α-Methyl-d-glucoside has been shown to be a non-metabolizable sugar which is accumulated against a concentration gradient by a Na+-dependent and phlorizin inhibited process by adult rat renal cortical slices incubatedin vitro at 37 °C. (2) The velocity of accumulation increased linearly with substrate concentrations up to 1.5 mM, but at higher concentrations obeyed saturable kinetics with an apparentKm of about 6 mM. (3) Uptake was enhanced as Na+ was increased from 0 to 100 mequiv/l. Higher Na+ concentrations caused no further effect. (4) A pH maximum of transport occurred between 7.35 and 8.0. (5) Glucoside uptake was inhibited byd-glucose,d-galactose,d-fructose,d-mannose andd-ribose. The inhibition byd-glucose andd-galactose was competitive with apparentKt of 24 and 53 mM, respectively. (6) Bothd-glucose andd-galactose accelerated the efflux of α-methyl-d-glucoside from preloaded cells. (7) Kidney cortex slices from 1-day-old rats were unable to accumulate α-methyl-d-glucoside to form a concentration gradient. The ability to concentrate the glucoside increased progressively after birth, reaching near normal in tissue from 15-day-old animals. The data indicate that the transport process in the newborn is rudimentary, failing also to display accelerated efflux phenomenon. Protons regulateelectrogenic sodium absorption in a variety of epithelia, including thecortical collecting duct, frog skin, and urinary bladder. Recently,three subunits (, , ) coding for the epithelial sodium channel(ENaC) were cloned. However, it is not known whether pH regulatesNa+ channels directly byinteracting with one of the three ENaC subunits or indirectly byinteracting with a regulatory protein. As a first step to identifyingthe molecular mechanisms of proton-mediated regulation of apicalmembrane Na+ permeability inepithelia, we examined the effect of pH on the biophysical propertiesof ENaC. To this end, we expressed various combinations of -, -,and -subunits of ENaC in Xenopusoocytes and studied ENaC currents by the two-electrode voltage-clampand patch-clamp techniques. In addition, the effect of pH on the-ENaC subunit was examined in planar lipid bilayers. We report that ,,-ENaC currents were regulated by changes in intracellular pH(pHi) but not by changes inextracellular pH (pHo).Acidification reduced and alkalization increased channel activity by avoltage-independent mechanism. Moreover, a reduction ofpHi reduced single-channel openprobability, reduced single-channel open time, and increased single-channel closed time without altering single-channel conductance. Summary Paleozoic carbonate ramp sedimentation has generally been described in terms of downlapping clinoforms composed of allochthonous
sediment derived from shallower environments. However, during transgressive episodes when carbonate sediment production is
low and down slope sediment transport by gravity becomes inactive, autochthonous carbonate sediment accumulates in vertical
stacks of essentially in situ sediment. Autochthonous outer ramp deposition is probably a part of many Paleozoic ramp strata,
but has heretofore not been recognized because of the general absence of adequate exposures.
Evidence of autochthonous, in situ deposition and preservation of sediments in a starved setting is well displayed in the
Alamogordo Member of the Lake Valley Formation in south central New Mexico. This evidence includes: 1) beds and bed sets that
are individually continuous and traceable along ramp slope for 32 km, 2) down-ramp sequential distributions of depth-sensitive
organisms and assemblages but patchy distribution of rock types, 3) lack of sedimentary structures indicative of transport,
4) well preserved, unabraded fossils, 5) the common occurrence of fossils in life position, 6) beds traceable into and through
mounds 7) bed thickness trends ascribed to biotic productivity, and 8) geopetal structures in original position.
Integrated paleontologic, sedimentologic, and stratigraphic data provide information about depositional processes and setting.
The depositional slope was approximately 0.5o based on the distribution of fossil algae; this is comparable to dips reported for other Mississippian homoclinal ramps.
An oxygen minimum zone may have impinged on the ramp during a major flooding event. Shifts in biotic gradients from bed to
bed reveal transgressive-regressive patterns that would not be resolvable without detailed paleontological evidence.
The Alamogordo Member formed as a result of transgressive and early highstand starved carbonate sedimentation along a narrow,
homoclinal outer ramp. The surface of maximum flooding and the boundary between the TST and HST are within the Alamogordo

Afternoon observations in summer comparing shoreline with inland atmospheric conditions were made during onshore winds at
Victoria, British Columbia, Canada. The onshore wind came from a cool water surface. Mean monthly water temperatures near
to shore were between 11 and 11.5° C. The onshore wind brought lower air, ground surface radiant and sky radiant temperatures;
lower humidity and greater wind speed. All of these combine to produce a cooler human environment at the shoreline than inland.
The relative importance of climatic elements in producing the cooler environment was assessed using sensitivity analyses with
eight different human thermal exchange models/indices. Air temperature and wind speed had the greatest effect, followed by
ground surface radiant temperature, sky radiant temperature and humidity. Wind speed is the most practical element to consider
when trying to maximize human comfort along the shoreline.

