Molecular species of epidermal growth factor carrying immunosuppressive activity

The suppression of antibody formation to sheep red cells in mice by partially purified fractions of mouse submaxillary gland was shown to be caused by epidermal growth factor (EGF). Purification of EGF by the method of Savage and Cohen resolved three components referred to as EGF a, EGF b, and EGF c. All three induced premature eye opening in neonatal mice, but only EGF a (identified as EGF 1-53) had full immunosuppressive activity. EGF c was shown by micropeptide mapping of chymotryptic and thermolytic digests and amino-terminal analysis to differ from EGF a only by the presence of beta-aspartyl instead of an asparaginyl residue. EGF b differed from EGF a in that it lacked the N-terminal asparagine. EGF shortened enzymatically at its carboxy terminal by two or five amino acids did not have any immunosuppressive activity. These findings suggest that, in contrast to some other biological effects of EGF, intact amino and carboxy terminals are required for the expression of immunosuppressive activity.     

Analysis of the HLA-ABC linkage disequilibrium: Decreasing strength of gametic association with increasing map distance

Summary 1242 HLA-ABC haplotypes of the North German population (Hamburg) as deduced by family analyses are described. They are in perfect agreement with recently published data by Mayr (1977) from Austria (Vienna) in all parameters tested: frequency of the single HLA-alleles, haplotype distribution and linkage disequilibrium values. Gametic association studies revealed that 69.4% of the B and C genes (map distance 0.2 cM) 36.9% of the A and C genes (0.6 cM), but only 23.2% of the A and B genes (0.8 cM) were significantly more often combined than expected due to their frequencies. From these findings it seems likely that the linkage disequilibrium within the MHC is rather due to a short evolutionary period than to selective forces. Some observations as to the most common European haplotype A1,B8 are discussed.     

Influence of N and K fertilization and growth temperature on 13C/12C ratios of timothy (Phleum pratense L.)

Summary Measurements have been reported of carbon isotope ratios of timothy grown at different temperatures and with varying nitrogen and potassium supplies. Both total plant tissue and extracted plant tissue have been analyzed. The ¹³C/¹²C ratios were found to vary both with temperature and with nutrient level; the highest values of ¹³C were found under the most optimum growth conditions.     

The transition of the thick ascending limb of Henle's loop into the distal convoluted tubule in the nephron of the rat kidney

Summary Examination of serial semithin sections of rat kidney cortex and a subsequent electron microscopic study of selected areas revealed that the characteristic epithelium of the cortical part of the thick ascending limb of Henle extends for a varying distance beyond the macula densa. The transition from the relatively thin epithelium of the thick ascending limb at this site to the three -or even four-fold thicker epithelium of the convoluted part of the distal tubule is sharply defined and occurs without the interposition of an intermediate cell type.The position of the macula densa at the end but still clearly within the ascending limb of Henle's loop is functionally interpreted to guarantee the separation of the sensor point macula densa from disturbing influences which might arise from the secretory activity of the subsequent tubular portion.Investigations supported by the Deutsche Forschungsgemeinschaft. Inhibition of compensatory kidney hypertrophy by hypothyroidism in the rat. Measurement of the mean diameter of glomeruli and proximal tubules]

The body, heart and kidney weights are reduced in the hypothyroid rat. In this animal, the diameter of the proximal tubule is significantly smaller than in euthyroid controls. The glomerular diameter is not affected by hypothyroidism. 21 days after uninephrectomy, the weight of the remaining kidney and the diameter of the proximal tubule increase significantly both in the hypothyroid and the euthyroid rats. But this compensatory renal hyperthrophy is definitely impaired in the hypothyroid animals.     

Cycloamylose-accelerated cleavage of acylimidazoles

Hydrolyses of N-trans-cinnamoylimidazole (1) and N-acetylimidazole (2) were accelerated by cyclohexaamylose (α-CA) and cycloheptaamylose (β-CA) at 25°C. The cleavage of the amide bond in 1 at pH 9.0 was accelerated by α-CA and β-CA by 28- and 38-fold, respectively, whereas the cleavage of the amide bond in 2 at pH 7.0 was accelerated by α-CA and β-CA by 50- and 28-fold, respectively. The β-CA-accelerated hydrolysis of 1 proceeded via binding, acylation of β-CA, and deacylation of β-CA trans-cinnamate, which is consistent with the pathway used by serine proteases. The deuterium oxide solvent isotope effects for acylation and deacylation steps indicate nucleophilic attack in acylation and general basic attack in deacylation. The present finding of the acceleration by cycloamyloses in the cleavages of amide bonds in 1 and 2 indicates that cycloamyloses are an excellent model for hydrolytic enzymes.     

Germination dimorphism and developmental flexibility in the ruderal weed Heterotheca grandiflora

Summary Heterotheca grandiflora Nutt. (Asteraceae, tribe Astereae) is one of the few native Californian plant species increasing its range as a weed. The production of dimorphic seed, together with flexible development in either the annual or biennial habit, may contribute to its range expansion. Well dispersed disc achenes germinate rapidly to high percentages while poorly dispersed ray achenes show considerable dormancy, germinating at a much lower rate to lower final percentages. Ray achenes appear more sensitive to environmental factors and have more specific germination requirements than do disc achenes. Thus, germination is distributed in space and time. Plants growing as annuals have one flowering period while those acting as biennials may flower up to three times, although seed production differs greatly among the stages. These factors aid in forming the general purpose genotype so frequently encountered in weedy species.     

T cell receptor/CD3-signaling induces death by apoptosis in human T cell receptor gamma delta + T cells.

mAb directed against the TCR/CD3 complex activate resting T cells. However, TCR/CD3 signaling induces death by apoptosis in immature (CD4+CD8+) murine thymocytes and certain transformed leukemic T cell lines. Here we show that anti-TCR and anti-CD3 mAb induce growth arrest of cloned TCR-gamma delta + T cells in the presence of IL-2. In the absence of exogenous IL-2, however, the very same anti-TCR/CD3 mAb stimulated gamma delta (+)-clones to proliferation and IL-2 production. In the presence of exogenous IL-2, anti-TCR/CD3 mAb induced the degradation of DNA into oligosomal bands of approximately 200 bp length in cloned gamma delta + T cells. This pattern of DNA fragmentation is characteristic for the programmed cell death termed apoptosis. These results demonstrate that TCR/CD3 signaling can induce cell death in cloned gamma delta + T cells. In addition, this report is the first to show that apoptosis triggered by TCR/CD3 signaling is not restricted to CD4+CD8+ immature thymocytes and transformed leukemic T cell lines but can be also observed with IL-2-dependent normal (i.e., TCR-gamma delta +) T cells.
