The pleiotropic effects of Calloselasma rhodostoma venom is caused by various toxins, among them kistrin and ancrod, which block platelet activation triggered by RGD-dependent integrins and the blood clotting cascade, respectively. Here, we demonstrate that rhodocetin, another component of this venom, acts as alpha2beta1 integrin inhibiting disintegrin and antagonizes important cellular responses to type I collagen. Cell adhesion, migration, and collagen lattice contraction in vitro were specifically inhibited by rhodocetin, whereas expression of collagen-degrading matrix metalloproteases was differently modulated. Moreover, cell invasion of HT1080 fibrosarcoma cells into a type I collagen matrix, but not into a fibrin gel or a basement membrane-extracted matrigel was efficiently blocked by rhodocetin. Unlike its natural ligand collagen, rhodocetin failed to cluster alpha2beta1 integrin, despite similar binding affinities. Hence, in the absence of focal adhesions cells do not attach firmly to rhodocetin and do not respond with any of alpha2beta1-triggered cell reactions, except for MMP-1 production. Therefore, this disintegrin may be a valuable tool to specifically target stromal tumor invasion and to manipulate other alpha2beta1 integrin-mediated functions, such as excessive scar contraction and fibrosis. Rhodocetin might be therapeutically useful because of its lack of interference with RGD-dependent integrins, low molecular mass, high solubility, and biochemical stability.  相似文献

In the humid forest zone of Southern Cameroon, farmers generally associate cocoa with native and exotic trees in complex agroforestry systems. Despite the socio-economic and ecological importance of these systems, few studies have investigated their plant composition. We investigated tree composition of cocoa agroforests along a gradient of market access, population density and resource use intensity in the humid forest zone of southern Cameroon, comprising (i) the sub-region of Yaoundé, (ii) the sub-region of Mbalmayo, and (iii) the sub-region of Ebolowa. Market access, population density and resource use intensity all decreased from the first to the third sub-region. We quantified the diversity of tree species associated with cocoa within individual agroforests, among agroforests in the same region, and among the three sub-regions, and classified the tree species according to their main uses. A total of 9.1 ha belonging to 60 cocoa agroforests were inventoried in 12 villages. We encountered a total of 206 tree species with an average of 21 tree species per agroforest. In the more urbanized area around Yaoundé, agroforests were less diverse than in the other sub-regions. In all of the agroforests, food producing tree species tended to be more frequent than other species. Two thirds of the food trees were native forest species and one third was introduced. From Ebolowa to Yaoundé, the density of food producing trees doubled and the density of exotic food-producing species increased relative to native species. Some local species producing high-value non-timber forest products were found in the agroforests, but their density was far lower than that of exotic tree species. The agroforests also provide medicine, charcoal and other products for household consumption and sale. We conclude that unless there are specific efforts to promote local forest tree species in cocoa agroforests, these will progressively lose importance with increasing market access, population pressure and land use intensity.  相似文献

Prostaglandin-E2 (PGE2) is known to trigger suicidal death of nucleated cells (apoptosis) and enucleated erythrocytes (eryptosis). In erythrocytes PGE2 induced suicidal cell death involves activation of nonselective cation channels leading to Ca2+ entry followed by cell shrinkage and triggering of Ca2+ sensitive cell membrane scrambling with phosphatidylserine (PS) exposure at the cell surface. The present study was performed to explore whether PGE2 induces apoptosis of nucleated cells similarly through cation channel activation and to possibly disclose the molecular identity of the cation channels involved. To this end, Ca2+ activity was estimated from Fluo3 fluorescence, mitochondrial potential from DePsipher fluorescence, phosphatidylserine exposure from annexin binding, caspase activation from caspAce fluorescence, cell volume from FACS forward scatter, and DNA fragmentation utilizing a photometric enzyme immunoassay. Stimulation of K562 human leukaemia cells with PGE2 (50 microM) increased cytosolic Ca2+ activity, decreased forward scatter, depolarized the mitochondrial potential, increased annexin binding, led to caspase activation and resulted in DNA fragmentation. Gene silencing of the Ca2+-permeable transient receptor potential cation channel TRPC7 significantly blunted PGE2-induced triggering of PS exposure and DNA fragmentation. In conclusion, K562 cells express Ca2+-permeable TRPC7 channels, which are activated by PGE2 and participate in the triggering of apoptosis.  相似文献

DIDS (4,4'-diisothiocyanato-stilbene-2,2'-disulfonic acid) is a commonly used blocker of plasma membrane anion channels and transporters. We observed that DIDS undergoes decomposition while stored in DMSO (dimethyl sulfoxide) forming a biologically active compound. One decomposition product, called IADS, was identified and synthesized. Voltage-clamp and patch clamp experiments on Xenopus laevis oocytes and human erythrocytes revealed that IADS is able to activate a plasma membrane cation conductance in both cell types. Furthermore, we found that IADS induces hemolysis in red blood cells of healthy donors but fails to hemolyze erythrocytes of donors with cystic fibrosis. Thus, IADS stimulated activation of a cation conductance could form the basis for a novel diagnostic test of cystic fibrosis.  相似文献

The Degradation Gradient Method (DGM) is a sophisticated technique for the assessment of range condition. It applies multivariate analyses of herbaceous species data to detect subtle degrees of overgrazing. The suitability of this multivariate method was tested in the central Highland Savanna of Namibia by comparing its results against a univariate analysis of herbaceous data in a simple but robust Range‐Unit Model. Despite aridity and topographical heterogeneity, the DGM performed unexpectedly well under these conditions. The relative instability of this dry savanna system favoured the applicability of the DGM by promoting a clear grazing gradient. Using species density data only resulted in an incorrect outcome of the multivariate analysis. The sensitivity of the DGM could be improved by combining density and cover data.  相似文献

Mutations in the human ClC-2 Cl(-) channel have been described to influence its function dramatically. To test for naturally occurring gene variants in a human population and their functionality, all 24 CLCN2 exons from a Central African population were sequenced. Six single amino acid exchanges in the intracellular N-terminus (P48R, R68H), in the pore domain (G199A), or in the intracellular C-terminus (R646Q, R725W, R747H) were identified at low frequency. Heterologous expression of these polymorphisms in Xenopus laevis oocytes demonstrated their functional significance as determined by two-electrode voltage-clamp. The polymorphisms R68H, R725W, and R747H exhibited faster voltage-stimulated gating as compared to the wild type channel, resulting in higher steady state currents of R725W. Probably due to decreased surface expression P48R, R68H, and R646Q mutants generated lower currents than the wild type channels. The inward currents of the mutated channels R725W, R747H, and G199A failed to increase during hypotonic swelling, a defect paralleled by impaired swelling-accelerated voltage-gating in one mutant (G199A). In conclusion, the Africans' gene pool comprises CLCN2 gene variants in the N-terminus, the C-terminus or the pore domain that affect surface expression and voltage- or cell-swelling-stimulated channel gating.  相似文献
