During the G1/S transition, p21 proteolysis is mediated by Skp2; however, p21 reaccumulates in G2 and is degraded again in prometaphase. How p21 degradation is controlled in mitosis remains unexplored. We found that Cdc20 (an activator of the ubiquitin ligase APC/C) binds p21 in cultured cells and identified a D box motif in p21 necessary for APC/C(Cdc20)-mediated ubiquitylation of p21. Overexpression of Cdc20 or Skp2 destabilized wild-type p21; however, only Skp2, but not Cdc20, was able to destabilize a p21(D box) mutant. Silencing of Cdc20 induced an accumulation of p21, increased the fraction of p21 bound to Cdk1, and inhibited Cdk1 activity in p21(+/+) prometaphase cells, but not in p21(-/-) cells. Thus, in prometaphase Cdc20 positively regulates Cdk1 by mediating the degradation of p21. We propose that the APC/C(Cdc20)-mediated degradation of p21 contributes to the full activation of Cdk1 necessary for mitotic events and prevents mitotic slippage during spindle checkpoint activation. 相似文献

To analyse the influence of cold ischemic time (CIT) (2–24 h) and of cryopreservation (liquid phase) on the viability of the
valvular fibroblasts and in the presence of apoptosis. Cardiac valves from 10 pigs were evaluated by anatomo-pathological
study of the wall, muscle and leaflet. At the same time, the presence of cellular death due to apoptosis was investigated
in two ways; directly on tissue by Apodetec system and by two-colour flow cytometry assay analyzing a suspension of fibroblast
from valve leaflets using Anexina V and propidium iodure (PI). We established three groups of samples to compare different
experimental conditions: 2 h of ischemia (group 1), 24 h of ischemia (group 2), and a programme of cryopreservation (−1°C/min)
after 2 h of ischemia, followed by storage in liquid nitrogen during a week and thawing was performed (group 3). The analysis
of viabilities showed slight differences between all three groups. The results indicated CIT of 24 h undergoing more structural
affectation than CIT of 2 h. Flow cytometry analysis did not show important differences between groups; however cryopreserved
samples (group 3) slightly less viability and a higher percentage of death by apoptosis than group 1 and 2 using flow cytometry.
Apoptosis was confirmed on tissue from all valves but mainly in samples of group 2 and group 3. In summary, the viability
of the valves in the case of ischemic times of 2 h, 24 h or after cryopreservation/thawing differs slightly. The death of

The NS neurons are nonspiking cells, present as pairs in each midbody ganglion of the leech nervous system, which display
a very extensive arborization. They were shown to regulate the coactivation of motoneurons. Here we have investigated the
electrophysiological properties of these neurons under the hypothesis that transmission along the extensive neurites requires
the aid of voltage-dependent conductances. The results indicate that NS neurons respond to electrical stimulation with a spike-like
event, which was not an all-or-none but rather a graded phenomenon that depended on the intensity and duration of the electrical
stimulus. The spike-like response was activated at a membrane potential of approximately −50 mV; its amplitude was a logarithmic
function of the extracellular Ca2+ concentration and was unaffected by a broad range of changes in the extracellular Na+ concentration; intracellular application of tetraethylammonium (TEA) caused a large increase in its amplitude and duration.
These data indicate that NS neurons bear voltage-dependent low-threshold Ca2+ and TEA-sensitive K+ conductances that could contribute to shaping synaptic signals, or transmission along the extensive neuritic tree. 相似文献

