Attomole (10(-18)mol) levels of RNA and DNA isolated from beer spoilage bacterial cells Lactobacillus brevis have been detected by the electrochemical sandwich DNA hybridization assay exploiting enzymatic activity of lipase. DNA sequences specific exclusively to L. brevis DNA and RNA were selected and used for probe and target DNA design. The assay employs magnetic beads (MB) modified with a capture DNA sequence and a reporter DNA probe labeled with the enzyme, both made to be highly specific for L. brevis DNA. Lipase-labeled DNAs captured on MBs in the sandwich assay were collected on gold electrodes modified with a ferrocene (Fc)-terminated SAM formed by aliphatic esters. Lipase hydrolysis of the ester bond released a fraction of the Fc redox active groups from the electrode surface, decreasing the electrochemical signal from the surface-confined Fc. The assay, shown to be efficient for analysis of short synthetic DNA sequences, was ineffective with genomic double stranded bacterial DNA, but it allowed down to 16 amole detection of 1563 nts long RNA, isolated from bacterial ribosomes without the need for PCR amplification, and single DNA strands produced from ribosomal RNA. No interference from E. coli RNA was registered. The assay allowed analysis of 400 L. brevis cells isolated from 1L of beer, which fits the "alarm signal" range (from 1 to 100 cells per 100mL).  相似文献

Endothelial cells release prostacyclin (PGI2) and nitric oxide (NO) to inhibit platelet functions. PGI2 and NO effects are mediated by cyclic nucleotides, cAMP- and cGMP-dependent protein kinases (PKA, PKG), and largely unknown PKA and PKG substrate proteins. The small G-protein Rac1 plays a key role in platelets and was suggested to be a target of cyclic nucleotide signaling. We confirm that PKA and PKG activation reduces Rac1-GTP levels. Screening for potential mediators of this effect resulted in the identification of the Rac1-specific GTPase-activating protein ARHGAP17 and the guanine nucleotide exchange factor ARHGEF6 as new PKA and PKG substrates in platelets. We mapped the PKA/PKG phosphorylation sites to serine 702 on ARHGAP17 using Phos-tag gels and to serine 684 on ARHGEF6. We show that ARHGAP17 binds to the actin-regulating CIP4 protein in platelets and that Ser-702 phosphorylation interferes with this interaction. Reduced CIP4 binding results in enhanced inhibition of cell migration by ARHGAP17. Furthermore, we show that ARHGEF6 is constitutively linked to GIT1, a GAP of Arf family small G proteins, and that ARHGEF6 phosphorylation enables binding of the 14-3-3 adaptor protein to the ARHGEF6/GIT1 complex. PKA and PKG induced rearrangement of ARHGAP17- and ARHGEF6-associated protein complexes might contribute to Rac1 regulation and platelet inhibition.  相似文献

Sibirotitan astrosacralis nov. gen., nov. sp., is described based on isolated but possibly associated cervical and dorsal vertebrae, sacrum, and previously published pedal elements from the Lower Cretaceous (Barremian?) Ilek Formation at Shestakovo 1 locality (Kemerovo Province, Western Siberia, Russia). Some isolated sauropod teeth from the Shestakovo 1 locality are referred to the same taxon. The phylogenetic parsimony analyses place Sibirotitan astrosacralis nov. gen., nov. sp., as a non-titanosaurian somphospondyl titanosauriform. The new taxon exhibits four titanosauriform and one somphospondylan synapomorphies, and one autapomorphy – a hyposphene ridge that extends between the neural canal and the postzygapophyses. It differs from all other Somphospondyli by having only five sacral vertebrae. The new taxon shares with Euhelopus and Epachtosaurus sacral ribs that converge towards the middle of the sacrum in dorsal view. Sibirotitan astrosacralis nov. gen., nov. sp., is only the second sauropod taxon from Russia and one of the oldest titanosauriform described so far in Asia.  相似文献

