Mitochondrial DNA sequences of selected regions of the small subunit and large subunit ribosomal RNAs and cytochrome b genes were analyzed for 10 gadid species, representing 8 genera within Gadidae, and 10 species representing 5 other gadiform families. Phylogenetic analyses revealed that Gadiculus is the most basal gadid genus, and that Trisopterus and Micromesistius constitute a relatively basal clade. Lotidae was identified as the family most closely related to Gadidae. Estimation of divergence times indicated that the most ancient Gadidae split between Gadiculus and the remaining gadid genera occurred about 20 million years ago. The clade including the most recent species (Gadus, Boreogadus, Merlangius, Melanogrammus, and Pollachius) diverged from the Trisopterus/Micromesistius clade approximately 12 million years ago.  相似文献

Understanding the factors that affect most severely the extinction risk of populations is crucial for maintaining biodiversity. An important general pattern derived from stochastic population theory is that time to extinction should decrease with increasing environmental stochasticity. Drake and Lodge recently provided one of the first pieces of experimental support for this simple prediction by artificially manipulating the dynamics of populations of Daphnia. A future challenge will be to include both demographic stochasticity and environmental stochasticity in such studies.  相似文献

Many phleboviruses (family Bunyaviridae) are emerging as medically important viruses. These viruses enter target cells by endocytosis and low pH-dependent membrane fusion in late endosomes. However, the necessary and sufficient factors for fusion have not been fully characterized. We have studied the minimal fusion requirements of a prototypic phlebovirus, Uukuniemi virus, in an in vitro virus-liposome assay. We show that efficient lipid mixing between viral and liposome membranes requires close to physiological temperatures and phospholipids with negatively charged headgroups, such as the late endosomal phospholipid bis(monoacylglycero)phosphate. We further demonstrate that bis(monoacylglycero)phosphate increases Uukuniemi virus fusion beyond the lipid mixing stage. By using electron cryotomography of viral particles in the presence or absence of liposomes, we observed that the conformation of phlebovirus glycoprotein capsomers changes from the native conformation toward a more elongated conformation at a fusion permissive pH. Our results suggest a rationale for phlebovirus entry in late endosomes.  相似文献

Summary Three species of calanoid copepods, Boeckella michaelseni, Parabroteas sarsii and Pseudoboeckella poppei were recorded from 6 freshwater localities in the Husvik area, South Georgia. Of the latter species two distinct size morphs occurred, with no overlap in size even in closely situated populations. The large morph was recorded in lakes, the small was found in ponds. The small morph did not coexist with the large predatory P. sarsii, and we suggest predation pressure from this species as the major cause for the observed distribution of these morphs. The pronounced size segregation as well as small morphological dissimilarities suggest that these morphs are reproductively isolated. While the large morphotype corresponds to that of P. poppei, the taxonomic affinities of the small morph are uncertain.  相似文献

Crustacean remains are not yet useful as parameters for assessing the recent acidification phenomenon, due to too few studies on their ecology. The changes observed in the sediments are related not just to changes in lake chemistry, but also to predatory and competitive interactions. Moreover, the habitat pH of the organism may differ from the measured lake pH, and may as such not be representative. Therefore, species in some acidic lakes may decrease their abundance, whereas the same species in nearby acidic lakes increase. The caution mainly concerns species with no clear pH distribution. Species predominant in acidic or alkaline lakes generally follow patterns more closely related to their physiology. Study of pelagic species, including both body size and shape measurements, should be included in the analysis. If all the ecological factors are included, animal as well as algal remains would form powerful tools to reveal lake histories.  相似文献

The purpose of this study was to evaluate the sensitivity of dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI), diffusion-weighted (DW)-MRI, in vivo MR spectroscopy (MRS), and ex vivo high-resolution magic angle spinning (HR MAS) MRS for the detection of early treatment effects after docetaxel administration. Docetaxel is an antitumor agent that leads to mitotic arrest, apoptosis, and mitotic catastrophe cell death. Gene expression analysis was performed to detect altered regulation in gene expression pathways related to docetaxel treatment effects. Histopathology was used as a measure of alterations in apoptosis and proliferation due to docetaxel. Experiments were performed using MCF7 mouse xenografts, randomized into a docetaxel (30 mg/kg) treatment group and a control group given saline. MRI/MRS was performed 1 day before treatment and 1, 3, and 6 days after treatment. Parametric images of the extracellular extravascular volume fraction (ve) transfer constant (Ktrans) and the apparent diffusion coefficient (ADC) were calculated from the DCE-MRI and DW-MRI data. Biopsies were analyzed by HR MAS MRS, and histopathology and gene expression profiles were determined (Illumina). A significant increase in the ADC 3 and 6 days after treatment and a significant decrease in total choline and a higher ve were found in treated tumors 6 days after treatment. No significant difference was found in the Ktrans between the two groups. Our results show that docetaxel induces apoptosis and decreases proliferation in MCF7 xenografts. Further, these phenomena can be monitored by in vivo MRS, DW-MRI, and gene expression.  相似文献
