label, but were higher than liver, kidney, lung, brain, muscle, and intestine. The specific activity of the lymph node was found to decrease during the 12–72-hr period following insemination. This result led to the hypothesis that the lymphatic system could serve as a route for the dissemination, to maternal tissues, of radioactivity originally associated with spermatozoa deposited in the uterus. Heat-inactivated spermatozoa, which have the potential for facilitating the first cleavage of fertilized embryos, exhibited a distribution pattern indistinguishable from untreated spermatozoa. Sperm protein kinase was found to survive the heat inactivation of spermatozoa. This stability was interpreted as being compatible with the kinase functioning as an intermediary in the transmission of sperm-dependent stimuli that control preimplantation embryo development in mice.     

Killing power of the red imported fire ant [Hym.: Formicidae]: a key predator of the boll weevil [Col.: Curculionidae]

The red imported fire ant,Solenopsis invicta Buren, was identified as a key predator of the boll weevil,Anthonomus grandis Boheman. Although key factor analysis does not guarantee that the key factor is the causative agent, the evidence of boll weevil mortality was so clear that the fire ant could be identified as causing the mortality. Ant predation on immature boll weevils during the summer of 1981 averaged 84%. When this predation was combined with other natural mortality factors, the weevil population density throughout the season never exceeded levels that required control measures.     

LATICIFERS IN THE BULB SCALES OF ALLIUM

The structure and distribution of laticifers has been studied in the bulb scales of 52 species and varieties of Allium. Differences in laticifer position, shape, and pitting in the cross wall of the observed specimens support Traub's separation of the subgenera Amerallium and Allium within the genus Allium. If the species examined are representative, anatomical evidence also justifies the distinction of the section Melanocrommyum and the subsection Cepa.     

Determination of nicotinamide N-oxide—a human excretory product

A method for the determination of nicotinamide N-oxide has been developed. It is based on the ability of the N-oxide to function as an electron acceptor in the xanthine oxidase catalyzed oxidation of xanthine. In simple mixtures the N-oxide can be converted quantitatively to nicotinamide and the latter determined by the cyanogen bromide method. The conversion is not always quantitative in complex mixtures, such as urine; an isotope dilution variation on the basic method permits the determination of the N-oxide in such situations. The basic method is applicable over the range 0.02–0.3 μmole of nicotinamide N-oxide.The new method has been used to verify the prominent excretory role of nicotinamide N-oxide in rodents. Application of the method to a study of human urines has permitted the detection of the N-oxide as an excretory metabolite in man. Only vanishingly small quantities of the N-oxide are excreted under normal conditions. However after the ingestion of 200 mg of nicotinamide, significant quantities of the N-oxide are detectable in human urine. Urine samples obtained from a number of other mammalian species contained little or no detectable nicotinamide N-oxide.     

Isolation of Cryptosporidium oocysts and sporozoites using discontinuous sucrose and isopycnic Percoll gradients

Techniques for the large-scale isolation of Cryptosporidium oocysts and sporozoites, obtained from the feces of experimentally infected Holstein calves, were developed employing discontinuous sucrose gradients and isopycnic Percoll gradients. The oocyst recovery method utilized 2 sequential discontinuous sucrose gradients followed by 1 Percoll gradient. Recovered oocysts were essentially free of debris and bacteria and represented 34% of the original oocyst suspension. Sporozoites were recovered from excystation mixtures on a single Percoll gradient. Sixty-three percent of the original sporozoites were recovered with 2.2% contamination by intact oocysts and virtually no oocyst walls.     

COMPARATIVE MORPHOLOGY OF THE CARPEL IN THE ROSACEAE. IV. POMOIDEAE: CHAMAEMELES,COTONEASTER, DICHOTOMANTHES,PYRACANTHA

The carpels of Chamaemeles, Cotoneaster, Dichotomanthes, and Pyracantha tend to be separate from one another, their sutures tend to be closed, and they become more or less bony at maturity. However, aside from having collaterally placed ovules, they do not appear to be structurally similar. There seem to be 2 different evolutionary trends in the ovular bundle–wing bundle relationship: in Pyracantha, progressive fusion between the ovular bundle and the wing bundle has led to the formation of a “ventral” bundle; in Cotoneaster, and possibly Chamaemeles, the wing bundle has become reduced and rather attenuated. A primitive pomoid state may be represented by the carpel of Dichotomanthes, which is completely free of the floral cup and in which wing and ovular bundles are separate. Differences in sutural closure appear only in Cotoneaster, and in species of that genus the wing bundles and ovular bundles tend to be fused if the suture is closed, and separate if it is open.     

Production of 2,3-Butylene Glycol from Citrus Wastes. II. The Bacillus polymyxa Fermentation

The conditions required for production of levo 2,3-butylene glycol by Bacillus polymyxa from citrus molasses were studied. Starter cultures required acclimatization to the substrate prior to inoculation of the fermentation medium. Maximal production of butylene glycol with minimal residual sugar was obtained with a medium consisting of molasses, diluted to 20 degrees Brix, and 0.4% urea. Optimal environmental conditions included aeration at 0.11 volumes of air per volume of medium per minute, maintenance of pH at 6.0 to 6.2, a fermentation temperature of 30 C, and a stirring rate of 420 rev/min. The concentration of butylene glycol obtained in the fermentation beer ranged from 2.3 to 4.4%. The optical rotation of the glycol ranged from [alpha](D) (23 degrees ) = -1.01 degrees to -10.45 degrees . The variation in rotation was probably due to the presence of contaminating substances in the distillate.
