The Western Australian camaenid genera Plectorhagada and Strepsitaurus have morphological similarities and mutually exclusive ranges near Cape Range. Sequences of ctyochrome c oxidase subunit I (COI) and 16S mitochondrial DNA (mtDNA) genes confirmed that the two genera are genetically close sister clades. Targeted sampling showed that Strepsitaurus, which is confined to Cape Range, lies within a hole in the distribution of the more broadly distributed Plectorhagada that occurs on the coastal strip surrounding Cape Range. Species of the two genera meet at the transition between the rocky Cape Range and the sandier coastal areas, providing a rare example of the close replacement of genera. Within each genus, mtDNA sequences confirmed the monophyly and genetic distinctness of species, with few exceptions that show the need for additional work, and with the addition of three new species in the Cape Range area. As is typical of Australian camaenids, distributions of congeneric species are mutually exclusive, but in some cases close proximity is associated with contrasting habitats, such as gorge endemics versus the species on top of Cape Range. In sympatry, Strepsitaurus rugus (Cotton, 1951) and Strepsitaurus williami Solem, 1997 are separated by microhabitat. These local associations with habitat indicate that ecological differences, and not simply allopatric divergence, contribute to the lack of sympatry between closely related Australian camaenids. © 2015 The Linnean Society of London  相似文献

Atriplex nummularia is a polyploid Australian saltbush which has been identified as a suitable species for use in the rehabilitation of agricultural land affected by salinity. We isolated 12 polymorphic loci for a preliminary assessment of genetic variability and structure within the species as a basis for a breeding programme. Preliminary screening of loci in 40 individuals from two populations revealed multibanded genotypes consisting of up to seven alleles in a single individual, with up to 29 alleles observed at a single locus. The multibanded patterns are consistent with the polyploid status of this species.  相似文献

Acta 735, 341-351 and 352-360) the ordinary Scatchard-type analysis has been shown to yield erroneous results when applied to the binding of large molecules to membranes or cells. Formulae have been given to treat the limiting cases of very thin and of very bulky ligands. These results are now extended to include ligands of any shape and cooperative interactions. As an example, data on the cooperative binding of polymyxin to charged lipid bilayers are reevaluated. Melittin induced voltage-dependent conductance in DOPC lipid bilayers   总被引:1,自引:0,他引:1  
Melittin-induced conductance was measured on planar bilayers made from dioleoylphosphatidylcholine. Upon application of a fixed voltage, the current response was monophasic and remained so even after prolonged observation times. The conductance of melittin-doped bilayers increased exponentially with voltage. In addition, an ohmic contribution appeared after some current had passed. The voltage-dependent conductance increased e-fold every 22 mV and was proportional to the fourth power of the aqueous monomeric peptide concentration, for all salt concentrations investigated (0.4-1.8 M NaCl). Discrete conductance steps could be resolved at all these salt concentrations. The amplitudes of these steps were highly variable. In each experiment, conductance was initially only observed for potentials which were positive on the side of peptide addition. As more and more current passed across the bilayer, the current-voltage curves became symmetric. The system needed some time to reach stationary current-voltage characteristics: about 50 min at pH 7 but only about 15 min at pH 8, suggesting involvement of the N-terminus (pK around 7.5) of melittin in the slow formation of a 'prepore'.  相似文献

Temperature is one of the main factors that determine sexual reproduction in terrestrial and emergent aquatic plant species. The effect of temperature on sexual reproduction and seed production of Glyceria maxima (Hartm.) Holmb. in the southern hemisphere is unknown. Glyceria maxima collections in February 2010 at three isolated infestations in KwaZulu-Natal failed to yield a single seed, only empty panicles. Laboratory experiments showed that vernalisation had no consistent effect on seed production. Field- and laboratory-grown plants produced seeds in the 2010/2011 season, because of having sufficient time at optimum temperatures required for seed production (1 491 and 1 585 hours, respectively), compared to a shorter period (1 352 hours) of suitable temperatures during the 2009/2010 growing season. An inadequate period of optimum temperatures (15–25°C) during seed production resulted in the lack of seeds in the field in the 2009/2010 growing season. This study showed that temperature and duration of exposure thereto during the seed-production period play vital roles in G. maxima sexual reproduction.  相似文献

The CpG Island Methylator Phenotype (CIMP) is fundamental to an important subset of colorectal cancer; however, its cause is unknown. CIMP is associated with microsatellite instability but is also found in BRAF mutant microsatellite stable cancers that are associated with poor prognosis. The isocitrate dehydrogenase 1 (IDH1) gene causes CIMP in glioma due to an activating mutation that produces the 2-hydroxyglutarate oncometabolite. We therefore examined IDH1 alteration as a potential cause of CIMP in colorectal cancer. The IDH1 mutational hotspot was screened in 86 CIMP-positive and 80 CIMP-negative cancers. The entire coding sequence was examined in 81 CIMP-positive colorectal cancers. Forty-seven cancers varying by CIMP-status and IDH1 mutation status were examined using Illumina 450K DNA methylation microarrays. The R132C IDH1 mutation was detected in 4/166 cancers. All IDH1 mutations were in CIMP cancers that were BRAF mutant and microsatellite stable (4/45, 8.9%). Unsupervised hierarchical cluster analysis identified an IDH1 mutation-like methylation signature in approximately half of the CIMP-positive cancers. IDH1 mutation appears to cause CIMP in a small proportion of BRAF mutant, microsatellite stable colorectal cancers. This study provides a precedent that a single gene mutation may cause CIMP in colorectal cancer, and that this will be associated with a specific epigenetic signature and clinicopathological features.  相似文献

目的:观察并分析奥关拉唑联合法莫替丁治疗反流性食管炎的临床效果。方法:选取2008年5月至2012年5月在本院确诊并治疗的反流性食管炎患者45例,随机平均分为三组。联合用药组(15例):每日早餐前口服20mg奥关拉唑,睡前口服20mg法莫替丁;奥关拉唑组(15例):每日口服两次奥美拉唑,每次20mg;法莫替丁组(15例):每日口服两次法莫替丁,每次20mg。每组的治疗时间均为8周。在内镜指导下观察并比较三组患者的胸痛、反酸和烧心等主要病征的改善情况,综合评价三种治疗方法的临床疗效。结果:联合用药组较其他两组获得的疗效更明显,患者的症状得到较好的改善,差异有统计学意义(P〈0.05)。结论:奥美拉唑联合法莫替丁能够有效地抑制胃酸的分泌,对于反流性食管炎的,临床治疗具有良好的效果。  相似文献

Camaenid land snails are often the focus of targeted biodiversity surveys in Australia, but can be difficult to locate because they hibernate for most of the year. Searches would be more effective if the density of live snails could be predicted from features of the habitat, or from the abundance of empty shells from long dead individuals. In 53 populations of Rhagada, the dominant genus of land snail in the Pilbara bioregion of Western Australia, the density of live snails is not correlated with the cover or height of spinifex (Triodia), the main habitat plant occupied by Rhagada. On the other hand, the density of empty shells on the soil surface does explain snail population density, but the relationship is weak (r2=0.209). Nevertheless, the data suggest that searches for live Rhagada are more likely to succeed in areas where empty shells are most abundant.  相似文献

Novel and classical modeling techniques used in recent work on small and large motor systems illustrate the different roles that models play in furthering our understanding of motor systems.
