The metabolism of the orally active anabolic steroid methyltestosterone in the equine was investigated by administration of the drug along with a tritiated traces. In this study some of the metabolites were identified and a radio immunoassay screen and immunoaffinity chromatography gel for methyltestosterone were also evaluated. Pathway intermediates, in particular the 17-methylandrostanediols, were studied to gain an insight into the most likely stereochemistry of the major metabolites. The predominant phase I biotransformations involve reduction of the A ring 3-oxo and 4-ene groups to yield predominantly 3β-hydroxy-5-androstane products and hydroxylation of the steroid nucleus at several positions. Epimerisation of the 17-methyl group also occurred. Ten steroids could be positively identified by comparison with authentic references materials and many other triol, tetrol and pentols were also observed. Phase II metabolites and sulphate conjugates in particular, were common. 相似文献

Degradation of fluoranthene, pyrene, benz[a]anthracene and dibenz[a,h]anthracene by Burkholderia cepacia 总被引:7,自引:1,他引:6
Microbiological analysis of soils from a polycyclic aromatic hydrocarbon (PAH)-contaminated site resulted in the enrichment of five microbial communities capable of utilizing pyrene as a sole carbon and energy source. Communities 4 and 5 rapidly degraded a number of different PAH compounds. Three pure cultures were isolated from community 5 using a spray plate method with pyrene as the sole carbon source. The cultures were identified as strains of Burkholderia ( Pseudomonas ) cepacia on the basis of biochemical and growth tests. The pure cultures (VUN 10 001, VUN 10 002 and VUN 10 003) were capable of degrading fluorene, phenanthrene and pyrene (100 mg l−1 ) to undetectable levels within 7–10 d in standard serum bottle cultures. Pyrene degradation was observed at concentrations up to 1000 mg l−1 . The three isolates were also able to degrade other PAHs including fluoranthene, benz[ a ]anthracene and dibenz[ a , h ]anthracene as sole carbon and energy sources. Stimulation of dibenz[ a , h ]anthracene and benzo[ a ]pyrene degradation was achieved by the addition of small quantities of phenanthrene to cultures containing these compounds. Photodynamic therapy (PDT) is a treatment combining a photosensitiser, molecular oxygen and visible light of characteristic wavelength to produce cytotoxic reactive oxygen species (ROS). Within our centre, a series of phenothiazinium salts were synthesised and initial characterisation studies performed to determine any potential use for PDT. All photosensitisers within the series were shown to have useful spectral properties for PDT, with absorbance lambdamax above 667 nm. The Log P values of the compounds were shown to range from -0.9 to > +2.0. Furthermore, Log P values were shown to be important in determining the site of subcellular localisation and as such the site of photooxidative damage. Derivatives with a Log P value of greater than +1.0 were shown to initially localise to the lysosomes then relocalise throughout the cytoplasm following illumination, whereas compounds with intermediate Log P values (-0.7 to +1.0) all remained lysosomal. Only methylene blue (Log P-0.9) was shown to redistribute to the nucleus upon illumination. Following treatment of RIF-1 cells with each phenothiazinium salt for 1 h and subsequent exposure to 665 nm laser light at a fluence rate of 10 mW cm(-2)(18 J cm(-2)), it was determined that the most potent photosensitiser was 260-fold more potent than methylene blue. Furthermore, the PDT efficacy of the photosensitisers was shown to be related to the level of mitochondrial damage induced directly following illumination. 相似文献

The Southern Plateau subantarctic region, southeast of New Zealand, is an important feeding area for birds, seals and fish, and a fishing ground for commercially significant species. The Southern Plateau is a major morphometric feature, covering approximately 433,620 km2 with average depth of 615 m. The region is noted for its relatively low levels of phytoplankton biomass and primary production that is iron-limited. In order to evaluate the implications of these attributes for the functioning of this ecosystem a steady-state, 19-compartment model was constructed using Ecopath with Ecosim software of Christensen et al. [www.ecopath.org]. The system is driven by primary production that is primarily governed by the supply of iron and light. The total system biomass of 6.28 g C m−2 is very low compared with systems so far modelled with a total system throughput of 1136 g C m−2 year−1. In the model, the Southern Plateau retains 69% of the biomass in the pelagic system and 99% of total production. Although fish are caught demersally, most of their food is part of production in the pelagic system. Top predators represent about 0.3% of total biomass and account for about 0.24 g C m−2 year−1 of food consumed made up of birds 0.058 g C m−2 year−1, seals 0.041 g C m−2 year−1, and toothed 0.094 g C m−2 year−1 and baleen whales 0.051 g C m−2 year−1. This amounts to 105,803 tonnes carbon over the whole of the Southern Plateau and is about 17% of the total amount of food eaten by non-mesopelagic fish. Mean transfer efficiencies between trophic levels II and IV of 23% are at the high end of the range reported in the literature. In the model, adult fish production is almost completely accounted for by the fisheries take (32%), consumption by seals (7%), toothed whales (21%), other adult fish (13%), and squid (20%). Fish and squid catches are at the trophic levels of 4.8 and 5.0, respectively. The gross efficiency of the fishery is 0.018% (catch/primary production). Although not all data come from direct knowledge of this system, the model reflects its general characteristics, namely a low primary production system dominated by the microbial loop, low sedimentation to the seafloor, high transfer efficiencies, a long food web and supporting high-level predators. 相似文献

Ca2+ - and Mg2+ -dependent ATPase activity (EC 3.6.1.3) in a plasma membrane-enriched fraction increased rapidly after in vivo application of physiologically active concentrations of triacontanol (TRIA) to the roots of barley ( Hordeum vulgare L. cv. Conquest) seedlings. Ca2+ - and Mg2+ -dependent ATPase activity was 64 and 85% higher, respectively, in the roots of seedlings germinated in the presence of growth-promoting concentrations of TRIA compared to controls. The increase in vivo was concentration dependent, with the greatest increase obtained at 2.3 n M TRIA. Maximal stimulation of ATPase activity of excised tissue treated with TRIA coincided with the temperature at which the barley was grown. At this temperature the plasma membrane is primarily in a mixed gel/liquid crystalline state. Pretreatment of barley roots with cyclohexamide did not alter ATPase stimulation by TRIA. Two to three times more [14 C]-TRIA (mg membrane protein)−1 was found associated with plasma membrane-enriched vesicles treated with TRIA than with vesicles enriched for mitochondrial membranes or for vesicles enriched for tonoplast, Golgi and rough endoplasmic reticulum. Both Ca2+ - and Mg2+ -dependent ATPase activity increased by 40–60% within 30 min of the addition of 2.3 n M TRIA to cell-free extracts of barley roots. The addition of octacosanol, the C28 analogue of TRIA, to cell-free extracts did not affect metal-dependent ATPase activity. Consistent with many studies in the green-house, simultaneous additions of equimolar amounts of TRIA and octacosanol to cell-free extracts resulted in inhibition of ATPase stimulation by TRIA.