Structurally and functionally, the human cornea is a highly specialized tissue. The corneal stromal collagen matrix is uniquely
transparent and yet maintains a mechanically tough and chemically impermeable barrier between the eye and environment. We
report for the first time that stromal keratocytes of the human cornea show cytogenetic abnormalities with a frequency that
is unprecedented among normal tissues. The abnormalities are acquired, clonal and nonclonal, primarily aneuploid in nature,
and present in normal as well as diseased corneas.

Dispersal and new colony formation in wild naked mole-rats: evidence against inbreeding as the system of mating 总被引:3,自引:1,他引:2
Early field work on naked mole-rats, Heterocephalus glaber,
suggestedthat small colonies are rare and that colonies can only formby
fissioning of existing colonies. Many researchers expectedthat this would
result in extreme inbreeding and high relatednesswithin colonies and would
thus explain the evolution of eusocialityin naked mole-rats. Here I report
evidence of dispersers andoutbreeding in colonies of wild naked mole-rats
that suggeststhat inbreeding is not the system of mating for this speciesand
that outbreeding is probably frequent. Wild dispersers havethe same
morphology as was reported for dispersers in laboratorycolonies. Low levels
of genetic variation in previous moleculargenetic studies of naked mole-rats
probably result from theviscous population structure typical of fossorial

Background
Newcastle disease (ND) outbreaks are global challenges to the poultry industry. Effective management requires rapid identification and virulence prediction of the circulating Newcastle disease viruses (NDV), the causative agent of ND. However, these diagnostics are hindered by the genetic diversity and rapid evolution of NDVs.Methods
An amplicon sequencing (AmpSeq) workflow for virulence and genotype prediction of NDV samples using a third-generation, real-time DNA sequencing platform is described here. 1D MinION sequencing of barcoded NDV amplicons was performed using 33 egg-grown isolates, (15 NDV genotypes), and 15 clinical swab samples collected from field outbreaks. Assembly-based data analysis was performed in a customized, Galaxy-based AmpSeq workflow. MinION-based results were compared to previously published sequences and to sequences obtained using a previously published Illumina MiSeq workflow.Results
For all egg-grown isolates, NDV was detected and virulence and genotype were accurately predicted. For clinical samples, NDV was detected in ten of eleven NDV samples. Six of the clinical samples contained two mixed genotypes as determined by MiSeq, of which the MinION method detected both genotypes in four samples. Additionally, testing a dilution series of one NDV isolate resulted in NDV detection in a dilution as low as 101 50% egg infectious dose per milliliter. This was accomplished in as little as 7 min of sequencing time, with a 98.37% sequence identity compared to the expected consensus obtained by MiSeq.Conclusion
The depth of sequencing, fast sequencing capabilities, accuracy of the consensus sequences, and the low cost of multiplexing allowed for effective virulence prediction and genotype identification of NDVs currently circulating worldwide. The sensitivity of this protocol was preliminary tested using only one genotype. After more extensive evaluation of the sensitivity and specificity, this protocol will likely be applicable to the detection and characterization of NDV.140.
Seed weight varies significantly within and among fruits of wild radish (Raphanus raphanistrum). To determine sources of this variation, we studied fertilization and seed development following controlled pollinations. Within fruits, central ovules were fertilized prior to distal ovules and attained greater seed size. Ninety-seven percent of the variation in mean seed wt per fruit was explained by an analysis of variance incorporating parental effects, pollination date, and the number of seeds per fruit. We document strong maternal effects on the number of ovules per ovary, the number of fertilized ovules per ovary, the number of seeds per fruit, and mean individual seed wt per fruit. Across females, pollen donor had a slight but significant effect on seed wt; no paternal effects on fertilization rate, zygote number, or seed number per fruit were detected. Within females, with one exception, pollen donor had no significant effect on these components of seed development. Stronger maternal main effects may be due to donor x recipient interactions, cytoplasmic factors, the genetic inequity within triploid endosperm, and/or strict maternal control over resource allocation. The large maternal effects relative to paternal effects may limit the rate at which natural selection acts on paternal traits expressed prior to seed maturation. 相似文献