Background
The huge amount of data generated by DNA chips is a powerful basis to classify various pathologies. However, constant evolution of microarray technology makes it difficult to mix data from different chip types for class prediction of limited sample populations. Affymetrix® technology provides both a quantitative fluorescence signal and a decision (detection call: absent or present) based on signed-rank algorithms applied to several hybridization repeats of each gene, with a per-chip normalization. We developed a new prediction method for class belonging based on the detection call only from recent Affymetrix chip type. Biological data were obtained by hybridization on U133A, U133B and U133Plus 2.0 microarrays of purified normal B cells and cells from three independent groups of multiple myeloma (MM) patients.Results
After a call-based data reduction step to filter out non class-discriminative probe sets, the gene list obtained was reduced to a predictor with correction for multiple testing by iterative deletion of probe sets that sequentially improve inter-class comparisons and their significance. The error rate of the method was determined using leave-one-out and 5-fold cross-validation. It was successfully applied to (i) determine a sex predictor with the normal donor group classifying gender with no error in all patient groups except for male MM samples with a Y chromosome deletion, (ii) predict the immunoglobulin light and heavy chains expressed by the malignant myeloma clones of the validation group and (iii) predict sex, light and heavy chain nature for every new patient. Finally, this method was shown powerful when compared to the popular classification method Prediction Analysis of Microarray (PAM).Conclusion
This normalization-free method is routinely used for quality control and correction of collection errors in patient reports to clinicians. It can be easily extended to multiple class prediction suitable with clinical groups, and looks particularly promising through international cooperative projects like the "Microarray Quality Control project of US FDA" MAQC as a predictive classifier for diagnostic, prognostic and response to treatment. Finally, it can be used as a powerful tool to mine published data generated on Affymetrix systems and more generally classify samples with binary feature values.996.
The effect of the Photosystem II (PSII) inhibitors dichlorophenyldimethylurea (DCMU) and bromonitrothymol (BNT) on the rate of the high-light induced D1 protein turnover was studied in whole cells of two cyanobacterial strains Synechocystis PCC 6803 and Synechococcus PCC 7942. In Synechocystis the D1 degradation was slowed down to a similar extent in the presence of either inhibitor compared with control cells. This slower degradation corresponded with the retardation of Photosystem II photoinactivation (PSIIPI) measured as a decline of PS II activity in the illuminated cells treated with chloramphenicol (CAP). The ongoing D1 synthesis in the presence of both PS II inhibitors was confirmed by unchanging PS II activity and the steady-state level of D1 during illumination in the absence of CAP. In Synechococcus cells both DCMU and BNT blocked the turnover of the 'low-light' D1 form (D1:1) but did not prevent the exchange of the 'high-light' form D1:2 for the D1:1 form. The similar effect of both herbicides on the D1 exchange was in contrast with their influence on the rate of PSIIPI. While DCMU had a pronounced protective effect, BNT significantly increased the rate of PS II photodamage. The fast BNT-induced decline of PS II activity was also observed in Synechocystis cells treated with azide, an inhibitor of reactive oxygen species scavenging enzymes. Therefore, we assume that the distinct sensitivity of the two cyanobacterial strains to BNT can be caused by different content and/or activity of these enzymes in each strain. 相似文献

Two pure peroxidase isoenzymes B1 and D4 were isolated from the upper parts of 10-day-old wheat seedlings by means of gel and ion-exchange chromatography. Their MWs were 85000 and 24000 respectively. B1 was unstable and under various conditions it was converted to another isoenzyme, electrophoretically identical with D4. B1 contains about 40% of neutral sugars: 17.2% arabinose, 15.3% galactose, 5% glucose and traces of mannose. D4 is free of neutral sugars. None of the isoenzymes contained amino sugars. B1 oxidizes ferulic and p-coumaric acids. This oxidation has two pH optima of 4.4 and 5.4–5.6 and is inhibited by high concentrations of substrates, cyanide and azide. B1 oxidizes IAA in the presence of phenolic cofactor and Mn2+ ions. IAA oxidation has two pH optima of 4.5 and 5.6 and is inhibited by high substrate concentration, cyanide and azide, and by a number of indole derivatives. The main products of IAA oxidation are 3-methyleneoxindole and indole-3-methanol. o- and p- diphenols induce a lag period prior to IAA oxidation. Ferulic acid is oxidized during this lag period, probably to a dimer. B1 is able to produce H2O2 from oxygen. Mn2+ ions, a phenolic cofactor and an electron donor (IAA or NADH) are needed. B1 oxidizes α-keto-γ- methylmercaptobutyric acid to ethylene. D4 has a low peroxidatic activity and is inactive as an IAA oxidase. Thus B1 is probably an active cell wall-bound peroxidase isoenzyme, whereas D4 is its decomposition product. 相似文献

Dual targeting of organellar seryl-tRNA synthetase to maize mitochondria and chloroplasts 总被引:2,自引:0,他引:2
Aminoacyl-tRNA synthetases (AARSs) play a critical role in translation and are thus required in three plant protein-synthesizing compartments: cytosol, mitochondria and plastids. A systematic study had previously shown extensive sharing of organellar AARSs from Arabidopsis thaliana, mostly between mitochondria and chloroplasts. However, distribution of AARSs from monocot species, such as maize, has never been experimentally investigated. Here we demonstrate dual targeting of maize seryl-tRNA synthetase, SerZMo, into both mitochondria and chloroplasts using combination of complementary methods, including in vitro import assay, transient expression analysis of green fluorescent protein (GFP) fusions and immunodetection. We also show that SerZMo dual localization is established by the virtue of an ambiguous targeting peptide. Full-length SerZMo protein fused to GFP is targeted to chloroplast stromules, indicating that SerZMo protein performs its function in plastid stroma. The deletion mutant lacking N-terminal region of the ambiguous SerZMo targeting peptide was neither targeted into mitochondria nor chloroplasts, indicating the importance of this region in both mitochondrial and chloroplastic import. 相似文献
