The Kinetics of Distribution of the Fat-Soluble Inert Gas Cyclopropane in the Body

A kinetic analysis is made of the experimentally measured time course of respiratory uptake of the highly fat-soluble, inert gas cyclopropane by normal human subjects. The analysis is based on the well-known perfusion-limited model in which a number of body compartments are arranged in parallel with the lungs via the circulating blood. Three distinct body compartments are derived from the data. These are tentatively identified as: (a) adipose tissue (b) fat-poor tissue of low perfusion such as resting muscle, skin, and connective tissue (c) fat-poor tissue of high perfusion such as brain, heart, gut, liver, and kidney. Blood flow rates to the several compartments are also derived from the data. The rates to compartments (a) and (b) are each approximately 10 per cent of the estimated total cardiac output. The derived perfusion (blood flow rate/compartment weight) of the three compartments are in the range, respectively, (a) 2 to 4, (b) 1 to 2.5, (c) 25 to 75 ml/min/100 gm. Uncertainties arising from the experimental data and from simplifications of the model (neglect of lung fill-up phase of uptake and gross diffusion of cyclopropane from one tissue into another) are discussed. The present type of uptake experiment is significant for the problems of total body fat determination, of gross body composition in relation to weight change, of gross shunting of blood flow from one compartment to another, of anesthesia by fat-soluble substances, and of decompression sickness.     

Simplified Aldehyde-Fuchsin Staining of Neurosecretory Cells

Gomori's original aldehyde-fuchsin method has been modified by the combination of Halmi's counter stain with Gabe's preparation, consisting of basic fuchsin, 1 gm; boiling water, 200 ml; with HC1, 2 ml and paraldehyde, 2 ml added after cooling and filtering. The solution so made was allowed to ripen 3-4 days at room temperature, and the precipitate which formed was filtered off and dried at 55-60°C. The staining solution consisted of 0.5 gm of the dry precipitate dissolved in 100 ml of 70% alcohol. The staining follows original procedures except that it is very important to bring slides from water to 70% alcohol before placing them in the aldehyde-fuchsin solution and also to remove all excess staining solution by rinsing in 95% alcohol after staining. The staining solution is stable for at least 6 mo.     

Sequencing, distribution, and inactivation of the dipeptidase A gene (pepDA) from Lactobacillus helveticus CNRZ32.

Previously, the gene for a general dipeptidase (pepDA) was isolated from a gene bank of Lactobacillus helveticus CNRZ32. The pepDA gene consists of a 1,422-bp open reading frame which could encode a polypeptide of 53.5 kDa. No significant identity was found between the deduced amino acid sequence of the pepDA product and the sequence for other polypeptides reported in GenBank. Southern hybridization studies with a pepDA probe indicated that the nucleotide sequence for pepDA is not well conserved among a variety of lactic acid bacteria. Growth studies indicated that a pepDA deletion had no detectable effect on growth rate or acid production by L. helveticus CNRZ32 in milk. Furthermore, no difference in total cellular dipeptidase activity was detected between the mutant and wild-type strains during logarithmic growth in MRS medium.     

Characterization of the Lactobacillus helveticus CNRZ32 pepC gene.

Sequence analysis of the aminopeptidase C gene (pepC) from Lactobacillus helveticus CNRZ32 identified a 1,332-nucleotide open reading frame coding for a polypeptide with motifs characteristic of cysteine proteinases. Homology to the pepC gene appears to be widely distributed among lactic acid bacteria.     

Occurrence and distribution of Legionella species in composted plant materials.

Legionellae were found in many samples of composted plant matter obtained from home gardeners and from facilities which undertook bulk composting. The predominant species isolated from these composts was Legionella pneumophila, the strains of which belonged to serogroups other than serogroup 1. Other Legionella species were present in many samples. Legionella longbeachae serogroup 1, which is implicated in human infections in South Australia, was present in samples obtained from two of six facilities composting large volumes of material and from 3 of 30 gardeners. Many of the species or strains isolated from composts have not been implicated as causative agents of legionellosis in South Austrailia, but some cause infection in healthy and immunosuppressed persons.     

Conservation of human Y chromosome sequences among male great apes: Implications for the evolution of Y chromosomes

Nine newly described single-copy and lowcopy-number genomic DNA sequences isolated from a flow-sorted human Y chromosome library were mapped to regions of the human Y chromosome and were hybridized to Southern blots of male and female great ape genomic DNAs (Gorilla gorilla, Pan troglodytes, Pongo pygmaeus). Eight of the nine sequences mapped to the euchromatic Y long arm (Yq) in humans, and the ninth mapped to the short arm or pericentromeric region. All nine of the newly identified sequences and two additional human Yq sequences hybridized to restriction fragments in male but not female genomic DNA from the great apes, indicating Y chromosome localization. Seven of these 11 human Yq sequences hybridized to similarly-sized restriction endonuclease fragments in all the great ape species analyzed. The five human sequences that mapped to the most distal subregion of Yq (deletion of which region is associated with spermatogenic failure in humans) were hybridized to Southern blots generated by pulsed-field gel electrophoresis. These sequences define a region of approximately 1 Mb on human Yq in which HpaII tiny fragment (HTF) islands appear to be absent. The conservation of these human Yq sequences on great ape Y chromosomes indicates a greater stability in this region of the Y than has been previously described for most anonymous human Y chromosomal sequences. The stability of these sequences on great ape Y chromosomes seems remarkable given that this region of the Y does not undergo meiotic recombination and the sequences do not appear to encode genes for which positive selection might occur. Facilitation of Sexual Receptivity by Ventromedial Hypothalamic Implants of the Antiprogestin RU 486

RU 486 is known primarily as an antagonist to progestins and glucocorticoids. However, RU 486 has also been shown to have agonistic progestational properties in biochemical and behavioral studies. In the current study, RU 486 was implanted directly into tim ventromedial hypothalamus (VMH) to test for facilitative action on the receptive behavior of female ovariectomized Long-Evans rats primed with 5 μg of estradiol benzoate. Cannulae containing RU 486, progesterone (P), or empty cannulae were implanted 48 hr after estrogen priming. The lordosis quotient and the lordosis score were assessed 4 hr after the cannulae were lowered by a standardized test consisting of 10 mounts by a stimulus male. P and RU 486 significantly facilitated receptivity compared to blank implants in terms of lordosis quotient and lordosis score, with no significant difference between the hormone treatments. While only a single dose of each treatment was given in the current study, RU 486 facilitated lordosis when implanted to the VMH as well as progesterone in contrast to our previous results where the steroids were administered systemically.