BACKGROUND: Alterations in maternal folate and homocysteine metabolism are associated with neural tube defects (NTDs). The role played by specific micronutrients and metabolites in the causal pathway leading to NTDs is not fully understood. METHODS: We conducted a case-control study to investigate the association between NTDs and maternal alterations in plasma micronutrients and metabolites in two metabolic pathways: methionine remethylation and glutathione transsulfuration. Biomarkers were measured in a population-based sample of women who had NTD-affected pregnancies (n = 43) and a control group of women who had a pregnancy unaffected by a birth defect (n = 160). We compared plasma concentrations of folate, vitamin B(12), vitamin B(6), methionine, S-adenosylmethionine (SAM), s-adenosylhomocysteine (SAH), adenosine, homocysteine, cysteine, and reduced and oxidized glutathione between cases and controls after adjusting for lifestyle and sociodemographic factors. RESULTS: Women with NTD-affected pregnancies had significantly higher plasma concentrations of SAH (29.12 vs. 23.13 nmol/liter, P = .0011), adenosine (0.323 vs. 0.255 mumol/liter; P = .0269), homocysteine (9.40 vs. 7.56 micromol/liter; P < .001), and oxidized glutathione (0.379 vs. 0.262 micromol/liter; P = .0001), but lower plasma SAM concentrations (78.99 vs. 83.16 nmol/liter; P = .0172) than controls. This metabolic profile is consistent with reduced methylation capacity and increased oxidative stress in women with affected pregnancies. CONCLUSIONS: Increased maternal oxidative stress and decreased methylation capacity may contribute to the occurrence of NTDs. Further analysis of relevant genetic and environmental factors is required to define the basis for these observed alterations.  相似文献

Formation of noncovalent complexes between tyrosinase from mushrooms and a cationic polyelectrolyte, polybrene (PB, poly (1,5-dimethyl-1,5-diazaundecamethyelene) bromide), was shown to activate and stabilize tyrosinase in water-ethanol mixtures. In the reaction of catechol oxidation in aqueous solutions, catalytic activity (k(cat)) of tyrosinase-PB complex ([PB]/[tyrosinase] molar ratio 100:1, per mole of polymer) in a wide range of pH was higher than that of free tyrosinase. In aqueous solutions and in water-ethanol mixtures at moderate concentrations of ethanol (10-40% v/v), the value of k(cat) of tyrosinase-PB complex exceeded the activity of free enzyme, from 1.2-2-fold, accompanied by the essential (up to 10-fold) increase in the value of the specificity constant (k(cat)/K(m)). The results are of practical importance for the construction of biocatalysts working successfully in polar organic media.  相似文献

Bioelectrocatalytic oxidation of theophylline was studied at gold and graphite electrodes modified with microbial theophylline oxidase (ThOx), a multi-cofactor redox enzyme capable of selective oxidation of theophylline. Gold electrodes were additionally modified with self-assembled monolayers (SAMs) of (-OH)- and (-NH(2))-terminated alkanethiols of different chain lengths, to achieve compatibility between ThOx and the electrode surface. On graphite, ThOx was either physically co-adsorbed with a surfactant didodecyldimethylammonium bromide (DDAB), or entrapped within an Os-redox-polymer film. At all electrodes, ThOx was bioelectrocatalytically active; direct electrochemistry of ThOx in the absence of theophylline was followed only at the SAM-modified gold electrodes. Direct electrochemistry of ThOx correlated with redox transformations of the heme domain of ThOx, with a E(o/)of -110+/-2 mV versus Ag|AgCl, at pH 7. Bioelectrocatalytic oxidation of theophylline was optimal at mixed (-OH)/(-NH(2))-terminated SAMs; co-adsorption of ThOx with DDAB improved the bioelectrocatalytic performance of the ThOx-electrode. In both cases, the response to theophylline was within the mM range. Alternatively, a reagentless ThOx-electrode based on ThOx cross-linked within the Os-redox-polymer matrix demonstrated a linear response to theophylline within the physiologically important 0.02-0.6mM (3.6-72 mg l(-1)) concentration range with a sensitivity of 52.1+/-7.8 mA cm(-2)M(-1).  相似文献
