Optimization of the process of enzymatic hydrolysis of keratin-containing stock aimed at obtaining hydrolysates of high biological value has been performed. The increasing of the stock/water weight ratio, the amount of the alkaline protease preparation from Acremonium chrysogenium added and the temperature of the reaction mixture resulted in an increase in the yield and antioxidant capacity of hydrolysis products. The molecular masses of soluble products obtained under optimal hydrolysis conditions ranged from 3.55 to 3.60 kDa. High antioxidant capacity, 100% bioavailability and a well-balanced amino acid composition was characteristic of the hydrolysis products.  相似文献

Various ratios of nutrient forms have been analyzed on the basis of the materials collected in Rybinsk Reservoir in the years 2001 to 2011. The analysis was used for assessment of the modern state of the reservoir. The total nitrogen (TN) to total phosphorus (TP) ratio is the most informative parameter. During the study period at all sampling stations, the average value of this ratio was 17.0. This value indicates the lack of limitation of development of phytoplankton by these nutrients. High values of the absolute concentration of nitrate nitrogen and its proportion in the total mineral nitrogen (which are important indicators of increasing eutrophication), as well as the percentage of inorganic phosphorus, were revealed in Volzhskii reach and the upper part of Sheksninskii reach of the reservoir. These values indicate the eutrophic state of the above areas of the reservoir.  相似文献

Two new pesticide preparations of cyclophos and alvison, a number of their components, two alvison metabolites have been studied to reveal their cytogenetic activity. According to the investigation in vivo the mutagenic effect of cyclophos manifests itself only with toxic doses. Professional contacts with the pesticide don't induce chromosome mutation in human peripheral lymphocytes. The alvison preparations generate chromosome mutations in somatic cells of people and animals. The components and metabolites of alvison have shown no visible genetic activity during in vivo and in vitro experiments.  相似文献

The frequency of recombination exchanges per unit length of DNA (Freuld) can be estimated by measuring the scale of the genetic map that is the mean statistical distance between two neighboring crossovers. The scales appear to be equal for the alternative pathways of recombination, RecBCD (wild-type cells) or RecF (recBC- sbcB- sbcC- genotypes). The absolute value of the scale depends on specific experimental conditions. recR, recQ, ruv, recJ and recN genes of the RecF pathway of recombination (recBC- sbcBC- cell genotypes) do not appear to be silent in wild-type cells where the RecBCD pathway predominates. On the contrary, these genes are responsible for the Freuld. The list recF504::Kmr greater than recQ61::Tn3 greater than ruv-54 greater than recJ284::Tn10 shows decreasing efficiency in inhibiting recombination exchanges by these mutations. The recN264 mutation gives a small, but opposite effect of increasing the frequency of recombination exchanges. The effect of the recF and recQ mutations appears to be additive, but that is not the case in combinations of ruv-54 with recF504::Kmr or recQ61::Tn3.  相似文献

Fungal Decomposition of Oat Straw during Liquid and Solid-State Fermentation   总被引:10,自引:0,他引:10  
White rot fungi (Coriolus hirsutus, Coriolus zonatus, and Cerrena maxima from the collection of the Komarov Botanical Institute of the Russian Academy of Sciences) and filamentous fungi (Mycelia sterilia INBI 2-26 and Trichoderma reesei6/16) were grown on oat straw–based liquid and solid media, as well as in a bench-scale reactor, either individually or as cocultures. All fungi grew well on solid agar medium supplemented with powdered oat straw as the sole carbon source. Under these conditions, the mold Trichoderma reesei fully suppressed the growth of all basidiomycetes studied; conversely,Mycelia sterilia neither affected the development of any of the cultures, nor did it show any substantial susceptibility to suppression by their presence. Pure solid cultures of basidiomycetes, as well as the coculture of Coriolus hirsutus andCerrena maxima,caused a notable bleaching of the oat straw during its consumption. When grown on the surface of oat straw–based liquid medium, the basidiomycetes consumed up to 40% of the polysaccharides without measurable lignin degradation (a concomitant process). Under these conditions, Mycelia sterilia decomposed no more than 25% of the lignin in 60 days, but this was observed only after polysaccharide exhaustion and biomass accumulation. In contrast, during solid-state straw fermentation, white rot fungi consumed up to 75% of cellulose and 55% of lignin in 83 days (C. zonarus), whereas the corresponding consumption levels for cocultures ofMycelia sterilia and Trichoderma reesei equaled 70 and 45%, respectively (total loss of dry weight ranged from 55 to 60%). Carbon dioxide–monitored solid-state fermentation of oat straw by the coculture of filamentous fungi was successfully performed in an aerated bench-scale reactor.  相似文献

Laccases of the basidiomycetes Cerrena unicolor 059, C. unicolor 0784, and Pleurotus oastreatus 0432 were subjected to a comparative study. The enzymes were isolated as homogeneous preparations with molecular weights of 55, 56, and 57 kD, respectively. The three enzymes were found to be glycoproteins. The carbohydrate moiety of the glycoproteins included mannose, galactose, and N-acetylglucosamine. The carbohydrate moieties of the laccases from C. unicolor 059, C. unicolor 0784, and P. oastreatus 0432 accounted for 17, 23, and 24%, respectively. The pH optima of the enzymes corresponded to 4.0, 3.75, and 5.6, respectively. Thermal stability tests (carried out at 40°C) demonstrated that the laccase of C. unicolor 0784 was characterized by the highest temperature resistance (the enzyme retained 25% activity after 172 h of incubation). The values of the Michaelis constant (K M) were determined for the reactions of oxidation of pyrocatechol, hydroquinone, and potassium ferrocyanide catalyzed by the laccases of the basidiomycetes.  相似文献
