Electrorotation—the spin of cells in rotating high frequency electric fields—has been used to investigate properties of human peripheral blood lymphocytes. The rotation spectra of lymphocytes deviate from those of single shell spheres. The deviations are caused by the electrical properties of the nucleus in the cell interior.Electrorotation allows the distinction between successfully stimulated lymphocytes and unstimulated cells after application of concanavalin A. Notwithstanding the fact that only a proportion of the cells will be mitogenically stimulated we detected an enhanced cell membrane conductivity for the whole cell population immediately after the addition of mitogen.  相似文献

Selective enrichment culture techniques were employed to obtain mixed cultures of methanogenic rods and sarcina from surface flooding waters and deep subsurface (~1650 m) oil-bearing sedimentary rocks and formation waters sampled from an old oil field in the U.S.S.R. previously reported to display active biological methanogenesis. The methanogens were selectively isolated as colonies on agar petri dishes that were incubated in a novel container. The general cellular and growth features of three Methanobacterium isolates were determined. These strains grew optimally at 37 to 45°C in anaerobic pressure tube cultures with a doubling time of 16 to 18 h on H2-CO2 and proliferated as autotrophs. Acetate addition significantly enhanced the final cell yield. Growth of these strains was completely inhibited by either 0.6 g of sodium sulfide per liter or 31.0 of sodium chloride per liter, but growth was not inhibited by either 0.3 g of sodium sulfide per liter or 1.0 g of sodium sulfate per liter. One novel isolate, Methanobacterium sp. strain ivanov, was grown on H2-CO2, and the stable-carbon isotopic fractionations that occurred during synthesis of methane, cell carbon, and lipids were determined. The results of this study were used to examine the anomalous relationship between the isotopic and chemical compositions of natural gas occurring in the deep subsurface environment of the oil field.  相似文献

In this first of a series of papers concerning the theoretical analysis of rate theory models for ion transport through rigid pores, the case of vanishing interactions is investigated. "Rigidity" means that ions crossing membranes through pores see a fixed structure of the pores, not changing in time. A single pore is considered to be a sequence of (n + 1) activation barriers separated by n energy minima. The explicit analytical treatment is restricted to pores with regular internal barrier structure, including the nonequilibrium situation of an applied electric field. In this case the connection with continuum diffusion models is demonstrated by performing in the limit n leads to infinity (n = number of binding sites within the pores) the transition to continuum. Thus, from diffusion equations describing a discrete number of jumps, the corresponding diffusion-like partial differential equations and boundary conditions are generated. For regular pores, from the time dependent solutions of the discrete equations, the corresponding solutions of the continuum equations are explicitly generated. The time-dependent relaxation behaviour of the discrete model is in good agreement with the continuum model if one assumes more than two binding sites in the pores.  相似文献

Wavelength-selective absorption of solar radiation within plant leaves allows penetration of visible radiation (400–700 nm) to the chloroplasts, while removing much of the damaging ultraviolet-B (UV-B, 280–320 nm) radiation. Flavonoids are important in this wavelength-selective absorption. Induction of flavonoid synthesis by solar radiation, and specifically by UV-B radiation, is discussed as this relates to the potential acclimation of plants to enhanced solar UV-B radiation that would result from stratospheric ozone reduction.  相似文献

A study of the fine structure of Caenorhabditis briggsae revealed several features not previously described in free-living nematodes. These were: chambered walls of the stoma, zonula adherens in the esophagus, daedaloid folds in the inner surface of the uterus and openings in the terminal web of the intestinal epithelium. When an excised corn (Zea mays) root pretreated with chloride was exposed for 10 minutes to pulse labeling with 30Cl and then transferred to unlabeled chloride, the activity in the xylem exudate reached a maximum about 4 minutes after pulse labeling was discontinued and then declined sharply. The rate at which labeled chloride was transported across the root into the xylem and basipetally therein was on the order of 75 to 250 centimeters per hour. Consequently, symplasmic movement of chloride in corn roots is fast and may not be rate-limiting in transfer from the root surface to the xylem. Experiments on pulse labeling with 22Na gave similar results. A large fraction of the absorbed 22Na was not translocated into the exudate but was tightly sequestered in a cell compartment, probably the vacuole.  相似文献

13C/12C ratios have been determined for plant tissue from 104 species representing 60 families. Higher plants fall into two categories, those with low δPDBI13C values (—24 to —34‰) and those with high δ 13C values (—6 to —19‰). Algae have δ 13C values of —12 to —23‰. Photosynthetic fractionation leading to such values is discussed.  相似文献

Under certain conditions of preparation, DNA, whether free or complexed with polylysine or histone KAP (I, fl), produce huge negative circular dichroism (CD) spectra with maxima at about 270nm. In order to investigate the cause of these spectra, reconstituted polylysine-DNA complex was used as a model system. It was found that the CD change of DNA in the complex is not a linear function of the fraction of base pairs bound. Such a CD spectrum is not changed despite dilution up to 128 folds for as long as 12 hours. Difference CD spectra taken between free DNA and any of the complexes are qualitatively the same, and are similar to those of free DNA and nucleohistone KAP (Fasman et al., Biochemistry 9, 2814-2822, 1970), free DNA and direct mixed polylysine-DNA complexes, or free DNA in high salt (Chang et al., Biochemistry12, 3028-3032, 1973). The suggestion is made that this CD spectrum might be caused by specific conformational changes in DNA, perhaps belonging to the family of B to C transitions followed by a further structural distortion of DNA due to aggregation of the nucleoprotein molecules.  相似文献
